Growing interest in extracellular vesicles (EVs) has prompted the advancements of protocols for improved EV characterization. As a high-throughput, multi-parameter, and single particle technique, flow cytometry is widely used for EV characterization. The comparison of data on EV concentration, however, is hindered by the lack of standardization between different protocols and instruments. Here, we quantified EV counts of platelet-derived EVs, using two flow cytometers (Gallios and CytoFLEX LX) and nanoparticle tracking analysis (NTA). Phosphatidylserine-exposing EVs were identified by labelling with lactadherin (LA). Calibration with silica-based fluorescent beads showed detection limits of 300 nm and 150 nm for Gallios and CytoFLEX LX, respectively. Accordingly, CytoFLEX LX yielded 40-fold higher EV counts and 13-fold higher counts of LA+CD41+ EVs compared to Gallios. NTA in fluorescence mode (F-NTA) demonstrated that only 9.5% of all vesicles detected in scatter mode exposed phosphatidylserine, resulting in good agreement of LA+ EVs for CytoFLEX LX and F-NTA. Since certain functional characteristics, such as the exposure of pro-coagulant phosphatidylserine, are not equally displayed across the entire EV size range, our study highlights the necessity of indicating the size range of EVs detected with a given approach along with the EV concentration to support the comparability between different studies.  相似文献

The scope of this research is a problem of parameters identification of a linear time-invariant plant, which (1) input signal is not frequency-rich, (2) is subjected to initial conditions and external disturbances. The memory regressor extension (MRE) scheme, in which a specially derived differential equation is used as a filter, is applied to solve the above-stated problem. Such a filter allows us to obtain a bounded regressor value, for which a condition of the initial excitation (IE) is met. Using the MRE scheme, the recursive least-squares method with the forgetting factor is used to derive an adaptation law. The following properties have been proved for the proposed approach. If the IE condition is met, then: (1) the parameter error of identification is bounded and converges to zero exponentially (if there are no external disturbances) or to a set (in the case of them) with an adjustable rate, (2) the parameters adaptation rate is a finite value. The above-mentioned properties are mathematically proved and demonstrated via simulation experiments.  相似文献

A novel process concept for the oxidative coupling of methane followed by the oligomerization to liquids has been developed within the frame of the EU integrated project OCMOL. This technology is based on process intensification principles via cutting‐edge structured microreactor technology. It is also a fully integrated industrial process through the re‐use and the recycling of by‐products, in particular CO2, at every process stage. The focus of this contribution is on the reaction engineering aspects of the core steps, i.e., catalysts, kinetics and reactor design for the methane coupling and reforming.  相似文献

In this article, we present an adaptive color similarity function defined in a modified hue‐saturation‐intensity color space, which can be used directly as a metric to obtain pixel‐wise segmentation of color images among other applications. The color information of every pixel is integrated as a unit by an adaptive similarity function thus avoiding color information scattering. As a direct application we present an efficient interactive, supervised color segmentation method with linear complexity respect to the number of pixels of the input image. The process has three steps: (1) Manual selection of few pixels in a sample of the color to be segmented. (2) Automatic generation of the so called color similarity image (CSI), which is a gray level image with all the gray level tonalities associated with the selected color. (3) Automatic threshold of the CSI to obtain the final segmentation. The proposed technique is direct, simple and computationally inexpensive. The evaluation of the efficiency of the color segmentation method is presented showing good performance in all cases of study. A comparative study is made between the behavior of the proposed method and two comparable segmentation techniques in color images using (1) the Euclidean metric of the a* and b* color channels rejecting L* and (2) a probabilistic approach on a* and b* in the CIE L*a*b* color space. Our testing system can be used either to explore the behavior of a similarity function (or metric) in different color spaces or to explore different metrics (or similarity functions) in the same color space. It was obtained from the results that the color parameters a* and b* are not independent of the luminance parameter L* as one might initially assume in the CIE L*a*b* color space. We show that our solution improves the quality of the proposed color segmentation technique and its quick result is significant with respect to other solutions found in the literature. The detection phase in computational contact mechanics can be subdivided into a global search and a local detection. When potential contact is detected by the former, a rigorous local detection determines which surface elements come or may come in contact in the current increment. We first introduce a rigorous definition of the closest point for non‐differentiable lower‐dimensional manifolds. We then simplify the detection by formulating an optimization problem subject to inequality constraints. The formulation is then solved using different techniques from the field of mathematical optimization, for both linear and quadratic finite element meshes. The resulting general and robust detection scheme is tested on a set of problems and compared with other techniques commonly used in computational geometry. The convective fluid flows with an interface are modeled using the classical Oberbeck–Boussinesq model of convection. The three dimensional solutions for the infinite domains with fixed heat-insulated boundaries and with the interface under action of a longitudinal temperature gradient are studied. Construction of the solutions for the flows of two immiscible fluids in a channel with a rectangular cross-section is carried out using a complete problem statement. The kinematic and dynamic conditions are prescribed at the interface. The additional condition of continuity of the tangential velocities, the conditions of continuity of temperature and of the thermal fluxes are assumed to be fulfilled on the interface. In the present paper the fluid flows are studied in the stationary case under conditions of gravity and microgravity. To investigate this problem numerically an iteration algorithm is introduced. This algorithm is based on a finite difference scheme (the alternating direction method) and it allows to find all the components of velocity for both phases and temperature distributions. The examples of flows which can be characterized as a combination of the translational and progressively rotational types of motion are presented.  相似文献

Transition metals incorporated into polymers lead to unusual or improved physical properties that significantly differ from those of purely organic polymers. A simple and practicable incorporation of diverse transition metals into any available polymer would make an important contribution to overcome some of the synthetic difficulties of metal‐polymer hybrid materials. Here, it is demonstrated that atomic layer deposition (ALD) can be a promising means to resolve some of those difficulties. It is found that even polytetrafluoroethylene (PTFE) with its great physical and chemical stability can be easily transformed into a transition metal–PTFE hybrid material simply by applying a metal‐oxide ALD process to PTFE. Upon metal incorporation into the PTFE, the molecular structure as well as mechanical properties (tensile behavior) of PTFE were observed to significantly change. For a better understanding of the changes to the material, experimental investigations using Raman spectroscopy, attenuated‐total‐reflection Fourier‐transform infrared spectroscopy, wide‐angle X‐ray diffraction, and energy‐dispersive X‐ray analysis were performed. In addition, with density functional theory calculations, potential bonding states of the incorporated metal into PTFE were modeled and predicted. The ALD‐based vapor‐phase approach for metal incorporation into a polymer could bring about rapid progress in the research area of metal–polymer hybrid materials.  相似文献

A 6-gap glass resistive plate chamber with a strip readout was tested on the beam of the U-70 accelerator at the Institute for High Energy Physics. A time resolution of ∼45 ps was attained at an efficiency of >98%. The position resolution along a strip was estimated to be ∼1 cm.  相似文献

An objective of experiments and finite element simulations was to check the stiffness, the strength and the fatigue resistance of the attachment of the First Wall panels onto a shield block of blanket modules according to the ITER 2001 design. The panel has a poloidal key at the rear side (in so-called option A with the rear access bolting) and it is attached by means of special studs located on a key-way in the shield block. Special device for a test of stud tensile pre-load relaxation during a thermal cycling was developed. True-to-scale panels, the shield block mock-up and simplified studs were fabricated and the assembly was loaded alternatively by radial moment, poloidal force or poloidal moment simulating the loading during off-normal plasma operations. Thermal cycling led to an acceptable stud pre-load relaxation. Mechanical cycling caused neither the pre-load relaxation nor the loss of the contact in the key-way nor a damage of the attachment system. The combination of poloidal moment and radial force during vertical displacement events (VDEs) seems to be a most dangerous case because it could lead to the loss of the key–key-way contact.  相似文献

Sequence‐independent or “click”‐type chemistry is applied for the preparation of novel π‐conjugated oligomers. A variety of bi‐functional monomers for Wittig–Horner olefination are developed and applied in a sequential protection–deprotection process for the preparation of structurally similar π‐conjugated oligomers. Selected oligomers are incorporated as the organic semiconductors in light‐emitting diodes and a field‐effect transistor, demonstrating the potential of the approach.  相似文献
