不同浓度的葡萄糖对SW480肿瘤细胞分泌IGF-1、TGF-β1的影响

摘    要:目的研究不同浓度葡萄糖对SW480肿瘤细胞胰岛素样生长因子1(IGF—1)、转化生长因子β1(TGF-β1)表达的影响,探讨糖尿病发生结直肠癌的相关因素。方法体外常规培养SW480肿瘤细胞。根据所用培养液外加葡萄糖浓度的不同,将实验所用细胞共分6组:0组(对照组)为RPMI1640培养液;1组为RPMI1640培养液+5mmol/L葡萄糖;2组为RPMI1640培养液+10mmol/L葡萄糖;3组为RPMI1640培养液+15mmol/L葡萄糖;4组为RPMI1640培养液+20mmol/L葡萄糖;5组为RPMI1640培养液+25mmol/L葡萄糖。每组设3个复孔,置于37℃、5%CO2饱和湿度的培养箱内培养48h,培养结束后收集细胞培养上清,ELISA法检测IGF-1、TGF—β1的浓度。结果①不同浓度的葡萄糖组IGF-1的表达无显著性差异(F=0.192,P均〉0.05)。②4组、5组TGF—β1的浓度明显高于0组,有显著性差异(F=26.036,P均〈0.01)。结论葡萄糖不能够上调SW480肿瘤细胞IGF-1的表达,但高浓度的葡萄糖能够上调SW480肿瘤细胞TGF—β1的表达。

Effect of different concentrations of glucose on secretions of IGF - 1 and TGF - β1 in SW480 tumor cells

Abstract:Objective It is to observe the effect of different concentrations of glucose on the secretions of insulin and insufin-like growth factor - 1 ( IGF - 1 ) and transforming growth factor ( TGF - β1 ) in SW480 tumor cells, and approach the relevant factors of development of colorectal cancer in diabetes mellitus. Methods SW480 tumor cells were cultured in vitro. Ac- cording to the concentrations of glucose in culture medium, the cells were divided into six groups: group 0 with RPMI1640, group 1 with RPMI1640 +5 mmol/L glucose, group 2 with RPMI1640 + 10 mmol/L glucose, group 3 with RPMI1640 + 15 mmol/L glucose, group 4 with RPMI1640 + 20 mmol/L glucose, group 5 with RPMI1640 + 25 mmol/L glucose. After added to different culture medium, the cells were cultured 48h, supernatant were collected. The concentrations of IGF- l and TGF- β1 in the supernatant were detected by specific enzyme-linked immunosorbent assay (ELISA). Results ①There was no signifi- cant difference in IGF - 1 level in SW480 tumor cells among groups of different concentrations of glucose ( all P 〉 0.05 ). ② The levels of TGF - β1 in group 4 and group 5 was significantly higher than that in group 0 (all P 〈0.01 ). Conclusion Glu- cose has no effect on the secretion of IGF - 1 in SW480 tumor cells. High concentration of glucose may up-regulate the secretion of TGF - 131 in SW480 tumor cells.
