黄铜矿表面生物吸附量的测定条件   总被引:2,自引:1,他引:1  
采用茚三酮比色法测定了在细菌浸矿的过程中,黄铜矿矿物表面吸附细菌细胞裂解液的吸光度,通过对测量波长、裂解时间、溶液pH值、加热时间、冷却时间等影响吸光度因素的实验研究,得出了黄铜矿表面细菌细胞裂解液吸光度的最佳测定条件为:波长560nm,裂解时间25min,溶液pH值7.0,加热时间15min,冷却时间6min.在最佳条件下绘制出了吸光度与矿物表面细菌吸附量之间关系的标准曲线.研究表明细菌裂解液的吸光值与细菌的浓度成正比.  相似文献

实验方法在两支25mL比色管中,分别加入1.0mL 1.0×10-3mol/L靛蓝胭脂红溶液、1.0 mLpH5.4六次甲基四胺-HCl溶液;在第一支比色管内加入一定量Al3+溶液,在两支比色管中加入2 mL0.02mol/L(NH4)2S2O8溶液、2.0mL5 g/LTritonX-100溶液;用水稀释至刻度,摇匀,放入(30±0.5)℃恒温水浴中加热30 min.以水作参比,在608 nm处,测量有Al3+反应液的吸光度为A,无Al3+反应液的吸光度为A0,计算△A=A0-A的值.  相似文献

D-核糖发酵液离心洗涤得菌体,采用超声波破碎(在40kW下,工作4s,间歇4s,破碎90次),制备无细胞抽提液.取无细胞抽提液80μL及100μmol/L葡萄糖、4μmol/LNADP、0.6mmol/LTris-HCl(pH=6.8)、10μmol/LMnSO4溶液各1mL,在37℃下保温10min,测定A340num的值,对比NADPH标准曲线,测定D-葡萄糖脱氢酶的酶含量.根据测定的时间及酶蛋白的含量,从而确定D-葡萄糖脱氢酶的酶活力的测定方法.  相似文献

采用苯酚-硫酸方法测定HQ0211菌裂解液中多糖含量,得出最佳的裂解时间为25 min,最佳吸收波长为430 nm,最佳冷却时间为10 min.在利用HQ0211菌对含砷金矿进行吸附氧化试验研究时,微生物细胞多糖含量随着溶液体系电位的变化而变化.在480~572 mV阶段,游离在溶液中的细菌的多糖质量浓度为0.033~0.123 mg/mL,吸附在含砷金矿石矿物上的细菌细胞多糖为5.3~8.9 mg/g;当电位达到638 mV时,溶液中游离的细菌细胞多糖质量浓度达1.155 mg/mL,吸附在矿物上的细菌细胞多糖可高达57.5 mg/g.  相似文献

研究了用坂口试剂-α-萘酚定量测定L-Arg的方法.确立了L-Arg水溶液和发酵液L-Arg含量测定的最佳条件分别为(1)0.0125mol/LNaOH0.8mL、0.00138mol/Lα-萘酚90μL、NaClO120μL,40℃水浴显色15min后,于室温下40min内测定;(2)0.0125mol/LNaOH0.8mL、0.00138mol/Lα-萘酚150μL、NaClO280μL,40℃水浴显色5min后置于冰水混合物中,40min内测定.根据发酵液L-Arg测定标准曲线,推导出其L-Arg含量的计算公式为X=117.65×A×N.  相似文献

基于Berthelot显色反应,采用正交设计法优化了测定大肠杆菌工程菌谷氨酸脱羧酶转化L-谷氨酸生成γ-氨基丁酸含量的条件.实验确定的最佳测定条件为:2.0mL 0.2mol/L pH6.0的醋酸缓冲液(内含0.1mmol/L的PLP,0.2mol/L的L-谷氨酸,稀释70倍),再依次加0.2mmol/L的硼酸缓冲液(pH9.0)1.0mL,0.5mL 6.0%重蒸苯酚和5.0mL 10%次氯酸钠,沸水浴加热10min后,迅速冰浴20min,待溶液出现蓝绿色后,加入60%(v/v)乙醇溶液4.0mL在640nm测定吸光值,绘制出标准曲线来计算GABA的含量.结果表明,该方法简便实用,相对误差小,适合实验室样品的快速检测.  相似文献

在0.32mol/L H2SO4溶液中,维生索B1在80℃水浴恒温条件下与KMnO4发生氧化还原反应.采用分光光度法在525 nm处测定KMnO4的吸光度变化值△A.△A值与维生素B1的浓度在4~40μg/mL范围内成良好的线性关系,相关系数R=0.9987,方法检出限为0.8 μg/mL.将该法用于药片中维生素B1含量的测定,相对标准偏差RSD为5.2%,平均回收率为92%,结果满意.  相似文献

采用氯化锌-软锰矿活化法制备核桃壳活性炭并研究其对废水中苯酚的吸附特性,结果表明:软锰矿的投加量占原料的5%、氯化锌浓度为3 mol/L、剂料比为1、活化温度600℃、活化时间10min是活性炭的最佳制备条件。在此条件下亚甲基蓝脱色力是123mL/g,碘吸附值945mg/g。在18℃、pH=2条件下,0.5g核桃壳活性炭对50mL的50mg/L苯酚溶液吸附240min吸附效果最佳,吸附效果优于市煤质活性炭。  相似文献

The Williston Basin is a significant petroleum province, containing oil production zones that include the Middle Cambrian to Lower Ordovician, Upper Ordovician, Middle Devonian, Upper Devonian and Mississippian and within the Jurassic and Cretaceous. The oils of the Williston Basin exhibit a wide range of geochemical characteristics defined as "oil families", although the geochemical signature of the Cambrian Deadwood Formation and Lower Ordovician Winnipeg reservoired oils does not match any "oil family". Despite their close stratigraphic proximity, it is evident that the oils of the Lower Palaeozoic within the Williston Basin are distinct. This suggests the presence of a new "oil family" within the Williston Basin. Diagnostic geochemical signatures occur in the gasoline range chromatograms, within saturate fraction gas chromatograms and biomarker fingerprints. However, some of the established criteria and cross-plots that are currently used to segregate oils into distinct genetic families within the basin do not always meet with success, particularly when applied to the Lower Palaeozoic oils of the Deadwood and Winnipeg Formation.  相似文献

The discovery of the prolific Ordovician Red River reservoirs in 1995 in southeastern Saskatchewan was the catalyst for extensive exploration activity which resulted in the discovery of more than 15 new Red River pools. The best yields of Red River production to date have been from dolomite reservoirs. Understanding the processes of dolomitization is, therefore, crucial for the prediction of the connectivity, spatial distribution and heterogeneity of dolomite reservoirs.The Red River reservoirs in the Midale area consist of 3~4 thin dolomitized zones, with a total thickness of about 20 m, which occur at the top of the Yeoman Formation. Two types of replacement dolomite were recognized in the Red River reservoir: dolomitized burrow infills and dolomitized host matrix. The spatial distribution of dolomite suggests that burrowing organisms played an important role in facilitating the fluid flow in the backfilled sediments. This resulted in penecontemporaneous dolomitization of burrow infills by normal seawater. The dolomite in the host matrix is interpreted as having occurred at shallow burial by evaporitic seawater during precipitation of Lake Almar anhydrite that immediately overlies the Yeoman Formation. However, the low δ18O values of dolomited burrow infills (-5.9‰~ -7.8‰, PDB) and matrix dolomites (-6.6‰~ -8.1‰, avg. -7.4‰ PDB) compared to the estimated values for the late Ordovician marine dolomite could be attributed to modification and alteration of dolomite at higher temperatures during deeper burial, which could also be responsible for its 87Sr/86Sr ratios (0.7084~0.7088) that are higher than suggested for the late Ordovician seawaters (0.7078~0.7080). The trace amounts of saddle dolomite cement in the Red River carbonates are probably related to "cannibalization" of earlier replacement dolomite during the chemical compaction.  相似文献

以AC-13级配为基础,将橡胶颗粒代替部分集料掺入混合料中,以低温弯曲试验为评价方法对不同橡胶颗粒掺量下沥青混合料的低温抗裂性进行研究,并引入应变能密度值对混合料的低温抗裂性进行综合评价.试验结果表明:橡胶颗粒沥青混合料试件的破坏微应变均超过2 300,满足冬寒区的技术指标;无论是否掺加橡胶颗粒,随着温度的下降,沥青混合料破坏时的最大弯拉强度增大,弯拉应变降低,劲度模量增大;弯曲应变能密度在胶粒掺量为1%左右时具有较大的弯曲应变能密度值,此时橡胶颗粒沥青混合料具有较好的低温抗裂性.  相似文献

理论推导与室内实验相结合,建立了低渗透非均质砂岩油藏启动压力梯度确定方法。首先借助油藏流场与电场相似的原理,推导了非均质砂岩油藏启动压力梯度计算公式。其次基于稳定流实验方法,建立了非均质砂岩油藏启动压力梯度测试方法。结果表明:低渗透非均质砂岩油藏的启动压力梯度确定遵循两个等效原则。平面非均质油藏的启动压力梯度等于各级渗透率段的启动压力梯度关于长度的加权平均;纵向非均质油藏的启动压力梯度等于各渗透率层的启动压力梯度关于渗透率与渗流面积乘积的加权平均。研究成果可用于有效指导低渗透非均质砂岩油藏的合理井距确定,促进该类油藏的高效开发。  相似文献

Quality traits in wheat (Triticum aestirum L.) were studied by quantitative trait locus (QTL) analysis in a recombinant inbred line (RIL) population, a set of 131 lines derived from Chuan 35050 × Shannong 483 cross (ChSh). Grains from RILs were assayed for 21 quality traits related to protein and starch. A total of 35 putative QTLs for 19 traits with a single QTL explaining 7.99-40.52% of phenotypic variations were detected on 10 chromosomes, 1D, 2A, 2D, 3B, 3D, 5A, 6A, 6B, 6D, and 7B. The additive effects of 30 QTLs were positive, contributed by Chuan 35050, the remaining 5 QTLs were negative with the additive effect contributed by Shannong 483. For protein traits, 15 QTLs were obtained and most of them were located on chromosomes 1 D, 3B and 6D, while 20 QTLs for starch traits were detected and most of them were located on chromosomes 3D, 6B and 7B. Only 7 QTLs for protein and starch traits were co-located in three regions on chromosomes 1D, 2A and 2D. These protein and starch trait QTLs showed a distinct distribution pattern in certain regions and chromosomes. Twenty-two QTLs were clustered in 6 regions of 5 chromosomes. Two QTL clusters for protein traits were located on chromosomes 1D and 3B, respectively, three clusters for starch traits on chromosomes 3D, 6B and 7B, and one cluster including protein and starch traits on chromosome 1D.  相似文献
