Linear alkylbenzene sulfonate (LAS) and polyoxyethylene lauryl ether (POLE) are the major surfactants in washing detergents. In the present study, we isolated surfactant-resistant bacteria from soil samples collected from a sports ground and a farm field. The samples were treated with 2.0% LAS or POLE at 25°C for 30 min and cultivated on agar plates at 25°C for several days, after which manifold bacterial colonies were isolated. Thereafter, we tested the ability of each bacterial isolate to resist the antibacterial activity of the surfactant. Ten LAS-resistant strains were isolated, and all were found to be Gram-negative bacteria such as Enterobacter and Pseudomonas. On the other hand, 18 POLE-resistant strains were isolated, of which 14 were Gram-positive bacteria including Bacillus and Microbacterium. Notably, one POLE-resistant strain was identified as Bacillus cereus, a potential causative agent for foodborne illness. The genera of LAS- and POLE-resistant bacteria did not overlap. Therefore, the combination of LAS and POLE could be more effective to eliminate soil bacteria from clothes and/or daily necessities.  相似文献

Submerged membrane bioreactors (MBRs) are now widely used for various types of wastewater treatment. One drawback of submerged MBRs is the difficulty in removing nitrogen because intensive aeration is usually carried out in the tank and the MBRs must therefore be operated under aerobic conditions. In this study, the feasibility of treating municipal wastewater by a baffled membrane bioreactor (BMBR), particularly in terms of nitrogen removal, was examined. Simultaneous nitrification/denitrification in a single and small reaction tank was possible by inserting baffles into a normal submerged MBR as long as wastewater was fed in the appropriate way. To examine the applicability of the BMBR, pilot-scale experiments were carried out using real municipal wastewater. Although neither external carbon addition nor mixed liquor circulation was carried out in the operation of the BMBR, average removal rates of total organic carbon (TOC), total phosphorus (T-P) and total nitrogen (T-N) reached 85%, 97% and 77%, respectively, with the hydraulic retention time (HRT) of 4.7h. Permeability of the membrane could be maintained at a high level throughout the operation. It was found that denitrification was the limiting step in removal of nitrogen in the BMBR in this study. Various types of monitoring carried out in the BMBR also demonstrated the possibility of further improvements in its performance.  相似文献

The liquid crystal display (LCD) technology allows several simple circuits to be built using thin film transistors, thus making fabrication of compact, integrated, large-area, and low-cost micro-pattern gaseous detectors possible. In this work, a single-grid-type micro-strip gas chamber (S-MSGC) using transparent electrodes based on the LCD technology was fabricated and successfully operated in several gas mixtures. The detector was coupled with a multi-pixel photon counter to detect an optical signal through the transparent substrate in Ar/CF4 gas. Both electrical and optical signals were measured and the light yield of the detector was acquired. Successful operation of the S-MSGC can be considered the very important first step for development of the next target of integrated devices.  相似文献

Some reports demonstrated successful genome editing in pigs by one-step zygote microinjection of mRNA of CRISPR/Cas9-related components. Given the relatively long gestation periods and the high cost of housing, the establishment of a single blastocyst-based assay for rapid optimization of the above system is required. As a proof-of-concept, we attempted to disrupt a gene (GGTA1) encoding the α-1,3-galactosyltransferase that synthesizes the α-Gal epitope using parthenogenetically activated porcine oocytes. The lack of α-Gal epitope expression can be monitored by staining with fluorescently labeled isolectin BS-I-B4 (IB4), which binds specifically to the α-Gal epitope. When oocytes were injected with guide RNA specific to GGTA1 together with enhanced green fluorescent protein (EGFP) and human Cas9 mRNAs, 65% (24/37) of the developing blastocysts exhibited green fluorescence, although almost all (96%, 23/24) showed a mosaic fluorescent pattern. Staining with IB4 revealed that the green fluorescent area often had a reduced binding activity to IB4. Of the 16 samples tested, six (five fluorescent and one non-fluorescent blastocysts) had indel mutations, suggesting a correlation between EGFP expression and mutation induction. Furthermore, it is suggested that zygote microinjection of mRNAs might lead to the production of piglets with cells harboring various mutation types.  相似文献

The reaction of N-(2,4-dinitrophenyl)pyridinium anion ( salt(A) ; A = Cl, FeCl4, and (CN)2N) with linear polyethyleneimine (LPEI; Mn = 20 380) and branched polyethyleneimines (BPEI1; Mn = 600, BPEI2; Mn = 10 000) at various molar feed ratios without using a catalyst resulted in pyridinium ring opening to yield ionic LPEI and BPEIs that were crosslinked by conjugated penta-2,4-dienylideneammonium (PDA) units, LPEI-PDA , BPEI1-PDA , and BPEI2-PDA , respectively. A model compound was synthesized by the reaction of salt(Cl) with diethylamine. The solubilities of BPEI1-PDA and BPEI2-PDA depended on the feed ratios between salt(Cl) and BPEI1 or BPEI2. Dipping LPEI-PDA into water and methanol yielded hydro- and organogels, respectively. UV–vis and reflection measurements revealed an expanded π-conjugation length between the polymer chains due to the through-space orbital interaction of the electrons on the two nitrogen atoms at the crosslinked positions in LPEI-PDA , BPEI1-PDA , and BPEI2-PDA . Cyclic voltammetry analysis suggested that the polymers underwent electrochemical oxidation. Measurement using a superconducting quantum interference device (SQUID) indicated that LPEI-PDA having FeCl4 anions was paramagnetic. © 2019 Wiley Periodicals, Inc. J. Appl. Polym. The online time-differential perturbed angular correlation (TDPAC) method was applied to a study of the physical states of a probe 19F, the β decay product of 19O (t1/2 = 26.9 s), implanted in highly oriented pyrolytic graphite. The observed magnitude of the electric field gradient at the probe nucleus, ∣Vzz∣ = 2.91(17) × 1022 V m−2, suggests that the incident 19O atoms are stabilized at an interlayer position with point group C3v. Exhibiting observed TDPAC spectra having a clear sample-to-detector configuration dependence, we demonstrate the applicability of the present online method with a short-lived radioactive 19O beam.  相似文献

A metal/insulator/semiconductor (MIS) structure of Ga0.47In0.53As has been prepared by applying an excimer laser photo-CVD process to depositing an SiNx insulating layer. It is found that interface state densities can be remarkably reduced by in situ photochemical etching with CC14 or CH3Br gases prior to SiNx deposition. A minimum value of the U-shape profile of the interface state density as low as 5 × 1011 cm-2eV-1is attained.  相似文献
