The aim of this study was the investigation of antimicrobial and antioxidant activities of different ethanolic extracts of propolis (EEP) collected from north‐western Algeria. Total polyphenols and flavonoids contents of extracts were measured. The UV absorption spectrum showed and confirmed their polyphenols constituents. All EEPs exerted antibacterial activity against Gram‐positive bacteria, but no effect on Gram‐negative bacteria. The minimum inhibitory concentration (MIC) of EEPs ranged from 0.01% to 2.6% v/v. The antioxidant activity was measured using ferric‐reducing power (FRAP), 1,1‐diphenyl‐2‐picrylhydrazyl (DPPH) free radical‐scavenging and ESR methods. Propolis TIA‐2 and MOH‐MAS samples showed the highest antioxidative capacity, after 35 min, while TIA‐1, NED‐TL and SFS‐SBA samples showed the highest antioxidative potential of measured EEPs, after 3 min. TIA‐2 sample showed the highest antibacterial, antioxidant activity and highest DPPH free radical‐scavenging activity as well as the highest polyphenols and flavonoids content, compared with other propolis samples.  相似文献

Dendritic cells (DCs), the most potent antigen-presenting cells inducing specific immune responses, are involved in the pathogenesis of atherosclerosis. In this inflammatory disease, DCs increase in number, being particularly abundant in the shoulder regions of plaques. Since the exposure to altered gravitational conditions results in a significant impairment of the immune function, the aim of this study was to investigate the effects of hypergravity on both the function of DCs and their interactions with the vascular wall cells. Monocytes from peripheral blood mononuclear cells of healthy volunteers were sorted by CD14+ magnetic beads selection, cultured for 6 days in medium supplemented with GM-CSF and IL-4, followed by a further maturation stimulus. DC phenotype, assessed by flow cytometry, showed a high expression of the specific DC markers CD80, CD86, HLA-DR and CD83. The DCs obtained were then exposed to hypergravitational stimuli and their phenotype, cytoskeleton, ability to activate lymphocytes and interaction with vascular wall cells were investigated. The findings showed that the exposure to hypergravity conditions resulted in a significant impairment of DC cytoskeletal organization, without affecting the expression of DC markers. Moreover, an increase in DC adhesion to human vascular smooth muscle cells and in their ability to activate lymphocytes was observed.  相似文献

Three different biochemical test systems were chosen based on their solubility to study the antioxidant activity of ginger extracts. Reducing power and DPPH. scavenging activity tests were considered to produce hydrophilic environments and the H2O2 test was considered as creating a lipophilic environment. The average yields were 10.23 ± 1.02% and 0.48 ± 0.19% for oleoresin and essential oil, respectively. The content of total phenols was 67.6 ± 1.08 mg GAE/g of dry extract. In terms of EC50, in hydrophilic environment standards, it showed the highest effects compared to ginger extracts, with oleoresin presenting more activity than essential oil. In contrast, except for quercetin, essential oil showed the best scavenging activity in inhibiting H2O2 compared to all other antioxidants. The present work demonstrated that, when using reducing power, DPPH· free radical scavenging and H2O2 scavenging assays, the same ginger extracts exhibit different antioxidant activities, which were affected not only by the extract itself but also by the chemical environment (hydrophilic/lipophilic).  相似文献

This study determined the ontogenic changes in learning and hippocampal protein kinase C (PKC) in C57 and DBA mice. Mice were tested on the visible- or hidden-platform versions of the Morris water task starting at 17, 24, 31, or 60 days of age. Both strains learned to locate the visible platform at all ages. C57 mice learned to solve the hidden-platform task when they were 24 days old, whereas DBA mice never learned to solve this task. Using a [–3H]-phorbol ester binding assay, the authors found that both strains had similar amounts of hippocampal PKC at 10 and 17 days of age but that C57 mice had significantly more PKC at 24, 31, and 60 days of age. Immunoblotting results revealed that C57 mice had more γ-PKC, but not α-PKC, than DBA mice. Thus, the development of performance differences in spatial learning between C57 and DBA mice parallels the ontogeny of hippocampal PKC. The exposure to microgravity conditions results in a significant impairment of the immune function. Many reports describe morphological and functional changes in T-lymphocytes, monocytes and neutrophil granulocytes cultured in microgravity, both real and modeled, but very few studies have been made on the effect of microgravity on dendritic cells (DCs) and DC differentiation. DCs are able to process antigens and are the most efficient cells in presenting them to T-lymphocytes, thus giving a crucial contribution to the rise of an effective immune response. The aim of this study was to investigate whether the maturation of DCs from monocytes of astronauts was altered postflight. Blood samples from a crew-member of the Eneide mission were collected before the flight, soon after the return to earth and one year after the mission. In order to generate DCs, monocytes were used as precursors. They were separated, cultured for 6 days in medium supplemented with granulocyte macrophage colony stimulating factor (GM-CSF) and interleukin 4 (IL-4), then furtherly stimulated for 24 hours with a cocktail of cytokines. Differentiation was assessed by flow cytometry and immunofluorescence, assaying the expression of typical DC markers. Gene expression was analysed by RT-PCR. Morphological and functional characteristics were studied by autofluorescence microscopy. The findings showed that the maturation of DCs from monocytes collected from the astronaut immediately postflight was altered. In comparison with controls, significant differences were found in expression of DC markers, expression of genes involved in DC maturation, morphological and functional characteristics.  相似文献

The beneficial health effects including antioxidant properties of mint (Mentha spicata) and honey have been extensively studied. However, there is no data about the effect of their associated use. In this study the total phenolic and flavonoid contents of individual extracts of mint and honey and their combination were determined. The antioxidant activity was investigated by using reducing power, 1,1-diphenyl-2-picrylhydrazyl (DPPH), 2,2´-azinobis-(3-ethylbenzothiazoline-6-sulfonic acid diammonium salt (ABTS), and chelating power methods. The results showed that individual extracts contained important quantity of phenolics and flavonoids and their combination was found to produce best antioxidant activity. A positive correlation between the phenolic/flavonoid contents and antioxidant activity, especially with free radical scavenging ability and chelating power, was observed. These findings suggest that combined mixture of honey and mint could be used as promising additive foods.  相似文献
