目的:建立RP-HPLC法测定血浆中异烟肼毒性代谢物肼的方法.方法:采用反相C18柱,以5mmol/L醋酸钠(用1 mol/L醋酸调节pH至5.0)-乙腈(35:65)为流动相,检测波长300nm.结果:肼血浆浓度在8.5~85.2μmol/L范围内线形关系良好(r=0.999),平均回收率为100.2%,日内、日间RSD均小于2%.结论:本方法准确并较为简便,为测定肼血浓以监测异烟肼毒性提供了实验方法.  相似文献

目的建立同时测定血清中异烟肼(INH)、利福平(RFP)、吡嗪酰胺(PZA)浓度的反相高效液相色谱法.方法 RFP检测条件:Symmetry C8色谱柱(5 μm,150 mm×3.9 mm),流动相为磷酸盐缓冲液(pH 5.5)∶甲醇=30∶70,流速0.45 ml/min,测定波长340 nm;INH、PZA检测条件:Symmetry C18色谱柱(5 μm,250 mm×4.6 mm),流动相为磷酸盐缓冲液(pH 6.0)∶乙腈=91∶9,流速0.8 ml/min,测定波长254 nm.结果以峰高外标法定量,线性范围:利福平为0.5~32 mg/L,INH为0.5~20 mg/L,PZA为1.0~60.0 mg/L,均具有良好的线性关系.3种药物的日内、日间变异均<10%.回收率>80%.结论该方法简便、快速,灵敏度和准确度较高,无干扰,用于上述3种药物复合制剂卫非特片的药代动力学研究,证明能满足人体血药浓度监测及药代动力学研究的需要.  相似文献

目的:研究异烟肼在电化学预处理玻碳电极上的电化学响应。方法:采用三电极系统,以电化学预处理玻碳电极为工作电极,饱和甘汞电极为参比电极,铂电极为对电极,用循环伏安法研究异烟肼的电化学行为。结果:异烟肼在预处理的玻碳电极上产生两个不可逆的还原峰,电位分别为-0.80V和-0.90V,其过程是一个表面吸附过程。发现在-0.90V出现的还原峰的峰电流与异烟肼的浓度在1.2×10-10~1.3×10-3mol/L的范围内成线性关系。异烟肼的检测限低达1.2×10-11mol/L(ta=40 s,S/N=3),平行8次测定1.2×10-6mol/L异烟肼的相对标准偏差为4.5%。在B ritton-Rob inson(B-R)缓冲溶液和在模拟尿样中的回收率分别为(101.7±3.8)%和(96.3±3.3)%。结论:该方法可灵敏测定微量异烟肼,可用于临床结核病患者药物浓度的监测。  相似文献

目的建立人血浆中替米沙坦浓度RP-HPLC荧光检测法。方法以缬沙坦为内标定量血浆中替米沙坦的浓度,血浆样品在酸性条件下用苯萃取后氮气吹干,残留物用流动相复溶后直接进样。样品分离用ZORBAXEclipseXDB-C18柱(4.6×150mm,5μm)和填充相同材料的预柱(4.6×12.5mm,5μm),流动相为甲醇-乙腈-10mmol/L磷酸二氢钾溶液(pH3.2)(5∶40∶55,v/v/v),流速为1.0mL/min,进样体积为20μL,荧光检测激发波长为250nm,发射波长为375nm,柱温为25℃。结果血浆中内源性物质不干扰测定结果,替米沙坦和内标的保留时间分别为8.8min和10.7min。在浓度1μg/L~180μg/L范围内线性关系良好,线性方程:A=0.2769R+0.03788,相关系数r=0.9995,检测限为1μg/L。浓度为2.0、30.0、70.0μg/L血浆质控样品的平均回收率分别为97.53%、104.6%和105.1%。低、中、高浓度血浆质控样品的日内和日间相对标准偏差(RSD)均小于7%。结论本研究建立的血浆中替米沙坦的浓度测定方法,具有准确、快速、简单,灵敏度高、线性范围宽等特点。该方法更适于替米沙坦血药浓度测定及替米沙坦药物代谢动力学研究。  相似文献

目的建立大鼠血浆中盐酸二甲双胍的检测方法。方法采用离子对RP-HPLC法测定大鼠血浆中药物浓度,色谱柱为Lichrospher C18(4.6mm×250 mm,5μm),流动相为3 mmol/L十二烷基磺酸钠水溶液-已腈(32∶68 v/v,PH=4)。结果在0.1~4.0 mg.L-1范围内盐酸二甲双胍峰面积与浓度线性关系良好(r=0.9996),最低检测浓度为0.03 mg.L-1,绝对回收率为83.2%~95.4%,准确度为96.7%~106.8%,日内、日间RSD均小于10%。结论样品处理简便快速,测定方法准确可靠,适合于盐酸二甲双胍的药代动力学研究。  相似文献

目的 建立用HPLC法测定异烟肼注射液含量的方法 .方法 HPLC法,采用Hypersil BDS C18柱,以乙腈-0.075 mol·L-1磷酸二氢钠溶液(体积比5:95)为流动相,流速为1.0 mL·min-1,检测波长262 nm.结果 异烟肼在9.90~99.00 μg·mL-1浓度范围内与峰面积线性关系良好(r=0.999 9),平均回收率100.3%(RSD=1.0%).结论 本法操作简便,快速准确,重复性好,可作为本品质量控制方法 .  相似文献

目的建立银荷方药物制备物及大鼠血浆中槲皮素、山奈酚、异鼠李素的含量测定方法,以研究银荷方药物制备物黄酮类成分的质量表征特性。方法制备物和血浆样品均加酸进行水解,并用乙醚-丙酮(14∶1)萃取法处理血浆样品,采用HPLC分析,以C_(18)为固定相,流动相为乙腈-0.2%磷酸水溶液梯度洗脱,在270 nm检测黄酮类苷元槲皮素、山奈酚、异鼠李素的含量。结果经方法学考证,槲皮素、山奈酚、异鼠李素的线性范围分别为1.49~29.70 mg/L(r=0.999 3),0.65~12.90mg/L(r=0.999 5),0.47~9.40 mg/L(r=0.999 0),日内、日间精密度RSD≤5.85%,血浆样品冻融稳定性良好,室温放置10 h,在-80℃条件下保存1个月的稳定性均良好,符合生物样品分析要求。槲皮素、山奈酚、异鼠李素在正常组中的血药浓度分别为7.25、1.71、3.73 mg/L,在血栓组中血药浓度分别为8.53、2.64、5.69 mg/L,在抗凝组中血药浓度分别为8.32、2.76、4.56 mg/L。结论本文所建立的检测方法适用于银荷方药物制备物黄酮类成分槲皮素、山奈酚和异鼠李素的体内含量测定,并可应用于分析黄酮类成分质量表征特性,为基于黄酮类成分的银荷方药物质量精准控制提供科学依据。  相似文献

目的建立全自动二维高效色谱法(2D-HPLC)测定血清中伏立康唑的浓度并应用于临床。方法第一维色谱柱为ASTON SC2柱(4.6mm×25mm,5μm),流动相为20mmol/L磷酸铵∶乙腈∶甲醇=3∶1∶1(V∶V∶V,磷酸调pH=5.4),流速1.2mL/min;第二维色谱柱为SHIMADZU C18柱(4.6 mm×150 mm,5μm),流动相为10mmol/L醋酸铵(三乙胺调pH7.0)∶10mmol/L醋酸铵(三氟乙酸调pH3.0)∶甲醇∶乙腈=30∶10∶10∶50(V∶V∶V∶V),流速1.0mL/min;中间柱为ASTON SH柱(3mm×10mm,5μm),辅助流动相为纯水。样品由一维色谱柱萃取,经中间柱捕获并转移至第二维色谱柱分析,检测波长256nm,柱温40℃,进样量200μL。结果伏立康唑在0.28~22.56μg/mL范围内线性关系良好,r=0.999 9,最低定量限为0.28μg/mL,方法回收率为99.01%~104.62%,提取回收率为85.81%~89.82%,日内、日间精密度RSD均小于9%。结论该方法操作简单、准确、灵敏,符合血清样品的测定要求,适用于伏立康唑临床血药浓度测定。  相似文献

目的:建立测定大鼠血浆多索茶碱血药浓度的高效毛细管电泳法.方法:样品用体积分数30%三氯醋酸沉淀蛋白,电解缓冲液为25 mmol/L磷酸盐缓冲液-50 mmol/L SDS(体积比为20:80,pH=7.0),分离通道为50cm×50 μm未涂层毛细管,紫外检测波长为273 nm,分离电压12 kV,采用峰面积内标法定量.结果:采用本法时,多索茶碱血浆浓度在2.5~80.0 mg/L范围内线性关系良好,标准曲线回归方程为y=-8.40×10-3 0.13X(r=0.998 1,P<0.05),方法回收率在允许范围内,日内和日间变异分别小于3.4%和7.5%.结论:本法稳定可靠、操作简单、分析速度快、分离效果好.  相似文献

Objective: To evaluatel the value of D-dimers in patients with acute aortic dissection (AAD). Methods: This study consisted of 16 patients with AAD and 27 non-AAD patients. Serum D-dimets were measured by Sta-Liatest D-DI immunoturbidimetric assay. Results: D-dimer level was higher (P < 0.001) in patients with AAD(7.91 ± 5.52 μg/ml) than that in non- AAD group(1.57±1.24 μg/ml). D-dimer was positive (>0.4 μg/ml) in all patients with AAD and in 10 control group patients (37%). Among patients with acute AAD, D-dimers tended to be higher in Stanford A than in Stanford B (8.67 ± 4.31 μg/ml vs. 3.24±1.27 μg/ml, P <0.01). D-dimer values tended to be higher in more extended disease(3.84 ± 1.65 μg/ml, 8.57 ± 3.58 μg/ml and 11.87 ± 5.69 μg/ml in thoracic aorta, thoracic and abdominal aorta, thoracic and abdominal aorta and iliacal arteries, respectively, P < 0.05 for both 8.57 ± 3.58 and 11.87 ± 5.69 vs. 3.84 ± 1.65 ). Including the control group into the analysis, we found a sensitivity of 100%, a negative predictive value of 100%, and a specificity of 66% and a positive predictive value of 64% for D-dimer in diagnosis of AAD in our patients with suspected AAD. Conclusion: D-dimer was elevated in patients with AAD. A negative D-dimer test result could be useful in excluding AAD.  相似文献

Objective: To set up a simple and reliable rat model of combined liver-kidney transplantation. Methods: SD rats served as both donors and recipients. 4℃ sodium lactate Ringer's was infused from portal veins to donated livers,and from abdominal aorta to donated kidneys, respectively. Anastomosis of the portal vein and the inferior vena cava (IVC) inferior to the right kidney between the graft and the recipient was performed by a double cuff method, then the superior hepatic vena cava with suture. A patch of donated renal artery was anastomosed to the recipient abdominal aorta. The urethra and bile duct were reconstructed with a simple inside bracket. Results: Among 65 cases of combined liver-kidney transplantation, the success rate in the late 40 cases was 77.5%. The function of the grafted liver and kidney remained normal. Conclusion: This rat model of combined liver-kidney transplantation can be established in common laboratory conditions with high success rate and meet the needs of renal transplantation experiment.  相似文献

Objective To observe blood pressure change with age in salt-sensitive teenagers whose salt sensitivity were determined by repeated testing.Methods Salt sensitivity was determined through intravenous infusion of normal saline combined with volume-depletion by oral diuretic furosemide in 55 teenagers. After five years, salt sensitivity was re-examined and subject blood pressure was followed up. Blood pressure changes in salt-sensitive teenagers were compared to that of non-salt sensitive teenagers over five years.Results After 5 years, the repetition rate of salt sensitivity determined by intravenous saline loading is 92.7%. In teenagers with salt sensitivity on the baseline, both the systolic blood pressure increments and increment rates were much higher than non-salt sensitive teenagers (12.7±12.1 mmHg vs. 2.8±5.2 mmHg, P< 0.01; 12.2%± 12.0% vs. 2.5% ±4.4%, P< 0.001,respectively). There was a similar trend for diastolic blood pressure (8.4 ± 6.4 mmHg vs. 3.7 ± 6.4 mmHg, P = 0.052; 13.2% ±10.6 % vs. 6.8%± 10.1%, P = 0.053, respectively).Conclusions Salt sensitivity determined by intravenous saline loading showed good reproducibility. Blood pressure increments with age were much higher in salt-sensitive teenagers than non-salt sensitive teenagers, especially in terms of systolic blood pressure.  相似文献

Shock wave lithotripsy (SWL) is a treatment of choice for upper urinary stones. However, this procedure is inappropriate for obese patients because the focus is often unable to reach the target owing to the limited focal distance in shock wave source. Although treating such patients in a blast path may increase the application length of shock wave source, it's difficult to find this path on the lithotripter monitor. Excess production of reactive oxygen species(ROS)of mitochondrion mediated by hyperglycemia is the common pathogenesis of angiopathic complications of diabetes.TCM holds that the damp from the dysfunction of spleen.kidney and liver is the causative factor of complications of diabetes.This is similar to the mechanism of Ros resulting in angiopathic complications of diabetes.When the angiopathic complications of type II diabetes mellitus(T2DM)are difierentiated as caused by turbid damp in TCM can be explained as ROS.Since the obstruction of pathogenic damp in channels and collaterals is said to be the main pathogenesis,the treating principle should be dissolving the damp to remove the obstruction.  相似文献

目的:评价使用安心颗粒对急诊经皮冠状动脉介入术(PPCI)术后生活质量的影响.方法:将160例接受PPCI的急性ST段抬高型心肌梗死患者随机分为安心颗粒组(术前顿服安心颗粒8.8g,术后安心颗粒4.4 g/次,每日2次)和对照组(仅接受基础药物治疗).所有患者均服用阿司匹林、氯吡格雷和阿托伐他汀.分别在入院时、出院前1d、出院后180 d时,应用心肌梗死多维度量表(MIDAS)、中文版SF-36评价量表对患者生活质量评分.并观察术后30 d以内的出血并发症、血小板减少症发生情况.结果:入院时和出院前1d,两组患者的心肌梗死MIDAS、SF-36量表评分比较无差异(P>0.05);出院后180 d时,与对照组比较,安心颗粒组MIDAS、SF-36评分明显减低(P<0.05);组内与入院时比较,两组出院前1d、出院后180 d时,MIDAS、SF-36评分均降低(P<0.05).两组患者在随访期间均无大量出血、少量出血、重度和极重度血小板减少症发生,安心颗粒组有4例、对照组有7例发生不明显出血(P>0.05).两组发生轻度血小板减少症的患者数比较无差异(P>0.05).结论:PPCI使用安心颗粒,能改善急性ST段抬高型心肌梗死患者的生活质量,且不增加出血风险.  相似文献

Objectives To explore serum cytokines levels (including IL-1 β, sIL-2R, IL-6, TNF-α, and IFN-v) and their significance in patients with acute coronary syndrome (ACS) and the subsequent follow-ups, with attempt to estimate the role of various serum inflammatory markers in the diagnosis and assessment of ACS.Methods The study population include 40 patients with acute myocardial infarction (AMI), 40 patients with unstable angina pectoris (UAP), and 40 controls. Among the 80 patients, 60 patients attended a follow up 4 months later. Serum inflammatory markers including IL-1 β, sIL-2R, IL-6, TNF-α, and IFN-v were measured by enzyme linked immunosorbent assay.Results Serum IL- 1 β, sIL-2R, IL-6, TNF-α were significantly higher in AMI group or UAP group compared to the control group and became significantly lower 4 months later in the follow-up patients. Serum levels of IFN-v shows no significant difference between AMI group or UAP group and controls, also showing no significant change when measured in follow up patients. There was no correlation between serum creatine kinase-MB isoenzyme levels and serum inflammatory markers either in UAP or AMI group. Furthermore, when divided into two subgroups using Wagner's QRS scoring system in the AMI group, there is no difference of each serum inflammatory marker between ≤ 6 scores group and > 6 scores group.Conclusion Serum levels of certain inflammatory markers may have some diagnostic value for ACS, and can be a useful marker reflecting disease stability.  相似文献
