In an attempt to identify genes that are involved in Drosophila embryonic cardiac development, we have cloned and characterized a gene whose function is required late in embryogenesis to control heart rate and muscular activity. This gene has been named held out wings (how) because hypomorphic mutant alleles produce adult animals that have lost their ability to fly and that keep their wings horizontal at a 90 degree angle from the body axis. In contrast to the late phenotype observed in null mutants, the How protein is expressed early in the invaginating mesoderm and this expression is apparently under the control of twist. When the different mesodermal lineages segregate, the expression of How becomes restricted to the myogenic lineage, including the cardioblasts and probably all the myoblasts. Antibodies directed against the protein demonstrate that How is localized to the nucleus. how encodes a protein containing one KH-domain which has been implicated in binding RNA. how is highly related to the mouse quaking gene which plays a role at least in myelination and that could serve to link a signal transduction pathway to the control of mRNA metabolism. The properties of the how gene described herein suggest that this gene participates in the control of expression of as yet unidentified target mRNAs coding for proteins essential to cardiac and muscular activity.  相似文献

The bottromycins are a family of highly modified peptide natural products, which display potent antimicrobial activity against Gram‐positive bacteria, including methicillin‐resistant Staphylococcus aureus. Bottromycins have recently been shown to be ribosomally synthesized and post‐translationally modified peptides (RiPPs). Unique amongst RiPPs, the precursor peptide BotA contains a C‐terminal “follower” sequence, rather than the canonical N‐terminal “leader” sequence. We report herein the structural and biochemical characterization of BotP, a leucyl‐aminopeptidase‐like enzyme from the bottromycin pathway. We demonstrate that BotP is responsible for the removal of the N‐terminal methionine from the precursor peptide. Determining the crystal structures of both apo BotP and BotP in complex with Mn2+ allowed us to model a BotP/substrate complex and to rationalize substrate recognition. Our data represent the first step towards targeted compound modification to unlock the full antibiotic potential of bottro‐ mycin.  相似文献

Multiphase TRansformation Induced Plasticity (TRIP) steels combine excellent ductility and high strength, making them ideally suited for shock absorbing parts in the automotive industry. When designing structures for impact, an understanding of the mechanical properties of materials under high strain rate conditions is essential. An extensive experimental program using a split Hopkinson tensile bar set-up was established in an effort to investigate the dynamic properties of various TRIP steel grades. Four different TRIP steels are described with varying contents of the alloying elements silicon, aluminium and phosphor. Moreover, several phenomenological models describing the strain rate and temperature-dependent mechanical behaviour are validated. TRIP steel grades in which aluminium is the main alloying element show high elongation values, whereas a high silicon content results in an increase in strength. The widely used Johnson-Cook model can describe the behaviour of TRIP steels and provides the opportunity to study its material and structural response.  相似文献

The beta2 integrin CR4 is involved in Mycobacterium tuberculosis phagocytosis by human mononuclear phagocytes through the opsonin C3bi. In this study, we demonstrate that M. tuberculosis can bind directly to monocyte-derived macrophages via CR4 in the absence of any opsonins. CR4-transfected CHO cells gave similar results, suggesting recognition by CR4 of bacterial structure. Furthermore, binding of M. tuberculosis transduced a potent signal, resulting in tyrosine phosphorylation of macrophage proteins, which was in part mediated by CR4.  相似文献

Genetic groups, also called unknown or phantom parents groups, are often used in dairy cattle genetic evaluations to account for selection that cannot be accounted for by known genetic relationships. With the advent of genomic evaluations, the theory of genetic groups was extended to the so-called single-step genomic BLUP (ssGBLUP). In short, genetic groups can be fitted in ssGBLUP through regression effects, or by including them in the pedigree and computing the adequate combined pedigree and genomic relationship matrix. In this study, we applied the so-called Quaas and Pollak transformation to a system of equations for single-step SNP BLUP (ssSNPBLUP), such that genetic groups can thereafter be included in the pedigree. The example in this study showed that including the genetic groups in the pedigree for ssSNPBLUP allowed reduced memory burden and computational costs in comparison to genetic groups fitted as covariates.  相似文献

Abalone meat is a delicacy worldwide, fetching high prices and a valuable source of income for the many countries farming and exporting this commodity. The quality of abalone is based on its unique sensory properties and an analytical metabolomics method for determining the compounds related to this would serve as a valuable tool for ensuring quality and consumer satisfaction. Metabolomics is a promising “omics” tool which can be applied towards this goal; however, widely applicable parameters for the evaluation of an untargeted gas chromatography mass spectrometry (GC-MS) metabolomic approach is still lacking. GC-MS is a popular and suitable metabolomics method due to its high separation power, reproducible retention times, and selective mass detection. The aim of this study was to establish a reliable untargeted GC-MS method for analyzing firstly a standard compound mixture consisting of 10 compounds representing various compound classes and secondly applying the method in an untargeted manner to abalone muscle samples. Using a standard compound mixture with a concentration range of 1 to 100 μg/mL, the limit of detection (LOD) ranged between 0.01 and 3.30 μg/mL, the limit of quantification (LOQ) resulted in values between 0.02 and 9.49 μg/mL, the accuracy determined was <1.5 μg/mL, and the precision displayed a coefficient of variance (CV) <25 %. When evaluating the method in terms of biological samples harvested, the repeatability and intermediate precision showed CV values <50 % for most compounds measured, allowing application of this method for metabolite profiling of abalone to answer important biological questions.  相似文献

We describe in this work an approach that individually combines constant initialization, free-space backpropagation, transport of intensity equation (TIE) and multi-height phase recovery with Fast Iterative Shrinkage-Thresholding Algorithm (FISTA) to predict reconstruction of the phase microspheres from diffracted field measurements. Life time test in direct borohydride fuel cell system   总被引:1,自引:0,他引:1  
The electric performances of direct borohydride fuel cells (DBFCs) are evaluated in terms of power density and life time with respect to the NaBH4 concentration. A DBFC constituted of an anionic membrane, a 0.6 mgPt cm−2 anode and a commercial non-platinum based cathode led to performances as high as 200 mW cm−2 at room temperature and with natural convection of air. Electrochemical life time test at 0.55 mA cm−2 with a 5 M NaBH4/1 M NaOH solution shows a voltage diminution of 1 mV h−1 and a drastic drop of performances after 250 h. The life time is twice longer with 2 M NaBH4/1 M NaOH solution (450 h) and the voltage decrease is 0.5 mV h−1. Analyses of the components after life time tests indicate that voltage loss is mainly due to the degradation of the cathode performance. Crystallisation of carbonate and borate is observed at the cathode side, although the anionic membrane displays low permeability to borohydride.  相似文献

Carbon allotropes exhibit an enormous range of properties due to their varied hybridizations. Different hybridizations are correlated with different geometrical structures. We address the visualization of both the geometry and the electronic density for different cases including diamonds with defects and mixed diamond/graphite/amorphous samples, in order to explore this connection more deeply.  相似文献
