H2滴眼液对苯扎溴铵导致的小鼠眼表损伤的预防作用

摘    要:目的 探讨H2滴眼液对苯扎溴铵导致的小鼠眼表损伤的预防作用。方法 选取SPF级BALB/c雄性小鼠30只,均取小鼠右眼作为实验眼。将30只小鼠随机分为实验组和对照组,各12只,其余的6只则作为空白对照不作任何处理。实验组采用1 g·L-1苯扎溴铵滴眼液+H2滴眼液滴眼,对照组采用1 g·L-1苯扎溴铵滴眼液+生理盐水滴眼,中间均间隔10 min,每天4次。分别在干预前及干预后1 d、7 d、14 d观察各组小鼠的泪膜破裂时间(tear break-up time,BUT)、基础泪液分泌试验(schirmer Ⅰ test,S Ⅰ t)及角膜荧光素染色(fluorescent test,FL)评分,并于干预14 d后取小鼠眼球,利用电子显微镜对角膜上皮进行观察与分析。结果 干预1 d后,实验组和对照组S Ⅰ t干预前后相比,差异均无统计学意义(均为P>0.05),两组的S Ⅰ t相比,差异亦无统计学意义(P>0.05);干预7 d、14 d后,对照组S Ⅰ t与干预前相比明显减小,差异均有统计学意义(均为P<0.05);两组S Ⅰ t相比,差异均有统计学意义(均为P<0.05),实验组S Ⅰ t均大于对照组。干预1 d后,两组BUT干预前后相比,差异均无统计学意义(均为P>0.05),两组BUT相比,差异亦无统计学意义(P>0.05);干预7 d、14 d后,对照组BUT与干预前相比明显减小,差异均有统计学意义(均为P<0.05);两组BUT相比,差异均有统计学意义(均为P<0.05),实验组BUT均长于对照组。实验组干预1~14 d角膜变化不明显,对照组干预1~14 d后FL染色区域增加。干预后 4 d 和 7 d,两组FL评分差异均有统计学意义(均为P<0.05),对照组均高于实验组。实验组角膜上皮细胞表面可见密集排列的微绒毛和微皱襞,而对照组角膜上皮细胞表面微绒毛、微皱襞部分平坦或出现融合、缺失。结论 H2滴眼液能通过保持角膜上皮的完整性从而改善干眼症。

Abstract:Objective To investigate the preventive effect of H2 eye drops on ocular surface damage induced by benzalkonium bromide in mice.Methods Thirty male BALB/c mice of SPF grade were selected.The right eyes of all mice were taken as experimental eyes.The 30 mice were randomly divided into an experimental group and a control group,12 in each group,and the remaining 6 were treated as blank controls without any treatment.The experimental group was treated with 1 g·L-1 benzalkonium bromide eye drops+H2 eye drops,while the control group was treated with 1 g·L-1 benzalkonium bromide eye drops+saline eye drops,with an interval of 10 minutes,four times a day.The tear break-up time (BUT),Schirmer Ⅰ test (S Ⅰ t) and corneal fluorescent test (FL) scores were observed before and 1 day,7 days and 14 days after intervention.The eyeballs of the mice were taken on day 14 after intervention,and the corneal epithelium was observed and analyzed by electron microscopy.Results After one day of intervention,there was no significant difference in S Ⅰ t between the two groups before and after intervention(P>0.05),there was no significant difference in S Ⅰ t between the two groups (P>0.05).After 7 days and 14 days of intervention,the S Ⅰ t of the control group decreased significantly compared with that before intervention,and the differences were statistically significant (both P<0.05),and there were significant differences in S Ⅰ t between the two groups (both P<0.05).The S Ⅰ t of the experimental group was higher than that of the control group.After one day of intervention,there was no significant difference in BUT between the two groups before and after intervention(P>0.05),there was no significant difference in BUT between the two groups (P>0.05).After 7 days and 14 days of intervention,the BUT of the control group decreased significantly compared with that before the intervention,and the differences were statistically significant (both P<0.05),and there were significant differences in BUT between the two groups (both P<0.05).The BUT of the experimental group was longer than the control group.The corneal changes of the experimental group were not obvious after 1-14 days of intervention.The FL staining area was enhanced after 1-14 days of intervention in the control group.On the 4th and 7th day after intervention,the differences of FL scores between the two groups were statistically significant (both P<0.05),and the FL scores of control group were higher than the experimental group.In the experimental group,dense microvilli and folds were observed on the surface of corneal epithelial cells,while in the control group,the surface of microvilli and folds were flat or fused or absent.Conclusion H2 eye drops can improve dry eye by maintaining the integrity of the epithelium.
