16 Hz、130 dB次声对小鼠记忆功能和脑超微结构的影响及姜黄素的治疗作用研究

摘    要:目的观察16Hz、130dB次声对小鼠记忆功能和脑超微结构的影响及不同剂量姜黄素的治疗作用。方法Morris水迷宫成绩相近的BALB/C小鼠接受16Hz、130dB的次声作用,2h/d,同时给予不同剂量的姜黄素,14d后再次评定水迷宫成绩,并取大脑额叶皮层做透射电镜,观察小鼠脑超微结构的变化。结果单纯次声暴露后,小鼠记忆功能下降(和空白组相比,P<O.05),神经细胞缺血性改变,胞浆内脂褐素增加,髓鞘变性,毛细血管水肿;姜黄素各组记忆功能改善(与单纯次声组比较,P<O.05),神经细胞轻度或没有缺血性改变,脂褐素少或没有,髓鞘变性及毛细血管水肿无明显改善。结论16Hz、130dB次声可引发小鼠脑皮质的脂质过氧化,使记忆功能受损。姜黄素可明显减轻次声引发的这些损害,机制可能与其抗氧化作用有关。其治疗作用有一定的部位选择性,主要集中在神经细胞内。

Abstract:Objective To explore the influence on memory function and cerebral ultrastructure of mice of exposure to 16Hz,130dB infrasound and the therapeutic effects of various doses of curcumin. Methods The BALB/C mice with equal learning capacity determined by Morris maze test were exposed to 16Hz,130dB infrasound for 2 hours daily for 14 days,and various doses of curcumin were given by intragastric administration after each time of the exposure.The learning capacity and cerebral ultrastructure of all the mice were observed and compared among groups. Results The memory function of the mice in the infrasound exposure groups decreased. Anoxic changes and lipofuscin accumulation were observed in cytoplast of the neurons. Myelin degeneration and capillary vessels edema were also observed. Compared with the pure infrasound exposure group,the memory function of curcumin-treated infrasound group mice improved significantly,the neurons showed little or no anoxic changes and only slight lipofuscin accumulation. But myelin degeneration and capillary vessels edema were not significantly ameliorated. Conclusion 16Hz,130dB infrasound may induce lipid peroxidation and decrease the memory function of mice,while curcumin can lighten these damages, the mechanism possibly lies in its effects of anti-peroxidation.
