The organization center of Cynops pyrrhogaster was divided into Parts 1, 2 and 3 of equal size (0.3×0.4 mm2) with presumptive fates as pharyngeal, pharyngeal+prechordal+trunk notochord, and trunk-tail notochord, respectively. Movements and changes in size and shape of each part were followed through gastrulation. Differentiation tendencies of each part were examined under three conditions: I, isolated; II, sandwiched with presumptive ectoderm; 111, sandwiched with presumptive ectoderm after preculture in isolation for various times. In I, Parts 2 and 3 differentiated into dorsal mesoderm. In II, each part induced dorsal mesoderm and neural tissues, the frequency being highest in Part 2 and lowest in Part 3. In III, Parts 1 and 2 realized their presumptive fates, through changes in inductive capacities from trunk-tail to head. This change progressed rapidly in Part 1, and slowly in Part 2. Part 3 required induction by neighbouring Part 2 to realize its presumptive fate. Changes of inductive capacity of Parts 1 and 2 respectively, were chronologically similar in normal development and in preculture experiments. Lastly, the primary presumptive pharyngeal zone at blastula was proposed to act as an initiator of the organization center, its programmed information being transmitted to Part 2, and then to Part 3.  相似文献

Mesodermal differentiation of dorsal marginal zone (DMZ) before and after invagination was analyzed by a series of combination experiments with different kinds of ectoderm.
Lower DMZ of early gastrula didn't show any axial-mesoderm (notochord and somitic mesoderm) but lateral mesoderm (mesenchyme, mesothelium, or blood cells) in combinant with non-competent ventral ectoderm, while combinant with competent ectoderm was found to have well-differentiated axial-mesoderm with deutero-spinocaudal neurals. The axial-mesoderms have origin in the ectoderm. Uninvaginated DMZ of middle gastrula also showed difference in mesodermal differentiation between competent and non-competent ectoderms; axial-mesoderm differentiation was much better in competent than in non-competent. The axial-mesoderm originated from the uninvaginated DMZ. Archenteron roof of late gastrula showed regional difference in mesodermal differentiation in both combinants with competent and non-competent. The present study further demonstrated that there was regionality in promoting effect of induced neurectoderm on axial-mesoderm differentiation of invaginated archenteron roof.
The present experiments suggest that the cranio-caudal and dorso-ventral axis formations of amphibian mesoderm are finally determined by sequential and reciprocal interactions between the mesodermal anlage and the overlying ectoderm. It should be also shown that lower DMZ acts to trigger a series of the sequential interactions during primary embryonic induction.  相似文献

The concentration of ions in plant cells and tissues is an essential factor in determining physiological function. In the present study, we established that concentration gradients of mobile ions exist in both xylem exudates and tissues within a barley (Hordeum vulgare) primary leaf. For K+ and NO3?, ion concentrations generally decreased from the leaf base to the tip in both xylem exudates and tissues. Ion gradients were also found for Pi and Cl? in the xylem. The hydathode strongly absorbed Pi and re‐translocated it to the rest of the plant, whereas Cl? was extruded. The ion concentration gradients developed early during leaf growth, increased as the tissue aged and remained under both high and low transpiration conditions. Measurement of the expression profiles of Pi, K+ and NO3? transporters along the longitudinal axis of the leaf revealed that some transporters are more expressed at the hydathode, but for most transporters, there was no significant variation along the leaf. The mechanisms by which longitudinal ion gradients develop in leaves and their physiological functions are discussed.  相似文献

Characterizing the carbon turnover in terrestrial ecosystems is critical for understanding and predicting carbon dynamics in ecosystems. We used in situ13C pulse labeling to track photosynthetic carbon fluxes from shoot to roots and to soil in a Kobresia humilis meadow on the Qinghai‐Tibet Plateau. We found that about 36.7% of labeled carbon was translocated out from the shoots within the first 24 h after photosynthetic uptake. This is equivalent to 66.1% of total 13C moving out from the shoot during the 32‐day chase period, indicating a rapid and large translocation of newly fixed carbon to belowground parts in these alpine plants. 58.7% of the assimilated 13C was transferred belowground. At the end of the chase phase, 30.9% was retained in living roots, 3.4% in dead roots, 17.2% lost as belowground respiration and 7.3% remained in the soil. In the four carbon pools (i.e., shoots, living roots, dead roots, and soil pools), living roots consistently had the highest proportion of 13C in the plant–soil system during the 32 days. Based on the 13C partitioning pattern and biomass production, we estimate a total of 4930 kg C ha?1 was allocated belowground during the vegetation growth season in this alpine meadow. Of this, roots accumulated 2868 kg C ha?1 and soils accumulated 613 kg C ha?1. This study suggests that carbon storage in belowground carbon pools plays the most important role in carbon cycles in the alpine meadow.  相似文献

Mesoderm formation in the presumptive trunk organizer was analyzed in gastrulae of Cynops pyrrhogaster. The presumptive trunk organizer showed little or no mesodermal differentiation in the beginning gastrula (0 h embryo). But as soon as the presumptive trunk organizer came into contact with the newly invaginated cranial archenteron roof, it rapidly formed mesoderm. This suggests that this differentiation was brought about by an inductive effect of the underlying cranial archenteron roof. For investigation of this possibility, the presumptive trunk organizer of 0 h embryos (Tr-0) and the newly invaginated cranial archenteron roof (presumptive pharyngeal endoderm and prechordal plate) of successive stages were cultured in isolation and by the sandwich technique. The newly invaginated presumptive pharyngeal endoderm and prechordal plate had no effect on mesoderm formation of the presumptive trunk organizer, and mesodermal differentiation of the combinations was similar to that of the Tr-0 alone. On the other hand, results showed that the prechordal plate, which came into contact with the still uninvaginated presumptive trunk organizer, stimulated dorsalisation of the weakly mesodermized trunk organizer. Based on these results, the stepwise process of mesoderm formation in the trunk organizer is discussed.  相似文献

In order to clone and analyse the avirulence gene AVR-Pia from Japanese field isolates of Magnaporthe oryzae , a mutant of the M. oryzae strain Ina168 was isolated. This mutant, which was named Ina168m95-1, gained virulence towards the rice cultivar Aichi-asahi, which contains the resistance gene Pia. A DNA fragment (named PM01) that was deleted in the mutant and that co-segregated with avirulence towards Aichi-asahi was isolated. Three cosmid clones that included the regions that flanked PM01 were isolated from a genomic DNA library. One of these clones (46F3) complemented the mutant phenotype, which indicated clearly that this clone contained the avirulence gene AVR-Pia . Clone 46F3 contained insertions of transposable elements. The 46F3 insert was divided into fragments I–VI, and these were cloned individually into a hygromycin-resistant vector for the transformation of the mutant Ina168m95-1. An inoculation assay of the transformants revealed that fragment V (3.5 kb) contained AVR-Pia . By deletion analysis of fragment V, AVR-Pia was localized to an 1199-bp DNA fragment, which included a 255-bp open reading frame with weak homology to a bacterial cytochrome- c -like protein. Restriction fragment length polymorphism analysis of this region revealed that this DNA sequence co-segregated with the AVR-Pia locus in a genetic map that was constructed using Chinese isolates.  相似文献

Phorbol esters as probes of the modulatory site on protein kinase C–an overview. The phorbol esters, diterpene derivatives produced by members of the family Euphorbiaceae, are of major scientific importance as tumour promoters. Their primary site of action is protein kinase C, an enzyme involved in the message transduction pathway for a large class of hormones which cause enhanced phosphatidylinositol turnover. Heterogeneity in the response of biological systems to the phorbol esters may be explained in part by heterogeneity in the phospholipids associated with protein kinase C, in its subcellular localization, and by proteolytic processing, as well as by isoforms of the enzyme. Development of other classes of activators of protein kinase C through computer modelling and natural product isolation suggests possible new approaches for intervention in the pathway.  相似文献

Photosynthesis and transpiration of excised leaves of Taraxacum officinale L. and a few other species of plants were measured, using an open gas analysis system. The rates of CO2 uptake and transpiration increased in two steps upon illumination of stomata-bearing epidermis of these leaves at a light intensity of 50 mW × cm−2. Abscisic acid inhibited only the second step of gas exchange. Illumination of the astomatous epidermis of hypostomatous leaves caused only the first step of gas exchange. These data indicate that the first and second steps arise from cuticular and stomatal gas exchange, respectively. The rate of the cuticular photosynthesis in a Taraxacum leaf reached saturation at a light intensity of 5 mW × cm−2, and the rates of the stomatal photosynthesis and transpiration reached saturation at a higher intensity of 35 mW × cm−2. The cuticular photosynthesis of a Taraxacum leaf was 18% of the stomatal photosynthesis at 50 mW × cm−2 and 270% at 5 mW × cm−2. The other species of leaves showed the same trend. The importance of cuticular CO2 uptake in leaf photosynthesis, especially under low light intensity was stressed from these data.  相似文献

Isotachophoretic analysis of ions was performed on guard cells of Vicia faba cv. Ryosai Issun with either open or closed stomata. In guard cells of open stomata, K+ and malic acid concentrations were 5–7 and 5–10 times higher, respectively, than in guard cells of closed stomata. The content of citric acid (plus isocitric acid) also increased during stomatal opening, but the increment was smaller than that of malic acid. Sodium ions, phosphoric and glyceric acids were present in low concentrations but did not increase during the opening. Other cations and anions could not be measured because of low concentrations. Malic acid provided 68–79% of the counter anions for the potassium taken up by guard cells during stomatal opening.  相似文献
