Insulin is a peptide hormone with many physiological functions, besides its use in diabetes treatment. An important role of insulin is related to the wound healing process—however, insulin itself is too sensitive to the external environment requiring the protective of a nanocarrier. Polymer-based nanoparticles can protect, deliver, and retain the protein in the target area. This study aims to produce and characterize a topical treatment for wound healing consisting of insulin-loaded poly-DL-lactide/glycolide (PLGA) nanoparticles. Insulin-loaded nanoparticles present a mean size of approximately 500 nm and neutral surface charge. Spherical shaped nanoparticles are observed by scanning electron microscopy and confirmed by atomic force microscopy. SDS-PAGE and circular dichroism analysis demonstrated that insulin preserved its integrity and secondary structure after the encapsulation process. In vitro release studies suggested a controlled release profile. Safety of the formulation was confirmed using cell lines, and cell viability was concentration and time-dependent. Preliminary safety in vivo assays also revealed promising results.  相似文献

Hydroxyapatite (HA) nanoparticles (NPs) doped with different radioisotopes for use as theranostic systems play an important role in scientific research nowadays due to their ability to simultaneously act in the treatment and diagnosis of various types of cancers. In this work, we describe the synthesis and characterization of a hydroxyapatite/tenorite nanocomposite functionalized with folic acid, representing a nanotheranostic material with potential for application as an agent in positron emission tomography imaging systems and to act specifically in the treatment and diagnosis of osteosarcoma. 64Cu and 32P were produced by nuclear activation in the TRIGA reactor at CDTN. The obtained samples were characterized by XRD with Rietveld refinement, XAFS, SEM, BET, TGA, FTIR, CHN, ICP-AES, XPS and gamma spectroscopy. We investigated how CuO grows in HA NPs, the stability of the interactions between CuO and HA constituents and the interactions between folic acid and the surface of the HA NPs. The results indicate the formation of a second phase (tenorite) besides hydroxyapatite, and that the interactions between the two phases are stable, resulting in a nanocomposite. Furthermore, the activation of 64Cu and 32P inside the HA matrix, through the exposition to a neutron flux, produces a theranostic material of interest for biological tests.  相似文献

In this study the potential of cloud point extraction formed by a non-ionic surfactant was used in order to separate polyphenols from industrial residues of camu-camu. The effects of operational conditions of the cloud point extraction (CPE) on the polyphenol recovery and volumetric ratio were investigated. The results showed a maximum recovery of 95.71% that was obtained using 7.0 wt% Triton X-114, native pH (3.25), and 80 wt% polyphenol extract at 30 °C. The use of cloud point extraction was successful to recover the polyphenols from agroindustrial residue since it is a simple as well as of low-cost technique.  相似文献

Magnetic polymers supports have proven to be valuable materials for enzyme immobilization, as they allow recovering the catalyst by magnetic separation, precluding the need for costly and time-consuming separation steps. In this study, magnetic copolymer supports were synthesized using styrene (STY) and different crosslinking agents (divinylbenzene, ethylene glycol dimethacrylate, or triethylene glycol dimethacrylate) and initiators (azobisisobutyronitrile or benzoyl peroxide) and used to immobilize Candida antarctica lipase B (CALB). The aim was to obtain biocatalysts with high enzymatic activity and satisfactory morphological properties for use in biotransformation reactions. Two morphological properties known to influence the immobilization yield were taken into consideration, specific surface area, and swelling index. Experimental data were compared to the predictions of a model based on molar balance, method of moments, numerical fractionation, and elementary gel structures. The high correlation (R2 = 0.9974) between experimental and predicted values demonstrated the suitability of the model for estimating the textural properties of enzyme supports. CALB was successfully immobilized, showing high hydrolytic activity (500–700 U g−1) and good thermal stability at 50°C. CALB/STY-EGDMA-M was 14 times more stable than free CALB. The results confirm the efficiency of the immobilization method and the suitability of the copolymers for enzyme immobilization.  相似文献

Penicillium camemberti lipase immobilized on a magnetized poly(styrene-co-divinylbenzene) was used as a biocatalyst for isopropyl palmitate synthesis. The reaction conditions were determined by 22 factorial central composite design. A mathematical model based on a simplified kinetic approach was developed to describe the system and validated with the experimental data. An assay carried out in a stirred-tank reactor confirmed the proposed model. The ester was purified and the properties such as density and water content were similar to those found in commercially available isopropyl palmitate.  相似文献
