旨在建立人截短型IL-6原核高效表达系统。用PCR方法获得截短的人IL-6基因(rhIL-6cDNA),将其插入克隆载体pUC18中,经证实后再克隆入表达载体pBV220中,鉴定阳性表达载体。结果建立了截短型IL-6基因的克隆载体,经测序证实完全正确;构建了截短型IL-6的原核表达质粒pBV220/rhIL-6并获得高效表达;初步纯化的表达蛋白的比活性为5×10  相似文献

采用PCR技术从人肝脏cDNA文库扩增出编码114~281位氨基酸的TRAIL基因片段,将之克隆至原核表达载体pET-30a、pQE-30和pJLA503中,上述重组载体经IPTG诱导表达后,SDS-PAGE鉴定显示pET-30a系统表达效率最高,目的蛋白占菌体总蛋白含量的40%左右;采用透析法或梯度稀释法对TRAIL胞外区片段包涵体进行复性,非还原SDS-PAGE的结果显示梯度稀释法明显好于透析法;纯化后的TRAIL胞外区片段蛋白具有明显的生物学活性,效果与标准品相似;通过添加不同的折叠促进剂对复性效率影响的研究,确立了TRAIL胞外区片段适宜的复性方法,复性效率达75%~80%.  相似文献

目的:构建重组人白细胞介素7(rhIL-7)的原核表达载体,在大肠杆菌中表达并鉴定.方法:以RT-PCR方法扩增编码IL-7的cDNA片段,插入至pET-3d载体中,构建rhIL-7的原核表达载体,阳性克隆经测序鉴定,转化至大肠杆菌BL21(DE3),优化rhIL-7表达条件,利用免疫印迹鉴定重组蛋白.结果:重组载体经DNA测序显示包含正确的rhIL-7编码序列,重组载体转入大肠杆菌后,经IPTG诱导后外源蛋白表达,相对分子质量为17 400,与IL-7理论分子质量一致;免疫印迹显示,外源蛋白可以被人IL-7特异性抗体识别,表明在原核表达的外源蛋白是rhIL-7.rhIL-7主要表达于包涵体中,包涵体表达优化条件为:温度为37 ℃、IPTG浓度为0.4 mmol/L、诱导时间为4 h.结论:成功的构建了rhIL-7的原核表达载体.  相似文献

目的 :探讨重组人碱性成纤维细胞生长因子 (rhbFGF)的纯化工艺和活性鉴定 .方法 :将rhbFGF工程菌大量扩增后通过包涵体提取、复性、阳离子交换、凝胶过滤等技术进行纯化 .结果 :纯化后的rhbFGF ,相对分子质量为 17× 10 3,蛋白纯度为 95 %以上 ,比活为 1 7× 10 6 U/mg ,对NIH3T3细胞具有明显的促分裂活性 .结论 :通过复性和两步层析的纯化方法可获得高纯度、高回收率和高生物活性的rhbFGF ,可用于实验室研究及临床试验  相似文献

应用RT-PCR和搭桥PCR技术,扩增肠激酶催化亚基的cDNA,测序正确后克隆至酵母分泌型表达质粒p9K中.将重组质粒p9K/EKLC转化至表达宿主pichia pastoris GS115,筛选得到高效表达重组牛肠激酶催化亚基工程菌.经甲醇诱导,目标蛋白肠激酶催化亚基(EKLC)表达并分泌至酵母培养基中.高密度发酵后,经超滤浓缩,采用SP Sepharose F.F一步纯化得到纯度达98%以上的重组目的蛋白,活性达到2.3 U/uL.上述结果表明,本实验成功获得了肠激酶催化亚基,它是一种能将融合蛋白中目标蛋白与载体蛋白裂解释放的有效工具酶.  相似文献

为了获得大量重组血管内皮生长因子(VEGF-A_(189))蛋白的可溶性表达,对重组VEGF-A_(189)工程菌进行发酵条件的优化并开展发酵研究.通过摇瓶培养进行可溶性发酵条件优化,在5 L发酵罐中发酵重组VEGF-A_(189)工程菌,将发酵产物经镍离子柱亲和层析纯化,并对发酵产物进行SDS-PAGE,Western Blot和ELISA等分析鉴定.结果表明,重组VEGF-A_(189)工程菌可溶性发酵条件为:37℃培养重组VEGF-A_(189)工程菌4 h,加入0.3 mmol/L的异丙基-β-D-硫代半乳糖苷(IPTG)于28℃诱导表达4 h.在5 L发酵罐中按比例放大发酵,在补料发酵3.5 h后菌体密度(A_(600nm))达到8.0,调整发酵温度到28℃,IPTG诱导4 h后的菌体密度为13.72,菌体产量为40 g/L.镍离子柱亲合层析纯化后重组VEGF-A_(189)纯度达到90%以上,回收率大于80%,Western Blot、ELISA分析结果显示,重组VEGF-A_(189)蛋白和抗VEGF抗体有良好的结合活性.结果表明经过发酵条件优化后发酵可获得有免疫结合活性的可溶性重组VEGF-A_(189)蛋白.  相似文献

在重组质粒pExSecI-IGF-Ⅰ的基础上,采用基因克隆方法合成了长链人胰岛素样生长因子(Long R3-IGF-Ⅰ)基因序列,构建基于该基因的重组原核表达质粒.将重组质粒转入E.coli,并对其表达条件进行了优化.实验结果表明,在37 ℃,IPTG终浓度0.6 mmol/L,诱导3 h条件下,Long R3-IGF-Ⅰ以包涵体形式可得到高效表达.将包涵体变性溶解后通过分离纯化,目的蛋白纯度达到95%以上.通过尿素梯度透析复性法对变性包涵体复性,复性率达到66%.  相似文献

采用大肠杆菌(Escherichia coli)作为宿主,通过300L发酵罐进行中试发酵表达重组人白细胞介素-13(rhIL-13)融合蛋白;经异丙基-β-D-硫代吡喃半乳糖苷(IPTG)诱导,离心发酵液收集菌体,菌体经溶解、变性、稀释复性及亲和层析捕获,获得融合蛋白;融合蛋白经肠激酶酶切后,再经亲和层析分离和分子筛精细纯化等步骤,获得高纯度rhIL-13原液.每升发酵液可获得高纯度的rhIL-13原液约30mg.通过本纯化工艺获得的rhIL-13原液经十二烷基硫酸钠-聚丙烯酰胺凝胶电泳(SDS-PAGE)法检测,纯度大于98.75%;经高效液相色谱分子排阻(HPLC-SEC)法检测,纯度为100%;质谱(MS)法测定rhIL-13分子质量为12 348u,与理论值基本一致;采用噻唑蓝(MTT)法进行比活性检测,比活大于8.0×106 IU/mg;采用HPLC-SEC法和MS法对rhIL-13二硫键数目和位置进行分析,结果显示二硫键数目和位置与理论一致.此操作方法可获得高纯度、高活性的rhIL-13原液,并且操作简单,易于放大,可实现工业化规模生产.  相似文献

为提高重组表达质粒pET32c-ZGA在大肠杆菌中的表达量,对表达重组风雨花凝集素基因工程菌的培养条件进行优化筛选,对影响蛋白质表达量和工程菌生长的因素如pH值、诱导温度、IPTG、接种量、诱导时间、补充碳源形式、诱导时机、无机离子等进行了探索.经过优化,重组融合目的蛋白占菌体总蛋白的61.8%,比优化前提高了13.4%.条件优化后,pET32c-ZGA工程菌经过放大培养,获得6 g/L的湿菌体量,超声波破碎菌体,SDS-PAGE证实重组融合蛋白以包涵体形式表达.  相似文献

旨在利用原核表达系统制备可溶性的PD-1胞外段(以下简称exPD-1)蛋白.以C57BL/6小鼠脾细胞cDNA为模板,扩增小鼠exPD-1基因,成功构建重组质粒pET-28a-exPD-1,并在E.coliBL21(DE3)中诱导表达.结果显示,小鼠exPD-1重组蛋白大小约为17kD,以包涵体形式存在.exPD-1包涵体蛋白经8mol/L尿素变性、Ni-NTA亲和层析后,采用尿素梯度透析法对蛋白进行复性,复性效果不佳,添加氧化/还原型谷胱甘肽复性后,复性率达到50%以上,提高了复性效率.小鼠exPD-1蛋白的成功制备,为进一步研究PD-1的免疫学功能奠定了材料基础.  相似文献

The Williston Basin is a significant petroleum province, containing oil production zones that include the Middle Cambrian to Lower Ordovician, Upper Ordovician, Middle Devonian, Upper Devonian and Mississippian and within the Jurassic and Cretaceous. The oils of the Williston Basin exhibit a wide range of geochemical characteristics defined as "oil families", although the geochemical signature of the Cambrian Deadwood Formation and Lower Ordovician Winnipeg reservoired oils does not match any "oil family". Despite their close stratigraphic proximity, it is evident that the oils of the Lower Palaeozoic within the Williston Basin are distinct. This suggests the presence of a new "oil family" within the Williston Basin. Diagnostic geochemical signatures occur in the gasoline range chromatograms, within saturate fraction gas chromatograms and biomarker fingerprints. However, some of the established criteria and cross-plots that are currently used to segregate oils into distinct genetic families within the basin do not always meet with success, particularly when applied to the Lower Palaeozoic oils of the Deadwood and Winnipeg Formation.  相似文献

Language is a means of verbal communication. People use language to communicate with each other. In the society, no two speakers are exactly alike in the way of speaking. Some differences are due to age, gender, statue and personality. Above all, gender is one of the obvious reasons. The writer of this paper tries to describe the features of women's language from these perspectives: pronunciation, intonation, diction, subjects, grammar and discourse. From the discussion of the features of women's language, more attention should be paid to language use in social context. The discovery of the prolific Ordovician Red River reservoirs in 1995 in southeastern Saskatchewan was the catalyst for extensive exploration activity which resulted in the discovery of more than 15 new Red River pools. The best yields of Red River production to date have been from dolomite reservoirs. Understanding the processes of dolomitization is, therefore, crucial for the prediction of the connectivity, spatial distribution and heterogeneity of dolomite reservoirs.The Red River reservoirs in the Midale area consist of 3~4 thin dolomitized zones, with a total thickness of about 20 m, which occur at the top of the Yeoman Formation. Two types of replacement dolomite were recognized in the Red River reservoir: dolomitized burrow infills and dolomitized host matrix. The spatial distribution of dolomite suggests that burrowing organisms played an important role in facilitating the fluid flow in the backfilled sediments. This resulted in penecontemporaneous dolomitization of burrow infills by normal seawater. The dolomite in the host matrix is interpreted as having occurred at shallow burial by evaporitic seawater during precipitation of Lake Almar anhydrite that immediately overlies the Yeoman Formation. However, the low δ18O values of dolomited burrow infills (-5.9‰~ -7.8‰, PDB) and matrix dolomites (-6.6‰~ -8.1‰, avg. -7.4‰ PDB) compared to the estimated values for the late Ordovician marine dolomite could be attributed to modification and alteration of dolomite at higher temperatures during deeper burial, which could also be responsible for its 87Sr/86Sr ratios (0.7084~0.7088) that are higher than suggested for the late Ordovician seawaters (0.7078~0.7080). The trace amounts of saddle dolomite cement in the Red River carbonates are probably related to "cannibalization" of earlier replacement dolomite during the chemical compaction.  相似文献

以AC-13级配为基础,将橡胶颗粒代替部分集料掺入混合料中,以低温弯曲试验为评价方法对不同橡胶颗粒掺量下沥青混合料的低温抗裂性进行研究,并引入应变能密度值对混合料的低温抗裂性进行综合评价.试验结果表明:橡胶颗粒沥青混合料试件的破坏微应变均超过2 300,满足冬寒区的技术指标;无论是否掺加橡胶颗粒,随着温度的下降,沥青混合料破坏时的最大弯拉强度增大,弯拉应变降低,劲度模量增大;弯曲应变能密度在胶粒掺量为1%左右时具有较大的弯曲应变能密度值,此时橡胶颗粒沥青混合料具有较好的低温抗裂性.  相似文献

理论推导与室内实验相结合,建立了低渗透非均质砂岩油藏启动压力梯度确定方法。首先借助油藏流场与电场相似的原理,推导了非均质砂岩油藏启动压力梯度计算公式。其次基于稳定流实验方法,建立了非均质砂岩油藏启动压力梯度测试方法。结果表明:低渗透非均质砂岩油藏的启动压力梯度确定遵循两个等效原则。平面非均质油藏的启动压力梯度等于各级渗透率段的启动压力梯度关于长度的加权平均;纵向非均质油藏的启动压力梯度等于各渗透率层的启动压力梯度关于渗透率与渗流面积乘积的加权平均。研究成果可用于有效指导低渗透非均质砂岩油藏的合理井距确定,促进该类油藏的高效开发。  相似文献

Quality traits in wheat (Triticum aestirum L.) were studied by quantitative trait locus (QTL) analysis in a recombinant inbred line (RIL) population, a set of 131 lines derived from Chuan 35050 × Shannong 483 cross (ChSh). Grains from RILs were assayed for 21 quality traits related to protein and starch. A total of 35 putative QTLs for 19 traits with a single QTL explaining 7.99-40.52% of phenotypic variations were detected on 10 chromosomes, 1D, 2A, 2D, 3B, 3D, 5A, 6A, 6B, 6D, and 7B. The additive effects of 30 QTLs were positive, contributed by Chuan 35050, the remaining 5 QTLs were negative with the additive effect contributed by Shannong 483. For protein traits, 15 QTLs were obtained and most of them were located on chromosomes 1 D, 3B and 6D, while 20 QTLs for starch traits were detected and most of them were located on chromosomes 3D, 6B and 7B. Only 7 QTLs for protein and starch traits were co-located in three regions on chromosomes 1D, 2A and 2D. These protein and starch trait QTLs showed a distinct distribution pattern in certain regions and chromosomes. Twenty-two QTLs were clustered in 6 regions of 5 chromosomes. Two QTL clusters for protein traits were located on chromosomes 1D and 3B, respectively, three clusters for starch traits on chromosomes 3D, 6B and 7B, and one cluster including protein and starch traits on chromosome 1D.  相似文献
