This work is based on formulating and optimizing controlled release (CR) valsartan (160 mg) tablets using different viscosity grades of the cellulosic polymer. The objective was to develop an effective once-daily drug delivery system of this cardiovascular agent. Central composite design was used for designing the formulations. Polymers used were Methocel® K4M, K15M and K100M. Compatibility of excipients with active was studied through FT-IR. Micromeritic properties were determined and formulations exhibiting appropriate flow characteristics were compressed. Swelling behavior and in vitro buoyancy effect were studied and response surface curves were constructed to optimize the formulation. Multi-point dissolution profiles of valsartan CR tablets at pH 1.2, 4.5 and 6.8 were obtained. Model-dependent and model-independent methods were performed including f2, stability test as per ICH guidelines and ANOVA. FT-IR studies revealed the compatibility of valsartan with all excipients. Formulation K4T9 (containing 25% K4M polymer) was selected to be the best optimized trial, based on physical properties and controlled release profile (23% at 4 h, 82% at 16 h and 100% at 24 h). Results of buoyancy and swelling behavior indicated that HPMC-K4M polymer exhibited excellent floating lag time and swelling indexes. In vitro drug release kinetics showed that formulation K4T9 displayed Korsmeyer–Peppas drug release pattern with r value > 0.99. The manufacturing process of K4T9 was also found to be reproducible with a shelf life period of 41 and 36 months at room temperature and accelerated conditions, respectively. Valsartan CR matrix-based formulation was successfully prepared with Methocel K4M retardant.  相似文献

The bone marrow (BM) microenvironment plays a crucial role in the development and progression of leukemia (AML). Intracellular reactive oxygen species (ROS) are involved in the regulation of the biology of leukemia-initiating cells, where the antioxidant enzyme GPx-3 could be involved as a determinant of cellular self-renewal. Little is known however about the role of the microenvironment in the control of the oxidative metabolism of AML cells. In the present study, a coculture model of BM mesenchymal stromal cells (MSCs) and AML cells (KG1a cell-line and primary BM blasts) was used to explore this metabolic pathway. MSC-contact, rather than culture with MSC-conditioned medium, decreases ROS levels and inhibits the Nrf-2 pathway through overexpression of GPx3 in AML cells. The decrease of ROS levels also inactivates p38MAPK and reduces the proliferation of AML cells. Conversely, contact with AML cells modifies MSCs in that they display an increased oxidative stress and Nrf-2 activation, together with a concomitant lowered expression of GPx-3. Altogether, these experiments suggest that a reciprocal control of oxidative metabolism is initiated by direct cell–cell contact between MSCs and AML cells. GPx-3 expression appears to play a crucial role in this cross-talk and could be involved in the regulation of leukemogenesis.  相似文献

In the current study, graphene oxide (GO) was prepared using green chemistry with modified Hummer's method without incorporating sodium nitrate (NaNO3). Solvent casting was employed to fabricate GO-doped poly(ethylene oxide) (PEO), that is, PEO/GO composites with various proportion of Na2SO4 and were then subjected to characterization via advanced spectroscopic techniques for different physicochemical aspects to estimate their potential applications as marketable products. XRD analysis explored that fabricated composites are more crystalline than neat PEO. PEO/GO/Na2SO4 composite films offered maximum crystallinity. SEM displayed the same trend. TG/DTA thermogram exposed better thermal stability than pristine polymer. FTIR studies confirmed complexation among hybrid's components. Elongation-at-break and Young's modulus displayed an enhancing behavior with an incremental loading of salt and filler. In terms of mechanical performance, composite of PEO with 0.37 wt % GO and 0.08 g salt was found to be an ideal composition during the course of study. © 2019 Wiley Periodicals, Inc. J. Appl. Polym. The application of solvent-aided crystallization (SAC) is based on the addition of a solvent, here 1-butanol, to crude biodiesel to catalyze the purification process by separating biodiesel from contaminants via crystallization process. Response surface methodology was applied to optimize the process parameters of SAC, represented by biodiesel purity. The purified biodiesel was analyzed by means of gas chromatography-mass spectrometry for the composition of the present fatty acid methyl ester (FAME). Under the predicted optimum process conditions within the experimental ranges for the highest biodiesel purity, the predicted biodiesel purity was 99.375 %.  相似文献

Adulteration in traded medicinal plants is a significant issue nowadays and use of these adulterated medicinal plants can impose harmful impact to end user. However, this problem can be overcome by ensuring the identification of traded medicinal plants which are used in making different herbal medicines. In this regard, palynological markers are considered to be an important taxonomic tool in the identification of original medicinal plant from its adulterant. Hence this study attempted to provide particular reliable palynological markers for distinguishing selected medicinal plants from their adulterants, that is, Cinnamomum verum versus Canella winterana, Cinnamomum tamala versus Cinnamomum obtusifolium, Gymnema sylvestre versus Gymnema lactiferum, Artemisia maritima versus Artemisia absinthium, Achillea millefolium versus Adhatoda vasaka, Sphaeranthus indicus versus Sphaeranthus africanus, Averrhoa carambola versus Butea monosperma, and Morus nigra versus Morus alba. Results demonstrated great variations in multiple palynological characters between original medicinal plant and its adulterant such as in pollen size, shape, colpi length, exine, intine thickness, and fertility. In equatorial view, circular to spheroidal shape of pollen was found in A. millefolium while oblate shape was observed in A. vasaka. Similarly B. monosperma pollen was 34 μm, whereas pollen of its adulterant A. carambola was 21 μm. Moreover, colpi length of A. maritima was 11.8 μm, whereas 4.5 μm in A. absinthium. Hence it can be concluded that palynological characters are commendably helpful in identification of genuine medicinal plant from its adulterant.  相似文献

Isobutanol is deemed to be a next‐generation biofuel and a renewable platform chemical. 1 Non‐natural biosynthetic pathways for isobutanol production have been implemented in cell‐based and in vitro systems with Bacillus subtilis acetolactate synthase (AlsS) as key biocatalyst. 2 – 6 AlsS catalyzes the condensation of two pyruvate molecules to acetolactate with thiamine diphosphate and Mg2+ as cofactors. AlsS also catalyzes the conversion of 2‐ketoisovalerate into isobutyraldehyde, the immediate precursor of isobutanol. Our phylogenetic analysis suggests that the ALS enzyme family forms a distinct subgroup of ThDP‐dependent enzymes. To unravel catalytically relevant structure‐function relationships, we solved the AlsS crystal structure at 2.3 Å in the presence of ThDP, Mg2+ and in a transition state with a 2‐lactyl moiety bound to ThDP. We supplemented our structural data by point mutations in the active site to identify catalytically important residues.  相似文献

Histogram equalization is the common method used for contrast enhancement. The mean brightness of the image is adjusted to middle of the permitted range and hence is not suitable for consumer electronics products. A novel contrast enhancement method using modified octagon histogram equalization is developed to overcome the drawback of conventional technique for gray scale images. The proposed algorithm is applied for boat image, microstructure of steel and human head. The contrast enhanced out of the images mentioned is obtained, and the efficiency of the algorithm is evaluated. Simulation results shows that the proposed method can enhance the different types of images effectively. Besides, the proposed contrast enhancement method using modified octagon histogram equalization has comparable performance with black and white stretching and adaptive histogram equalization.  相似文献
