摘    要:以固定化乙酰胆碱酯酶作敏感物质、银基汞膜电极作基础电极,制备了安培型生物传感器。采用单扫描伏安法考察了传感器对有机磷农药敌敌畏的响应特性。以乙酰硫代胆碱作底物,在0.05mol/LpH=7.0的硼砂缓冲溶液中,底物水解产物硫代胆碱在-0.33V处有灵敏的还原峰。在1.0×10-8~1.0×10-6mol/L浓度范围内对敌敌畏进行了测定,校准曲线呈现良好的线性关系,r=0.9988。抑制率为5%时,对敌敌畏的检出限为2.8×10-10mol/L。

Abstract:A acetylcholinesterase(AChE) amperomerometric biosensor is prepared for determination of organophosphorus insecticide, such as dichlorvos. The response characteristic of the AChE biosensor is studied by linear sweep voltammetry. With acetylthiocholine as substrate, in 0.05 mol/L borax buffer solution(pH=7.0), a sensitive voltammetric reduction peak of thiocholine formed by acetylthiocholine hydrolysis is observed. The peak potential is -0.33 V(vs.SCE). The calibration graphs are linear from 1.0×10~(-8) to 1.0×10~(-6) mol/L for dichlorvos. The detection limit (5% of inhibition) for dichlorvos is found to be 2.8×10~(-10) mol/L.
