| 摘 要： | 目的利用猪肺制备血管紧张素转换酶 (ACE)。方法将新鲜猪肺匀浆 ,6 0 0 0×g离心 30min后 ,经 1 .6～ 2 .6mol/L (NH4 ) 2 SO4 分级沉淀 ,SephadexG 2 0 0 ,QHyperD离子交换色谱 ,得到经SDS PAGE显示一条带的ACE纯品。结果猪肺 30 0g可制备出ACE 1 1 .2 5mg,活力回收率为 2 0 .70 % ,比活力为 1 4 80 .0u/mg,较组织匀浆上清提纯提高 42 8.99倍。结论此法结果较理想 ,可用于较大规模制备ACE |

| Abstract: | PurposeTo purify angiotension converting enzyme(ACE)from hog lung tissue. MethodsAfter hog lung homogenate processed with 1.6～2.6 mol/L ammonium sulfate fraction and so on,then by using of gel filtration and anion exchange chromatography column.Results11.25 mg protein containing ACE was obtained from 300 g hog lung tissue.The purified ACE specific activity reached to 1 480.0 units/mg, which was 428.99 times of the crude extract. SDS PAGE test showed a single bond and gave an apparent M r about 180 kD.ConclusionThe method to be able to obtain a high yield of purified ACE from hog lung laid a foundation for preparation of ACE on a large scale. |
