Chronic infections are considered one of the most severe problems in skin wounds, and bacteria are present in over 90% of chronic wounds. Pseudomonas aeruginosa is frequently isolated from chronic wounds and is thought to be a cause of delayed wound healing. Invariant natural killer T (iNKT) cells, unique lymphocytes with a potent regulatory ability in various inflammatory responses, accelerate the wound healing process. In the present study, we investigated the contribution of iNKT cells in the host defense against P. aeruginosa inoculation at the wound sites. We analyzed the re-epithelialization, bacterial load, accumulation of leukocytes, and production of cytokines and antimicrobial peptides. In iNKT cell–deficient (Jα18KO) mice, re-epithelialization was significantly decreased, and the number of live colonies was significantly increased, when compared with those in wild-type (WT) mice on day 7. IL-17A, and IL-22 production was significantly lower in Jα18KO mice than in WT mice on day 5. Furthermore, the administration of α-galactosylceramide (α-GalCer), a specific activator of iNKT cells, led to enhanced host protection, as shown by reduced bacterial load, and to increased production of IL-22, IL-23, and S100A9 compared that of with WT mice. These results suggest that iNKT cells promote P. aeruginosa clearance during skin wound healing.  相似文献

ABSTRACT

In order to develop a glass production method that inhibits formation of the Mo-bearing secondary phase, the so-called yellow phase (YP), and also to clarify its formation behavior, vitrification tests using a small-scale liquid-fed ceramic melter (LFCM) were conducted. Chemically simulated PUREX liquid waste containing Na, Mo and other fission elements was fed into a molten glass pool in the melter together with several sizes of feed glass. When the glass beads with a diameter of around 2 mm were fed, some YP was contained in the vitrified glass products at 25 wt% waste loading. While, almost no YP was detected in the glass products when glass powder with a diameter of less than 63 μm was used. The microscopic analysis of the cold-cap samples revealed that powderization of the feed glass contributed to the facilitation of Na dissolution to the feed glass in the cold-cap. This quick intake of Na to the feed glass prevented the formation of liquid Na2MoO4 aggregation and provided homogeneous Mo dispersion in the cold-cap with forms of alkali earth and/or rare earth molybdates. The homogeneous dispersion of Mo resulted in the fast and complete dissolution of Mo in the glass melt.  相似文献

The cellulose binding domain (CBD) of cellulosome-integrating protein A from Clostridium thermocellum NBRC 103400 was genetically fused to FMN-dependent NADH-azoreductase (AZR) and glucose 1-dehydrogenase (GDH) from Bacillus subtilis. The fusion enzymes, AZR-CBD and CBD-GDH, were expressed in Escherichia coli Rosetta-gami B (DE3). The enzymes were purified from cell-free extracts, and the specific activity of AZR-CBD was 15.1 U/mg and that of CBD-GDH was 22.6 U/mg. AZR-CBD and CBD-GDH bound strongly to 0.5 % swollen cellulose at approximately 95 and 98 % of the initial protein amounts, respectively. After immobilization onto the swollen cellulose, AZR-CBD and CBD-GDH retained their catalytic activity. Both enzymes bound weakly to 0.5 % microcrystalline cellulose, but the addition of a high concentration of microcrystalline cellulose (10 %) improved the binding rate of both enzymes. A reactor for flow injection analysis was filled with microcrystalline cellulose-immobilized AZR-CBD and CBD-GDH. This flow injection analysis system was successfully applied for the determination of glucose, and a linear calibration curve was observed in the range of approximately 0.16–2.5 mM glucose, with a correlation coefficient, r, of 0.998.  相似文献

The voltage variation in a remote system is large when the system is connected by long‐distance AC cables due to the cable capacitance. In Japan, the longest 54‐km 66‐kV AC submarine cable interconnection between the Kyushu mainland and Goto Islands was commissioned in 2005. It was requested to mitigate the voltage variation caused by switching off and on one circuit of the two circuits in the AC cables when a fault occurs. Since the conventional voltage control methods such as transformer tap changer or shunt capacitor and reactor banks are not sufficient because of their slow response time, a static var compensator (SVC) was installed on the Goto Islands. In such an application, an SVC control method should be developed so as not to override the existing voltage control systems. This paper describes the SVC control method developed for the Goto Islands AC interconnection project, which can be applied to similar situations. The effectiveness of the control method was verified by the results of effective value simulation and of field testing, which was implemented before the SVC was commissioned in 2007. © 2013 Wiley Periodicals, Inc. In patient-specific arterial fluid–structure interaction (FSI) computations the image-based arterial geometry comes from a configuration that is not stress-free. We present a method for estimation of element-based zero-stress (ZS) state. The method has three main components. (1) An iterative method, which starts with an initial guess for the ZS state, is used for computing the element-based ZS state such that when a given pressure load is applied, the image-based target shape is matched. (2) A method for straight-tube geometries with single and multiple layers is used for computing the element-based ZS state so that we match the given diameter and longitudinal stretch in the target configuration and the “opening angle.” (3) An element-based mapping between the arterial and straight-tube configurations is used for mapping from the arterial configuration to the straight-tube configuration, and for mapping the estimated ZS state of the straight tube back to the arterial configuration, to be used as the initial guess for the iterative method that matches the image-based target shape. Biopolymer based composites have been employed in numerous applications with increasing interest not only due to renewable, eco-friendly nature, but also because of the flexibility in processing conditions and competitive cost of their end products. The conductive materials from biopolymers have been found applicable in robots, medical imaging, sensitive membranes, actuators, visual displays, electronic wiring and shielding, and components in batteries. Cellulose is one of the most abundant biopolymers in the nature, which has received special attention for development of conducting materials due to biocompatibility for protein and drug immobilization and ability to form the composites with synthetic polymers. The present review is aimed to provide concisely the current status in this field of conducting composites from cellulose, with brief discussions of associated problems and future applications.  相似文献

The effects of morphological changes on the thermal expansion, toughness and heat resistance of polyamide‐6 (PA)/styrene–ethylene–butylene–styrene (SEBS)/polyphenylene ether (PPE) blends were investigated. Compared with the typical ‘sea (PA matrix)–island (PPE domain)–lake (SEBS in PPE domain)’ morphology, an injection‐molded ternary blend with a preferential distribution of SEBS component at the interface between PA and PPE exhibited a low coefficient of linear thermal expansion (CLTE) in the flow direction. This low CLTE was ascribed to the deformation of SEBS and PA into a co‐continuous microlayer network structure during injection molding. Consequently, the expansion preferentially occurred towards the thickness direction. Further CLTE reduction either by a change in PA viscosity or by the selective location of an inorganic filler was examined, and its influences on impact strength and heat resistance are discussed based on transmission electron microscopy observations. © 2015 Society of Chemical Industry  相似文献

Electrical and physical characteristics of the Al2O3/InGaAs interfaces with (1 1 1)A and (1 0 0) orientations were investigated in an attempt to understand the origin of electron mobility enhancement in the (1 1 1)A-channel metal-insulator-semiconductor field-effect-transistor. The (1 1 1)A interface has less As atoms of high oxidation states as probed by X-ray photoelectron spectroscopy. The electrical measurements showed that energy distribution of the interface traps for the (1 1 1)A interface is shifted toward the conduction band as compared to that for the (1 0 0) interface. Laterally-compressed cross-section transmission electron microscopy images showed that the characteristic lengths of the interface roughness are different between the (1 1 1)A and (1 0 0) interfaces. The contributions of the Coulomb and roughness scattering mechanisms are discussed based on the experimental results.  相似文献
