上肢熏洗剂治疗上肢损伤后遗症的临床分析

我院协定处方“上肢熏洗剂”以威灵仙、伸筋草等10种中草药组成,具有活血、舒筋、祛风、补肾功效,使用水煎熏蒸和浸洗的方法治疗上肢损伤后关节功能障碍,经临床152例观察,疗效满意,总优良率为94.1%。     

Synergistic Antitumor Interaction of Human Monocyte Chemotactant Protein-1 Gene Transfer and Modulator for Tumor-Infiltrating Macrophages

Purpose. In order to evaluate the possibility of synergistic antitumor gene therapy by the gene delivery of monocyte chemotactant protein-1 (MCP-1/MCAF/IE), the effect of a biological response modulater for macrophages on tumor progression of gene transfected tumor cells was studied. Methods. Cachexia-inducing adenocarcinoma cells (cell line colon 26, clone 20) were transfected with either a control plasmid or MCP-1 cDNA. Results. The production of MCP-1 reached 70-80 ng/ml in vitro when transfectant cells were cultured at a cell density of 1 × 10⁵ cells/ml for 3 days. Transfection of MCP-1 cDNA did not affect the growth ratein vitro. Also, MCP-1-transfectants formed tumors after intra-footpad inoculation similar in size to the parental cells. The number of infiltrating macrophages in the primary tumor of the transfectant rapidly increased from the 3rd to 5th day after inoculation as revealed by immunohistochemical staining using an antibody against mouse macrophages. An earlier, greater, but no longer-lasting increase in tumor-infiltrating macrophages was induced in tumors by MCP-1 transfection was compared to that induced by the parent cells. On the 10th day after the inoculation, the tumor-infiltrating macrophages in mice inoculated MCP-1 transfectants were decreased to a level similar to that of the parent cells. Groups of mice were treated intraperitoneally with LPS at different times after the inoculation. Tumor cells producing high levels of MCP-1 were significantly lysed by macrophages treated with LPS, whereas parental or control transfected cells were not. Conclusions. Combination immunotherapy can provide a rationale for the application of MCP-1 treatment to increase immunological responses to cancer.     

Histamine-releasing factors

There is increasing evidence that human basophils accumulate at sites of chronic inflammation, and, in particular, in allergic asthma. Investigators have, therefore, become very interested in identifying proteins that activate these cells. Recently, the gene encoding a candidate for this function, a novel molecule, the IgE-dependent histamine-releasing factor, was cloned.     

血小板激活因子对大鼠脑血管通透性的影响及药物的保护作用

颈内动脉注射血小板激活因子(PAF),再给伊文思蓝,可见脑实质染色程度加深,而颈内动脉只注射伊文思蓝,脑实质未见染色。而我们合成的新药SZ-1可剂量依赖性地抑制PAF诱导的脑实质伊文思蓝染色程度的加深。在体外培养的脑微血管平滑肌细胞上,PAF能显著刺激~(14)-花生四烯酸的释放,而SZ-1能剂量依赖性地抑制这种释放,提示PAF在脑内产生的损害除与其他因素相关外,还与其刺激花生四烯酸释放有密切关系,SZ-1对PAF引起的脑部损害有保护作用。     

Pathogenesis of IgA nephropathy: In vitro activation of human mesangial cells by IgA immune complex leads to cytokine secretion

IgA immune complex (IC) plays a crucial role in the pathogenesis of IgA nephropathy (IgAN). As IgA-IC is not itself cytotoxic, other mediators may be involved in the pathogenesis. In order to elucidate the mechanisms by which IgA-IC mediates renal injury in IgAN, the ability of IgA-IC to ‘activate’ cultured human mesangial cells (HMC) was studied. HMC were incubated with nephritogenic IgA-IC, containing a MOPC-315 plasmacytoma-derived IgA anti-dinitrophenyl (DNP) and DNP-conjugated bovine serum albumin. The cells showed morphological changes, an accelerated rate of proliferation, and increased production of interleukin-1 (IL-1), interleukin-6 (IL-6), platelet activating factor (PAF) and generation of superoxide anion. The enhancement of IL-1 and IL-6 mRNA expression in HMC incubated with IgA-IC was identified by dot blot analysis. Northern blot hybridization also demonstrated an augmented IL-6 mRNA expression in HMC treated with IgA-IC. These results suggest that nephritogenic IgA-IC may amplify the proliferation of HMC and the production of immune/chemical mediators and superoxide anion thereby resulting in the renal lesions of IgAN.     

银杏叶对银屑病患者中性粒细胞趋化反应的影响

血小板活化因子在银屑病发病中起重要作用，其拮抗剂的研究对银屑病的治疗有重要意义。应用琼脂糖玻片法对２０例银屑病患者外周血中性粒细胞的趋化反应进行了观察，发现银杏叶能抑制ＰＡＦ引起的趋化反应，为银杏叶治疗银屑病提供了理论依据。     

益气活血系列方治疗膜性肾炎31例临床观察

肾活检诊断为膜性肾炎病人61例,中药组31例,激素组30例。分别予益气活血系列方和激素治疗,同时测定治疗前后24小时尿蛋白、血浆白蛋白、血脂、肾功能等。并检测中药组治疗前后IgG、IgA、IgM、C_3、CD_3、CD_4、CD_8、CD_4/CD_8、TXB_2、6—Keto—PGF_(1α)、TPA、PAI、SOD、LPO。结果:中药组总有效率为74.2％,激素组为46.6％,中药组治疗后24小时尿蛋白、血浆白蛋白、血脂均明显改善(P<0.01或P<0.05),激素组治疗前后无明显差异(P>0.05)。中药组治疗后IgG、CD_8、CD_4/CD_8、TPA、TXB_2、LPO、SOD均较前有明显改善(P<0.01或 P<0.05)。     

重组人BAFF112-285的制备及活性分析

目的 制备具有功能活性的重组人TNF家族的B细胞激活因子(B cell activating factor belonging to the TNF family,BAFF)胞外区112—285氨基酸残基段(rhBAFF112—285)，为BAFF的深人研究奠定基础。方法 提取人HL-60细胞总RNA，经RT—PCR扩增编码人BAFF胞外区112—285氨基酸残基(BAFF112—285)的cDNA，行序列测定后，构建于原核表达载体pQE—80L并转化大肠杆菌DH5α，经IPTG诱导表达rhBAFF112—285，Ni^2 —NTA层析纯化，进而经^3H—TdR掺人实验检测其免疫学活性。结果 RT—PC双扩增得到了525bp的DNA片段，序列分析与GenBank中报道的编码人BAFF112—285的cDNA序列一致，该胞外区片段在大肠杆菌中获得了高效表达，表达水平达细菌总蛋白的45．7％，经纯化后纯度可达98．4％，活性检测证实其能明显刺激外周血淋巴细胞活化。结论 利用大肠杆菌可高效表达rhBAFF112—285，所获纯化产物具有生物学活性，所获结果为进一步研究创造了条件。     

Pituitary adenylate cyclase activating polypeptide induces vascular relaxation and inhibits nonvascular smooth muscle activity in the rabbit female genital tract

In vitro effects of two bioactive forms of pituitary adenylate cyclase activating polypeptide (PACAP): PACAP–38 and PACAP–27 were studied on rabbit vascular and non–vascular smooth muscle. Segments of the ovarian artery and muscle strips from the fallopian tube were used. Two series of experiments were performed on vessels: the dose–response relationship of PACAP–38 (10–10–7 M) was established on noradrenaline– (NA, 10–6 M) contracted vessels. In the other set of experiments the contractile effect of 10–8–10–4 M NA added cumulatively, was studied on arterial segments incubated with PACAP–38 (10–7 M), PACAP–27 (10–7 M) or VIP (10–7 M). The effect of PACAP–38, PACAP–27 and VIP (10–10–10–⁶ M) was investigated on spontaneously contracting smooth muscle of the fallopian tube. Longitudinally as well as transversally cut specimens were investigated. PACAP–38 produced a significant dose–related relaxation on the NA–precontracted vessels. However, pre–incubation of the vessels with 10–7 M PACAP–38, PACAP–27 and vaso active intestinal polypeptide (VIP) did not induce a general rightward shift of the NA concentration–response curves, although a tendency to inhibition in the low–dose interval was observed. The peptides caused a significant, dose–dependent inhibition of both frequency and amplitude on the fallopian tube smooth muscle activity. The effects of the three peptides on longitudinally as well as transversally cut specimens were alike.     

补肾活血中药对系膜增生性肾炎大鼠MMP-2表达的影响

目的:研究补肾活血中药对系膜增生性肾小球肾炎的基质金属蛋白酶-2(MMP-2)表达的影响。方法:取雄性SD大鼠54只,实验设正常对照组、模型组、补肾活血药物干预组,观察MMP-2的表达。结果:通过免疫组化、流式细胞术、免疫蛋白印记以及RT-PCR等方法表明模型组大鼠MMP-2表达较正常对照组明显减少,经补肾活血中药治疗后,MMP-2活性可部分恢复。结论:补肾活血中药能上调MMP-2的表达及活性的部分恢复,使得胶原合成减少,在延缓肾小球疾病的进展,防治肾小球硬化方面具有确切疗效。
