Targeting tumor vasculature through specific endothelial cell markers represents a promising approach for cancer treatment. Here our aim was to construct an antibiotic resistance gene-free plasmid encoding shRNAs to simultaneously target two endothelial cell markers, CD105 and CD146, and to test its functionality and therapeutic potential in vitro when delivered by gene electrotransfer (GET) and combined with irradiation (IR). Functionality of the plasmid was evaluated by determining the silencing of the targeted genes using qRT-PCR. Antiproliferative and antiangiogenic effects were determined by the cytotoxicity assay tube formation assay and wound healing assay in murine endothelial cells 2H-11. The functionality of the plasmid construct was also evaluated in malignant melanoma tumor cell line B16F10. Additionally, potential activation of immune response was measured by induction of DNA sensor STING and proinflammatory cytokines by qRT-PCR in endothelial cells 2H-11. We demonstrated that the plasmid construction was successful and can efficiently silence the expression of the two targeted genes. As a consequence of silencing, reduced migration rate and angiogenic potential was confirmed in 2H-11 endothelial cells. Furthermore, induction of DNA sensor STING and proinflammatory cytokines were determined, which could add to the therapeutic effectiveness when used in vivo. To conclude, we successfully constructed a novel plasmid DNA with two shRNAs, which holds a great promise for further in vivo testing.  相似文献

Immersion tests with different stainless steels have been performed, while the pH was stepwise decreased and then increased again. During 8.5-day exposure, the depassivation and repassivation pH values as a function of pitting resistance equivalent number were determined. There is always a gap between both pH values (depassivation and repassivation), indicating that for every steel, there are conditions where an existing passive layer can be maintained but cannot be rebuilt after depassivation. In such environments, the passive layer is thicker, consisting mainly of molybdenum and iron rich oxides, while chromium is dissolved. Usually, depending on conditions, the passive layer is more chromium-rich, especially the inner layer. This is relevant, for example, for acidizing jobs in oil and gas industry, proving that repassivation after acidizing will happen promptly, when the pH is increased again.  相似文献

This article investigates building thermal mass control of commercial buildings to reduce utility costs with a particular emphasis on the individual impacts of both adaptive comfort criteria and of heat waves. Recent changes in international standards on thermal comfort for indoor environments allow for adaptation to the weather development as manifested in comfort criteria prEN 15251.2005 and NPR-CR 1752.2005 relative to the non-adaptive comfort criterion ISO 7730.2003. Furthermore, since extreme weather patterns tend to occur more frequently, even in moderate climate zones, it is of interest how a building's passive thermal storage inventory responds to prolonged heat waves. The individual and compounded effects of adaptive comfort criteria and heat waves on the conventional and optimal operation of a prototypical office building are investigated for the particularly hot month of August 2003 in Freiburg, Germany. It is found that operating commercial buildings using adaptive comfort criteria strongly reduces total cooling loads and associated building systems energy consumption under conventional and building thermal mass control. In the case of conventional control, total operating cost reductions follow the cooling loads reductions closely. Conversely, the use of adaptive comfort criteria under optimal building thermal mass control leads to both lower and slightly higher absolute operating costs compared to the optimal costs for the non-adaptive ISO 7730. While heat waves strongly affect the peak cooling loads under both conventional and optimal building thermal mass control, total cooling loads, building energy consumption and costs are only weakly affected for both control modes. Passive cooling under cost-optimal control, while achieving significant total cost reductions of up to 13%, is associated with total energy penalties on the order of 1–3% relative to conventional nighttime setup control. Thus, building thermal mass control defends its cost saving potential under optimal control in the presence of adaptive comfort criteria and heat waves.  相似文献

The changing forms and concentrations of arsenic through aluminium-based coagulation treatment processes were tracked for three drinking-water treatment plants. This has provided direct evidence of where and how arsenic is removed. In general, soluble As(V) is converted to particulate As(V) by adsorption during rapid mixing, and is removed along with naturally occurring particulate arsenic predominantly by clarification. Soluble As(III) tracks through the treatment processes and is converted to soluble As(V) during final chlorination. The ability of a water treatment process to achieve the maximum acceptable concentration for arsenic in drinking water is dependent on the concentration of As(III) in the source water.  相似文献

This paper evaluates the use of upflow anaerobic sludge bed (UASB) bioreactors (30 degrees C, pH=7.0) to remove selenium oxyanions from contaminated waters (790 microg Se L(-1)) under methanogenic and sulfate-reducing conditions using lactate as electron donor. One UASB reactor received sulfate at different sulfate to selenate ratios, while another UASB was operated under methanogenic conditions for 132 days without sulfate in the influent. The selenate effluent concentrations in the sulfate-reducing and methanogenic reactor were 24 and 8 microg Se L(-1), corresponding to removal efficiencies of 97% and 99%, respectively. X-ray diffraction (XRD) analysis and sequential extractions showed that selenium was mainly retained as elemental selenium in the biomass. However, the total dissolved selenium effluent concentrations amounted to 73 and 80 microg Se L(-1), respectively, suggesting that selenate was partly converted to another selenium compound, most likely colloidally dispersed Se(0) nanoparticles. Possible intermediates of selenium reduction (selenite, dimethylselenide, dimethyldiselenide, H(2)Se) could not be detected. Sulfate reducers removed selenate at molar excess of sulfate to selenate (up to a factor of 2600) and elevated dissolved sulfide concentrations (up to 168 mg L(-1)), but selenium removal efficiencies were limited by the applied sulfate-loading rate. In the methanogenic bioreactor, selenate and dissolved selenium removal were independent of the sulfate load, but inhibited by sulfide (101 mg L(-1)). The selenium removal efficiency of the methanogenic UASB abruptly improved after 58 days of operation, suggesting that a specialized selenium-converting population developed in the reactor. This paper demonstrates that both sulfate-reducing and methanogenic UASB reactors can be applied to remove selenate from contaminated natural waters and anthropogenic waste streams, e.g. agricultural drainage waters, acid mine drainage and flue gas desulfurization bleeds.  相似文献

Persistent methyl sulfone (MeSO2-) and hydroxylated (HO-) polychlorinated biphenyl (PCB) metabolites have emerged as important classes of environmental contaminants in vertebrate, aquatic biota and humans. In the present study, PCB, MeSO2-PCB and HO-PCB concentrations and congener patterns were determined in the whole blood and adipose tissue of male (n = 7) and female (n = 12) polar bears (Ursus maritimus) of random age (3-25 years of age), and collected in 1999-2001 from the Ittoqqortoormiit/Scoresby Sound area in central East Greenland. There was no significant difference (P < 0.05) between males and females with respect to PCB or PCB metabolite concentrations in either tissue. The mean sum (Sigma) PCB concentrations were 7020+/-3366 ng/g lipid weight (lw) (range 2708-18148 ng/g lw) and 46.1+/-44.6 ng/g wet weight (ww) (range 12.6-204.2 ng/g ww) in adipose and blood, respectively. The mean Sigma-HO-PCB concentration in whole blood was 182.3+/-72.1 ng/g ww (range 93.8-382.1 ng/g ww). The mean Sigma-HO-PCB to Sigma-PCB concentration ratios in whole blood were 4.59+/-3.58 (range 1.03-11.88) and 8.30+/-5.56 (range 2.16-19.47) in females and males, respectively, which are the highest ratios reported so far for polar bears from any population, or for any free-ranging animal. Sigma-HO-PCB concentrations were greater than all other major classes of organochlorines (i.e. Sigma-PCBs, Sigma-MeSO2-PCBs, Sigma-chlordanes (CHLs), Sigma-hexachlorocyclohexanes (HCHs) and Sigma-chlorobenzenes (CBzs). The mean Sigma-MeSO2-PCB concentrations were 699+/-836 ng/g lw (range 127-3920 ng/g lw) and 10.9+/-9.6 ng/g ww (range 4.3-52.1 ng/g ww) in the adipose and blood, respectively. Regardless of age and sex, in both adipose and whole blood the MeSO2-PCB congener pattern was dominated by 3'- and 4'-MeSO2-CB101 and -CB87, and 4-MeSO2-CB149 (approx. 70% of the Sigma-MeSO2-PCBs). Minor differences in the MeSO2-PCB congener pattern were observed between blood and adipose, which suggests the possible influence of metabolite structure on mobilization and/or deposition to the adipose tissue. Sixteen HO-PCB congeners and one di-HO-PCB congener were identified, and five HO-PCB isomers and one di-HO-PCB isomer were detected. However, congener patterns were dominated by 4'-OH-CB120, 4-HO-CB146/3-HO-CB153, 4-OH-CB187, 4'-HO-CB172, 4-HO-CB193 and 4,4'-di-HO-CB202 (> 10 ng/g ww). HO-PCB congener patterns in whole blood were not significantly different (P < 0.05) between males and females. Other chlorinated phenolic contaminants, pentachlorophenol (0.3+/-0.3 ng/g ww) and 4-HO-heptachlorostyrene (7.5+/-2.9 ng/g ww) were also detected in blood. To our knowledge, this is to first report comparing PCBs, MeSO2-PCBs and HO-PCBs in whole blood and adipose tissue in a free-ranging wildlife species. HO-PCBs and MeSO2-PCBs are both important circulating contaminants in polar bears from this eastern Greenland population. Given the known toxicities of PCB metabolites, this population of polar bear may be experiencing health risks due to exposure to a complex loading of organohalogen contaminants that include HO-PCB and MeSO2-PCB metabolites.  相似文献
