目的探讨小胶质细胞活化与慢性前列腺炎疼痛大鼠脊髓兴奋性氨基酸变化的关系。方法完全福氏佐剂和3%角叉菜胶前列腺内注射造成慢性前列腺炎疼痛模型,脊髓插管给药胶质细胞活化抑制剂propento-fylline干扰慢性前列腺炎疼痛模型大鼠,免疫组织化学方法观察正常组、疼痛组、干扰组脊髓节段(L6~S1)背角的小胶质细胞的活化情况,并用氨其酸分析仪测量3组脊髓背解谷氨酸和天门冬氨酸的变化。结果脊髓背角小胶质细胞活化阳性细胞数疼痛组明显增加,干扰组明显减少,疼痛组谷氨酸和天门冬氨酸明显增多,干扰组明显减少。结论小胶质细胞活化是慢性前列腺炎疼痛大鼠脊髓背角兴奋性氨基酸变化的重要原因之一。  相似文献

目的 探讨电针激痛点对慢性肌筋膜疼痛综合征(myofascial pain syndrome,MPS)大鼠L4~6脊髓背角星形胶质细胞神经胶质纤维酸性蛋白(glial fibrillary acidic protein,GFAP)和小胶质细胞特异表达补体受体-3的单克隆抗体(OX-42)表达的影响。方法 将24只雄性SD大鼠随机分为正常组、模型组、电针组,每组8只。采用打击结合离心运动方式建立MPS模型。电针组选取激痛点,电针15min/次,1次/d,治疗7d。采用HE染色法观察大鼠激痛点处肌肉组织形态学变化;肌电图仪检测大鼠激痛点处自发电位变化;免疫组化法检测大鼠L4~6脊髓背角GFAP及OX-42的阳性细胞表达。结果 与正常组相比,模型组大鼠自发电位活动频率显著增多(P<0.05);与模型组相比,电针组自发电频率显著下降(P<0.05)。与正常组比较,模型组GFAP、OX-42阳性细胞表达增多(P<0.05);与模型组比较,电针组GFAP、OX-42阳性细胞表达减少(P<0.05)。结论 电针激痛点可降低MPS大鼠激痛点处自发电活动频率,抑制脊髓背角GFAP、OX-42阳性细胞过表达,其机制或与调节中枢敏化密切相关。  相似文献

目的:观察坐骨神经结扎处脉冲射频(PRF)对慢性坐骨神经压迫损伤(CCI)模型大鼠脊髓背角小胶质细胞和星形胶质细胞活化水平的影响,探讨坐骨神经PRF镇痛机制与脊髓背角小胶质细胞和星形胶质细胞活化水平的关系。方法:雄性SPF级SD大鼠40只随机分为CCI假造模PRF假治疗组(SS组)、CCI假造模PRF治疗组(SP组)、CCI造模PRF假治疗组(CS组)和CCI造模PRF治疗组(CP组),每组10只。CCI造模成功后4 d行PRF治疗,在坐骨神经结扎处行标准PRF (120s、42℃)。于CCI造模前1d (D0)及造模后1、3、5和7 d (D1、D3、D5和D7)测定大鼠患侧机械缩足反射阈(MWT)和热缩足反射潜伏期(TWL)。疼痛行为学测试完成后(D7)取患侧L3~5脊髓背角,Western blotting法检测脊髓背角小胶质细胞特异性蛋白(Iba-1)和胶质原纤维酸性蛋白(GFAP)的蛋白表达水平。结果:与SS组比较,CCI造模后不同时间点CS组大鼠患侧出现足外翻、跛行、脚趾弯曲聚拢和行走时抬足等行为学表现,MWT和TWL均明显下降(P<0.01),脊髓背角Iba-1和GFAP蛋白表达水平明显升高(P<0.05);与CS组比较,CP组大鼠(D5和D7) PRF后足外翻、跛行、脚趾弯曲聚拢和行走时抬足等行为学变化明显缓解,MWT和TWL值明显升高(P<0.01),脊髓背角Iba-1蛋白表达水平明显下调(P<0.05),GFAP蛋白表达水平差异无统计学意义(P>0.05)。结论:坐骨神经PRF能有效缓解CCI模型大鼠的神经病理性疼痛(NP),坐骨神经PRF可抑制脊髓背角小胶质细胞活化水平,其镇痛机制可能与抑制脊髓背角小胶质细胞活化有关。  相似文献

目的 研究坐骨神经损伤后脊髓背角小胶质细胞活化状态和活化类型的变化规律。方法 大鼠随机分为对照组(n=24)、实验组(n=24)。实验组采用结扎坐骨神经主干的方法构建大鼠坐骨神经损伤模型。测量大鼠疼痛行为学数据,于术后第1,7,14天取材,采用免疫荧光染色技术检测大鼠腰段脊髓背角不同激活状态的小胶质细胞变化;通过qRT-PCR验证不同类型小胶质细胞相关标记物的变化趋势。结果 假手术组大鼠在术后14 d内脊髓背角小胶质细胞形态和数量无明显改变,小胶质细胞标记物也无明显变化。术后1 d,CCI大鼠小胶质细胞形态和数量无明显变化,但促炎型(M1型)标记物增加,提示M1型小胶质细胞活化。术后7 d和14 d,CCI大鼠小胶质细胞数量显著增加,标记物检测显示以M1型活化为主,抑炎型(M2型)小胶质细胞活化不明显。结论 大鼠脊髓背角小胶质细胞在坐骨神经损伤后早期即开始活化,活化持续到至少术后两周,在此期间均以M1型小胶质细胞活化为主。  相似文献

目的 探讨神经病理性疼痛大鼠脊髓背角小胶质细胞离子型嘌呤受体(ionotropic purinoceptor,P2X)P2X4的表达及其意义.方法 雌性SD大鼠20只,随机分为坐骨神经保留性损伤(SNI)组和对照组.SNI组大鼠于左后肢制备SNI模型,对照组大鼠仅暴露坐骨神经后立即缝合手术切口.术后观测机械疼痛阈值的变化,于术后第7天断头处死大鼠,截取L4~6段脊髓,分离左右侧脊髓.采用免疫组化法检测脊髓背角P2X4受体及蛋白激酶p38MAPK的表达.结果 与对照组相比,SNI模型组大鼠机械刺激50%缩爪阈值显著降低(P<0.01).SNI模型组大鼠手术侧脊髓背角活化的小胶质细胞表面P2X4受体的表达水平(456.86±32.21)较非手术侧(106.27±12.16)明显增高(P<0.01),同时SNI模型组大鼠手术侧脊髓背角p38MAPK表达水平(399.95±32.42)较非手术侧(123.63±15.47)明显增高(P<0.01).结论 神经病理性疼痛大鼠模型脊髓背角活化小胶质细胞P2X4受体表达增加,可能通过激活p38MAPK促进了疼痛的发生.  相似文献

目的观察神经痛大鼠电针治疗前后脊髓背角与痛觉相关的孤啡肽受体mRNA表达的变化,探讨电针对神经痛的治疗作用。方法30只SD大鼠随机分为3组,每组10只,对照组为正常大鼠,自由生长;神经痛组为大鼠坐骨神经慢性限制性损伤致神经痛模型;电针组为术后第7天给予神经痛大鼠电针治疗30 min。各组动物处死后,取脊髓背角组织,冰冻切片,采用原位杂交的方法,观察各组脊髓背角Ⅰ~Ⅵ层内与痛觉相关孤啡肽受体mRNA的变化。结果大鼠坐骨神经结扎后出现痛敏,电针能明显抑制大鼠痛敏,与神经痛组比较缩爪潜伏期(PWL)明显延长(P<0.01);对照组脊髓背角Ⅰ~Ⅵ层有少量孤啡肽mRNA阳性细胞,神经痛组大鼠术后第7天时脊髓背角孤啡肽mRNA阳性细胞数明显多于对照组,2组比较差异具有显著性(P<0.001);电针组脊髓背角Ⅰ~Ⅱ层、Ⅴ~Ⅵ层孤啡肽mRNA阳性细胞数与神经痛组比较,差异具有显著性(P<0.05)。Ⅲ~Ⅳ层孤啡肽mRNA阳性细胞数与神经痛组比较无显著性差异(P>0.05)。结论电针对神经痛的治疗作用可能与脊髓背角内孤啡肽受体有关。  相似文献

目的:研究癫痫大鼠海马小胶质细胞在癫痫持续状态后不同时间点激活的特点。方法:采用锂-匹罗卡品癫痫大鼠及免疫组化方法比较不同时间点OX-42阳性细胞的增殖及形态变化。结果:小胶质细胞在癫痫持续状态(SE)后被激活;OX-42阳性细胞在SE 3~7 d时增加达到一个峰值,7~14 d时细胞的增加处于一个相对稳定状态,而14 d以后增加的OX-42阳性细胞逐渐下降,大约在21 d时细胞数恢复到较先前稍高的水平。结论:癫痫发作后的炎症病理损伤与小胶质细胞的增殖活化密切相关,小胶质细胞的激活是构成癫痫炎症损伤机制的重要环节。  相似文献

目的:建立并评估可量化型小鼠坐骨神经缩窄性神经病理痛模型。方法:用分级坐骨神经结扎的方法建立动物模型,即将小鼠随机分为N0、N1、N2、N4组,暴露坐骨神经主干部位,N1、N2、N4组小鼠分别结扎坐骨神经1、2、4道,每个结之间间隔1 mm;N0组只游离神经,不作任何结扎。通过测定小鼠右后爪热痛阈值,电镜观察坐骨神经超微结构,免疫组化检测小胶质细胞OX-42的荧光积分光密度值(IOD),酶联免疫吸附(ELISA)法测定脊髓白介素(IL-1β、IL-6)和肿瘤坏死因子(TNF-α)的浓度对模型进行评估。结果:与N0组相比,N1、N2和N4组术后热痛阈值较术前明显下降,坐骨神经纤维广泛洋葱化改变,轴索溶解或消失,雪旺氏细胞水肿、坏死,异常髓鞘所占比例增加,脊髓IL-1β、IL-6和TNF-α表达不同程度上升;N2、N4组脊髓小胶质细胞OX-42的IOD显著增加。结论:小鼠可量化型坐骨神经缩窄模型呈现不同程度的神经病理性疼痛、坐骨神经不同程度的髓鞘病变、脊髓小胶质细胞活化及脊髓IL-1β、IL-6和TNF-α表达增加;这一模型的建立可为神经病理性疼痛等的研究提供更利于准确评估的病理模型。  相似文献

目的:探讨改良第二跖趾关节修复掌指关节背侧缺损的应用解剖。方法:选取就诊的掌指关节背侧缺损患者共68例,将患者随机分为两组各34例,A组行改良第二跖趾关节修复治疗,B组行拇短伸肌半腱移位修复,对比两组患者随访痊愈率和并发症等现象。结果:术后3个月,A组优9例,良24例,差1例,优良率为91.18%;B组优8例,良18例,差8例,优秀率为76.47%;组间比较具有统计学意义(P<0.05)。结论:改良第二跖趾关节修复治疗掌指关节背侧缺损能取得满意的效果,有效提高痊愈率,缩短患者康复时间,让患者早日恢复活动能力,具有积极的临床推广意义。  相似文献

<正>209352 Local X-irradiation promotes regeneration of spinal cord central nervous system in a rat model/Shen Yixin(沈忆新,Dept Orthop,2nd Affil Hosp Suzhou Univ,Suzhou 215004)…∥Chin J Surg.-2009,47(12).-934~936Objective To evaluate the effect of local X-irradiation on spinal cord injury by using physiology,kinology,electrophysiology and histology method.Methods 46 female Spargen-Dawley rats were subjectied to spinal cord injury by weight dropping on T11-12.All animals were divided ito 3 groups randomly.One of the animal groups was irradiated with 10 Gy at the lesion site;another was irradiated with 20 Gy,the other without irradiation is regarded as sham-group.The animals were euthanized at different time points at 4 and 12 weeks after irradiation.Spinal cord callus was assessed by using physiology,kinology,and electrophysiology and histology method.Results In all the groups,the NF at 14 weeks were found higher than that of 6 weeks.Both 10 Gy irradiated and 20 Gy irradiated groups were higher than those of group at each time point (P<0.05).The MBP decreased at 14 weeks in irrdiated groups(P<0.05),but increases at 14 weeks in sham-group(P<0.05),the MBP of irradiated groups was lower than that of sham group at 14 weeks (P<0.05).The GFAP and Nogo-A at 14 weeks were higher than that at 6 weeks in all the groups (P<0.05),and there was no statistical significance with physiology,kinology,electrophysiology test in all groups.Conclusion A self-repair mechanism exists after SCI,which will last at least 14 weeks.Local irradiation promotes the regeneration of spinal cord system after injury to some extent.11 refs,2 tabs.  相似文献
