Pregnancy by Assisted Reproductive Technology Is Associated with Shorter Telomere Length in Neonates

Telomere length (TL) influences the development of lifestyle-related diseases, and neonatal TL may influence their prevalence. Various factors have been reported to affect neonatal TL. Although the fetus is exposed to multiple conditions in utero, the main factors affecting the shortening of neonatal TL are still not known. In this study, we sought to identify factors that influence fetal TL. A total of 578 mother-newborn pairs were included for TL analysis. TL was measured in genomic DNA extracted from cord blood samples using quantitative PCR. The clinical factors examined at enrollment included the following intrauterine environmental factors: maternal age, assisted reproductive technology (ART) used, body mass index (BMI), gestational diabetes mellitus (GDM), maternal stress, smoking, alcohol consumption, preterm delivery, small-for-gestational-age, neonatal sex, and placental weight. Univariate and multivariate regression analyses were used to verify the relationship between neonatal TL and these clinical factors. The median neonatal TL to single-copy gene ratio was 1.0. Pregnancy with ART was among the 11 factors associated with shorter neonatal TL. From multiple regression analysis, we determined that neonatal TL was significantly shorter for pregnancies in the ART group than in the other groups. Lysine demethylase 5 C (KDM5C) controls epigenetic gene expression and is attracting great interest in the field of chemical epigenetics. KDM5C has emerged as a therapeutic target for anti-prostate cancer agents, and recently we identified triazole 1 as an inhibitor of KDM5C. Compound 1 exhibited highly potent KDM5C-inhibitory activity in in vitro enzyme assays, but did not show strong anticancer effects. Therefore, a different approach is needed for the development of anticancer agents targeting KDM5C. Here, we attempted to identify KDM5C degraders by focusing on a protein-knockdown strategy. Compound 3 b , which was designed based on compound 1 , degraded KDM5C and inhibited the growth of prostate cancer PC-3 cells more strongly than compound 1 . These findings suggest that KDM5C degraders are more effective as anticancer agents than compounds that only inhibit the catalytic activity of KDM5C. 相似文献

Collisions of birds, especially endangered species, with wind turbines is a major environmental concern. Automatic bird monitoring can be of aid in resolving the issue, particularly in environmental risk assessments and real‐time collision avoidance. For automatic recognition of birds in images, a clean, detailed, and realistic dataset to learn features and classifiers is crucial for any machine‐learning‐based method. Here, we constructed a bird image dataset that is derived from the actual environment of a wind farm and that is useful for examining realistic challenges in bird recognition in practice. It consists of high‐resolution images covering a wide monitoring area around a turbine. The birds captured in these images are at relatively low resolution and are hierarchically labeled by experts for fine‐grained species classification. We conducted evaluations of state‐of‐the‐art image recognition methods by using this dataset. The evaluations revealed that a deep‐learning‐based method and a simpler traditional learning method were almost equally successful at detection, while the former captures more generalized features. The most promising results were provided by the deep‐learning‐based method in classification. The best methods in our experiments recorded a 0.98 true positive rate for bird detection at a false positive rate of 0.05 and a 0.85 true positive rate for species classification at a false positive rate of 0.1. 相似文献

The elemental composition of a single yeast, green alga, or red blood cell (RBC) was precisely determined by using inductively coupled plasma-mass spectrometry (ICP-MS) operating in fast time-resolved analysis (TRA) mode. The technique is known as single-cell (SC)-ICP-MS. Phosphorus, sulfur, magnesium, zinc, and iron were detected in the three types of cell. The elemental composition of yeast and green alga obtained by SC-ICP-MS was consistent with results obtained from conventional ICP-MS measurements following acid digestion of the cells. Slight differences were found in the measured values between SC-ICP-MS and the conventional ICP-MS results for RBC. However, the SC-ICP-MS results for S and Fe in RBC were closer to the estimated values for these elements that were calculated from the level of hemoglobin in RBCs. The data suggest that SC-ICP-MS is suitable for the analysis of various cell types, namely, fungus, plant, and animal cells. 相似文献

A micro fabrication is presented to manufacture hydrophobic surfaces with micro-scale structures. Hydrophobicity is controlled with the shape and the alignment of micro pillars in the structure. The structures are manufactured in large areas at high production rates in the following processes: (1) the structure is fabricated on a tool by focused ion beam sputtering; (2) the reverse structure is formed on a metal plate by incremental stamping using the structured tool; and (3) the structure is transferred onto plastic plates by molding. A consecutive stamping is also proposed to fabricate several structures on a surface accurately with a structured tool, in which the moving pitch of the structured tool is numerically controlled. The effect of the surface topography on hydrophobicity is discussed with measuring contact angles on the structured surfaces in the water droplet tests. Hydrophobicity on the plastic plate is associated with the solid fraction on the structured surface based on the Cassie–Baxter model. We have investigated the crystal growth of single-phase MnSi1.75−x
by a temperature gradient solution growth (TGSG) method using Ga and Sn as solvents and MnSi1.7 alloy as the solute, and measured the thermoelectric properties of the resulting crystals. Single-phase Mn11Si19 and Mn4Si7 crystals were grown successfully using Ga and Sn as solvents, respectively. The typical size of a grown ingot of Mn11Si19 was 2 mm to 4 mm in thickness and 12 mm in diameter, whereas Mn4Si7 had polyhedral shape with dimensions in the range of several millimeters. The single-phase Mn11Si19 has good electrical conduction (ρ = 0.89 × 10−3 Ω cm to 1.09 × 10−3 Ω cm) compared with melt-grown multiphase higher-manganese silicide (HMS) crystals. The Seebeck coefficient, power factor,
and thermal conductivity were 77 μV K−1 to 85 μV K−1, 6.7 μW cm−1 K−2 to 7.2 μW cm−1 K−2, and 0.032 W cm−1 K−1, respectively, at 300 K. 相似文献
