奥美沙坦酯对自发性高血压大鼠血清蛋白质表达谱的影响

摘    要:目的探讨奥美沙坦酯对自发性高血压大鼠(SHR)降压作用的蛋白质组学机制。方法选取雌雄各半12周龄的SHR,随机分为对照组(n=10)、奥美沙坦酯组(n=10)。对照组给予0.9%生理盐水灌胃,奥美沙坦酯组给予奥美沙坦酯配制的混悬液灌胃,两组均连续灌胃四周后,分离血清,每组随机各取5份血清样品。基质辅助激光解析电离飞行时间质谱(MALDI-TOF-MS)测定肽质量指纹图谱(PMF),采用数据库检索鉴定各肽段对应的蛋白质,获取差异表达蛋白质的信息。结果对照组、奥美沙坦酯组凝胶上的平均蛋白点数分别为(476±30)、(508±55)个,两组经统计分析有20个显著差异点,奥美沙坦酯干预后表达增强13个,降低5个,蛋白点消失2个。对这20个差异蛋白点进行质谱分析,鉴定出的8个蛋白质分别为线粒体膜蛋白、锌指蛋白、T细胞受体、视黄醇结合蛋白、转甲状腺蛋白A链、Rho GTPase、假想蛋白(2个)。结论奥美沙坦酯对高血压的作用机制可能与线粒体膜蛋白、锌指蛋白、T细胞受体、转甲状腺蛋白A链高表达,视黄醇结合蛋白、Rho GTPase低表达有关。
