体内外脓毒症损伤对Leptin表达水平的影响

摘    要:目的:通过观察体内脓毒症时肺组织及脂多糖(或内毒素,LPS)诱导的体外炎症模型中leptin水平的变化,探讨leptin在脓毒症损伤情况下的作用及意义。方法:建立小鼠的脓毒症损伤模型和LPS诱导的ECV-304细胞炎症损伤模型,用高灵敏的leptin放射免疫分析检测肺组织及细胞上清中leptin水平。结果:脓毒症6h肺组织内leptin显著低于假手术组,而脓毒症12h恢复于假手术组水平之上;leptin保护组6h肺组织内lepin明显高于单纯的脓毒症组,而消炎痛保护组却没有此变化。LPS刺激使ECV-304细胞分泌leptin明显下降(P〈0.05),且成时间依赖性。结论:lenin可能作为一种炎性因子在脓毒症损伤中发挥一定的保护作用。

Abstract:Objective To explore the role of leptin under septic conditions through in vivo and in vitro experimental models.Methods Sixty mice model of sepsis were prepared with cecal puncture.Twenty models were given intraperitoneal leptin or indomethacin(10 each) after preparation of the model and sacrificed 6h later.Another twenty models were given intraperitoreal drug for two times(oh and 6h) and were sacrificed at 12h.Twenty models were given no drug and sacrificed at 6h or 12h(10 at each time).Twenty animals underwent sham operation and served as controls.Leptin levels in the lung tissue homogenate of the sacrificed animals were measured with RIA.In vitro ECV-304 cell line cultures were treated with different strength of lipopoly-sacchride(LPS) and leptin levels in the supernatant were detected.Results At 6h,leptin contents in the lung tissue of models receiving,no drug were significantly lower than those in controls and the levels recovered to normal at 12h.Leptin levels in animals given intraperitoneal leptin were not decreased at 6h,but levels in animals receiving indomethacin were also much decreased at 6h.At 12h,leptin contents reocvered in all animals.With in vitro experiment,the leptin contents in the ECV-304 cell line culture supernatant treated with LPS were significantly lower than those in cultures not treated with LPS(P<0.05).Conclusion As an inflammatory cytokine,leptin might play some protective role in sepsis.
