Abdominal aortic aneurysm (AAA) is an inflammatory disease associated with marked changes in the cellular composition of the aortic wall. This study aims to identify microRNA (miRNA) expression in aneurysmal inflammatory cells isolated by laser microdissection from human tissue samples. The distribution of inflammatory cells (neutrophils, B and T lymphocytes, mast cells) was evaluated in human AAA biopsies. We observed in half of the samples that adventitial tertiary lymphoid organs (ATLOs) with a thickness from 0.5 to 2 mm were located exclusively in the adventitia. Out of the 850 miRNA that were screened by microarray in isolated ATLOs (n = 2), 164 miRNAs were detected in ATLOs. The three miRNAs (miR-15a-3p, miR-30a-5p and miR-489-3p) with the highest expression levels were chosen and their expression quantified by RT-PCR in isolated ATLOs (n = 4), M1 (n = 2) and M2 macrophages (n = 2) and entire aneurysmal biopsies (n = 3). Except for the miR-30a-5p, a similar modulation was found in ATLOs and the two subtypes of macrophages. The modulated miRNAs were then evaluated in the plasma of AAA patients for their potential as AAA biomarkers. Our data emphasize the potential of miR-15a-3p and miR-30a-5p as biomarkers of AAA but also as triggers of ATLO evolution. Further investigations will be required to evaluate their targets in order to better understand AAA pathophysiology. 相似文献

The removal of lead from drinking water was investigated to develop a point-of-use water filter that could meet the regulation imposed by the new European Directive 98-83 lowering lead concentration in drinking water below 10 μg L−1. The objective of this research was to assess the potential of different adsorbents (zeolites, resins, activated carbon, manganese oxides, cellulose powder) to remove lead from tap water with a very short contact time. To begin, the repartition of the lead species in a tap water and a mineral water was computed with the computer model CHESS. It showed that in bicarbonated waters lead is mainly under lead carbonate form, either in the aqueous or in the mineral phase. Batch experiments were then conducted to measure the equilibrium adsorption isotherms of the adsorbents. Then, for five of them, dynamic experiments in micro-columns were carried out to assess the outlet lead concentration level. Three adsorbents gave rise to a leakage concentration lower than 10 μg L−1 and were then selected for prototypes experiments: chabasite, an activated carbon coated with a synthetic zeolite and a natural manganese oxide. This article evaluates the effect of shear stress on the composition of biofilm bacterial communities. For the first time, a Conical Couette-Taylor Reactor (CCTR) was used to develop biofilms at varying shear stresses (from 0.055 to 0.27 Pa) and provided a useful model for studying the effect of hydrodynamics on biofilms. The composition, diversity and dynamics of biofilm bacterial communities were analysed using the PCR-SSCP fingerprint method. Results clearly demonstrate a link between shear stress and composition of the microbial communities. High shear stresses decrease biofilm diversity and the analysis of biofilm community dynamics suggests that shear stress would slow down biofilm maturation and tend to maintain a young biofilm. 相似文献

Penile plethysmography (PPG) is the gold standard for the assessment of sexual interests, especially among sex offenders of children. Nonetheless, this method faces some ethical limitations inherent to the nature of its stimuli and could benefit from the improvement of its ecological validity. The use of computer-generated characters (CGC) in virtual immersion for PPG assessment might help address these issues. A new application developed to design made-to-measure anatomically correct virtual characters compatible with the Tanner developmental stages is presented. The main purpose of this study was to determine how the virtual reality (VR) modality compares to the standard auditory modality on their capacity to generate sexual arousal profiles and deviance differentials indicative of sexual interests. The erectile responses of 22 sex offenders of children and 42 non-deviant adult males were recorded. While both stimulus modalities generated significantly different genital arousal profiles for sex offenders of children and non-deviant males, deviance differentials calculated from the VR modality allowed for significantly higher classification accuracy. Performing receiver operating characteristic analyses further assessed discriminant potential. Auditory modality yielded an area under the curve (AUC) of 0.79 (SE = 0.059) while CGC in VR yielded an AUC of 0.90 (SE = 0.052). Overall, results suggest that the VR modality allows significantly better group classification accuracy and discriminant validity than audio stimuli, which provide empirical support for the use of this new method for PPG assessment. Additionally, the potential use of VR in interventions pertaining to self-regulation of sexual offending is addressed in conclusion. 相似文献

The Nightingallery project encouraged participants to converse, sing, and perform with a musically responsive animatronic bird, playfully interacting with the character while members of the public could look on and observe. We used Nightingallery to frame an HCI investigation into how people would engage with one another when confronted with unfamiliar technologies in conspicuously public, social spaces. Structuring performances as improvisational street theatre, we styled our method of exhibiting the bird character. We cast ourselves in supporting roles as carnival barkers and minders of the bird, presenting him as if he were a fantastical creature in a fairground sideshow display, allowing him the agency to shape and maintain dialogues with participants, and positioning him as the focal character upon which the encounter was centred. We explored how the anthropomorphic nature of the bird itself, along with the cultural connotations associated with the carnival/sideshow tradition helped signpost and entice participants through the trajectory of their encounters with the exhibit. Situating ourselves as secondary characters within the narrative defining the performance/use context, our methods of mediation, observation, and evaluation were integrated into the performance frame. In this paper, we explore recent HCI theories in mixed reality performance to reflect upon how genre-based cultural connotations can be used to frame trajectories of experience, and how manipulation of roles and agency in participatory performance can facilitate HCI investigation of social encounters with playful technologies. 相似文献
