灯盏乙素对缺氧缺血性新生大鼠脑缺氧诱导因子-1α和水通道蛋白9表达水平的影响

摘    要:目的:研究灯盏乙素对缺氧缺血性脑损伤(HIBD)新生大鼠脑缺氧诱导因子-1α(HIF-1α)和水通道蛋白9(APQ9)表达水平的影响。方法:建立HIBD新生大鼠模型,将新生7日龄SD大鼠随机分为假手术组(sham组)、缺氧缺血组(HIBD组)、灯盏乙素高、中和低剂量组、地塞米松阳性对照组。于缺氧缺血7 d后,观察大鼠体质量增长率,脑组织HE染色观察脑组织病理改变,测定脑含水量,免疫荧光法检测脑HIF-1α蛋白表达,逆转录-多聚酶链反应(RT-PCR)法检测脑HIF-1α、APQ9mRNA表达,ELISA法检测脑caspase-3活性。结果:灯盏乙素能显著升高HIBD大鼠体质量增长率,降低脑组织含水量,抑制脑组织HIF-1α和AQP9mRNA表达,降低脑caspase-3活性。结论:灯盏乙素可减轻HIBD大鼠脑组织水肿,降低HIF-1α和AQP9表达,抑制神经细胞凋亡,发挥脑保护作用。

Abstract:OBJECTIVE To explore the effect of scutellarin on the expression of hypoxia-induced factor-1α (HIF-1α) and aquaporin 9 (AQP9) in neonatal rats brain after hypoxia-ischemia brain damage. METHODS Models of 7-day-old neonatal rats with HIBD were established. The rats were randomly divided into six groups:the sham group, hypoxic-ischemic brain damage (HIBD) group, scutellarin treated groups of three different doses, and dexamethasone group. After HIBD for 7 days, the brain water content and weight growth rate were detected by physical methods. The brain tissue morphological changes were observed by HE staining. The HIF-1α and AQP9 mRNA were measured by RT-PCR. The expression of HIF-1α was observed by immunofluorescence staining, and brain caspase-3 activity was measured by ELISA. RESULTS The brain water content in scutellarin treated groups decreased. Weight growth rate in scutellarin treated groups increased. HIF-1α and AQP9 mRNA expressions were inhibited by scutellarin. Caspase-3 activity in scutellarin treated groups decreased. CONCLUSION Scutellarin protects the brain tissues with HIBD probably through the relief of cerebral edema, down regulation of the expression of HIF-1α and AQP9, and inhibition of cell apoptosis.
