The objective of this study was to examine the influence of drug amount and mixing time on the homogeneity and content uniformity of a low-dose drug formulation during the dry mixing step using a new gentle-wing high-shear mixer. Moreover, the study investigated the influence of drug incorporation mode on the content uniformity of tablets manufactured by different methods. Albuterol sulfate was selected as a model drug and was blended with the other excipients at two different levels, 1% w/w and 5% w/w at impeller speed of 300?rpm and chopper speed of 3000?rpm for 30?min. Utilizing a 1?ml unit side-sampling thief probe, triplicate samples were taken from nine different positions in the mixer bowl at selected time points. Two methods were used for manufacturing of tablets, direct compression and wet granulation. The produced tablets were sampled at the beginning, middle, and end of the compression cycle. Rate constants of reactions of ozone with non-ionized solutes, such as aliphatic alcohols, olefins, chlorosubstituted ethylenes, substituted benzenes and carbohydrates, have been determined from the absolute rates with which ozone reacts in the presence of various concentrations of these compounds in water. They have been tested by comparison with the relative rates by which pairs of these solutes are transformed by ozone. Different experimental methods have been developed to determine such rate constants in the range from 10−2 to 105 M−1 s−1. Interferences between the direct reactions of ozone and reactions due to its preliminary decomposition to secondary oxidants could be eliminated. The kinetics of all the reactions studied are first order with respect to ozone and solute concentration. The rate constants of many types of organic compounds in water are of the same order of magnitude as in organic solvents. Substituted benzenes, however, react in water about 100 times faster. They obey a linear free energy relationship with p = −3.1 when based on δp+ values. Comparisons of rate constants with chemical structures of the reacting groups show that all reactions of ozone are highly selective and electrophilic. The kinetic data allow explanation of the chemical effects of ozone observed in water treatment practice.  相似文献

We propose an autoregressive conditional duration (ACD) model with periodic time-varying parameters and multiplicative error form. We name this model periodic autoregressive conditional duration (PACD). First, we study the stability properties and the moment structures of it. Second, we estimate the model parameters, using (profile and two-stage) Gamma quasi-maximum likelihood estimates (QMLEs), the asymptotic properties of which are examined under general regularity conditions. Our estimation method encompasses the exponential QMLE, as a particular case. The proposed methodology is illustrated with simulated data and two empirical applications on forecasting Bitcoin trading volume and realized volatility. We found that the PACD produces better in-sample and out-of-sample forecasts than the standard ACD.  相似文献

The concept of the Global Petroleumspace is an analytical tool which engages the roles which different oil actors play in the development of new urban ideas and built forms. Coined by Hein, this concept contributes to enriching our understanding of globalization, modernity, and architectural history and their impacts on space through time. Petroleum is modern industry’s fuel par excellence. For much of the world, it is the arrival of petroleum on the local scene that introduces modernity with its attendant spaces, forms, materials, and discourses. To frame this new discourse on urban development and petromodernity, three events were organized: ‘Petroleumscape Roundtable’ held as part of the 17th IPHS Conference at TU Delft, ‘Petroleum Modernism Symposium’, organized at the Illinois Institute of Technology, and ‘The Global Petroleumscape Conference’ held at the Faculty of Architecture at TU Delft. This article briefly recounts and reflects upon the scholarly discussions which took place at these events, in order to outline an emergent discourse on petroleum’s imbrication in architecture and planning.  相似文献

Global and regional land cover studies need to apply complex models on selected subsets of large volumes of multi-sensor and multi-temporal data sets that have been derived from raw instrument measurements using widely accepted pre-processing algorithms. The computational and storage requirements of most of these studies far exceed what is possible on a single workstation environment. We have been pursuing a new approach that couples scalable and open distributed heterogeneous hardware with the development of high performance software for processing, indexing and organizing remotely sensed data. Hierarchical data management tools are used to ingest raw data, create metadata and organize the archived data so as to automatically achieve computational load balancing among the available nodes and minimize input/output overheads. We illustrate our approach with four specific examples. The first is the development of the first fast operational scheme for the atmospheric correction of Landsat Thematic Mapper scenes, while the second example focuses on image segmentation using a novel hierarchical connected components algorithm. Retrieval of the global Bidirectional Reflectance Distribution Function in the red and near-infrared wavelengths using four years (1983 to 1986) of Pathfinder Advanced Very High Resolution Radiometer (AVHRR) Land data is the focus of our third example. The fourth example is the development of a hierarchical data organization scheme that allows on-demand processing and retrieval of regional and global AVHRR data sets. Our results show that substantial reductions in computational times can be achieved by the high performance computing technology.  相似文献

Architecture's daily impact on its users is the result of neither concentration nor focused attention. Preoccupied with everyday tasks, most people do not stop to observe the architectural object as a work of art. In this paper I investigate the content that may be present when architecture is experienced in the state Walter Benjamin calls 'distraction'. Using a phenomenological model of attention and my applied research, I propose a plausible model for the Lived Experience of the Built Environment (LEBEN). I further consider the possible components of the everyday experience of architecture and the urban surroundings, and suggest an integrative structure for it. Part I explains the need for the research and presents its objectives. Part II discusses the phenomenological model of attention and awareness. Parts III and IV present the LEBEN model, briefly describing its sources in the literature, and applied research, based on a workshop that examined multi-subjective perspectives. I note the research assumptions, methodology and the challenges facing such an investigation, eg, whether it is possible to study a phenomenon that exists beyond the realm of focal attention. Part V introduces the core-themes and categories distilled from my research (edge, depth, change, atmosphere and affordances) and positions them in the LEBEN model.  相似文献

It must be assumed that current petri dish primary hepatocyte culture models do not supply sufficient amounts of oxygen and thus cause anaerobic metabolism of the cells. This is contrary to the physiologic state of the cells. In vivo the liver is a highly vascularized organ with a rather high blood flow rate of a mixture of arterial and venous blood. The aim of the present study was to show the oxygen dependence of primary rat hepatocytes in long-term culture and to define appropriate conditions that could allow hepatocytes to maintain tissue specific functions in an aerobic environment. To this purpose matrix overlaid hepatocytes were either cultured on gas-permeable (fluorinated hydrocarbon films) or gas-impermeable (polystyrene) supports at 10% and 20% ambient oxygen concentration (v/v), respectively. Tissue-specific functions were assessed by studying albumin and urea secretion as well as xenobiotic metabolism. The mRNA expression and catalytic activities of the cytoprotective antioxidant enzymes mitochondrial manganese superoxide dismutase (MnSOD), cytosolic copper and zinc superoxide dismutase, peroxisomal catalase, and cytosolic glutathione peroxidase were investigated to assess intracellular responses to the defined variations in oxygen supply. Hepatocytes could successfully be maintained at aerobic conditions in long-term culture on gas-permeable PTFE films. At 50% (10%, v/v) of currently used oxygen levels lactate accumulation was prevented, a plateau-like albumin secretion reestablished, urea secretion improved, and xenobiotic metabolism proceeded at physiological rates. mRNA expression of cytoprotective enzymes responded to the pericellular availability of oxygen and was most pronounced in the case of MnSOD. However, the biggest stress factor for the hepatocytes still appeared to be the isolation procedure, as mRNA expression and catalytic activities were most elevated shortly thereafter. In conclusion, this study clearly shows the oxygen dependence of primary rat hepatocytes in long-term culture and indicates means to establish appropriate conditions for the aerobic culture of primary rat sandwich hepatocytes with full maintenance of function. The long-term culture of hepatocytes on oxygenating supports at in vivo-like oxygen tensions therefore appears to be more physiologic and beneficial for the cells.  相似文献
