There is a need for effective vaccine delivery systems and vaccine adjuvants without extraneous excipients that can compromise or minimize their efficacy. Vaccine adjuvants cytosine–phosphate–guanosine oligodeoxynucleotides (CpG ODNs) can effectively activate immune responses to secrete cytokines. However, CpG ODNs are not stable in serum due to enzymatic cleavage and are difficult to transport through cell membranes. Herein, DNA microcapsules made of CpG ODNs arranged into 3D nanostructures are developed to improve the serum stability and immunostimulatory effect of CpG. The DNA microcapsules allow encapsulation and co‐delivery of cargoes, including glycogen. The DNA capsules, with >4 million copies of CpG motifs per capsule, are internalized in cells and accumulate in endosomes, where the Toll‐like receptor 9 is engaged by CpG. The capsules induce up to 10‐fold and 20‐fold increases in tumor necrosis factor (TNF)‐α and interleukin (IL)‐6 secretion, respectively, in RAW264.7 cells compared with CpG ODNs. Furthermore, the microcapsules stimulate TNF‐α and IL‐6 secretion in a concentration‐ and time‐dependent manner. The immunostimulatory activity of the capsules correlates to their intracellular trafficking, endosomal confinement, and degradation, assessed by confocal and super‐resolution microscopy. These DNA capsules can serve as both adjuvants to stimulate an immune reaction and vehicles to encapsulate vaccine peptides/genes to achieve synergistic immune effects.  相似文献

The antitermite (termiticidal and antifeedant) activity of Protium javanicum Burm. f. extract was investigated. The ethyl acetate fraction was active. Scopoletin (1), quercetin, and stigmasterol were isolated by bioassay-guided fractionation. Scopoletin had the highest activity among the three compounds. In order to investigate the structure-activity relationship (SAR) of the methoxy and hydroxy groups at the C-6 and C-7 positions of the coumarin skeleton, we synthesized several coumarin derivatives whose chemical structures are similar to scopoletin. Scopoletin exhibited the strongest termiticidal activity among the 10 compounds tested, followed by 6-methoxycoumarin (3), 6-hydroxycoumarin (7), and umbelliferone (8). All compounds except coumarin (9) showed antifeedant activity.  相似文献

Emission of inorganic particulate matter (PM) from the incineration of dewatered sewage sludge has been investigated in a novel ash melting furnace. The sludge containing 79 wt% water was incinerated in an oxygen-enriched atmosphere at the primary temperature of 1400 °C, and its unburned volatile was combusted at 1100 °C in a secondary combustion chamber. A 13-stage low-pressure-impactor and the conventional impinger methods were employed for PM sampling at the outlet of the secondary combustion chamber. The results indicate that, PM is dominated by volatile and semi-volatile elements including Br, Cl, P, S, Na, K, Zn, As, Cu, Mn and Ni. Less refractory elements were found. PM has two major fractions: <0.22 and ?0.22 μm. Their chemical forms as well as water solubility are different between two fractions. The majority of Br, nearly half of Cl, and 40% of S and P are present in the small fraction. They are mostly water-soluble due to the association with alkali elements and heavy metals. The water-insoluble calcium sulfate and calcium/iron phosphate were, however, found in the large fraction of PM. Regarding the cations, the water solubilities of Na, K, Mn and Ni are close to their proportions partitioned into the small fraction of PM, since their water-soluble species were preferentially formed in this fraction. A relatively weak correlation for Al, Ca and As, while no such a correlation were found for Cu, Zn and Fe, due to the complex compounds formed for them.  相似文献
