In order to identify proguanylin-secreting cells, we have raised an antiserum against the synthetic fragment of human proguanylin (1-15) and have examined the proguanylin-positive cells in the human and rat gastrointestinal tract by immunohistochemical methods. Numerous proguanylin (1-15)-immunoreactive cells were found in the gastrointestinal tract. They were either pyramidal or spindle shaped in the stomach. Spindle-shaped cells, frequently possessing long slender processes, were located at the base of the pyloric epithelium and did not extend to the lumen. In the duodenum and jejunum, these cells were mostly pyramidal in shape and often had a slender process towards the lumen. The immunostaining was completely blocked by the human proguanylin (1-15) fragment. Paneth and goblet cells were negative against this antiserum. The number of serotonin-positive cells was much larger than that of proguanylin-positive cells in all the segments tested. The number of proguanylin-positive cells decreased from the jejunum to the ileum and very few cells were observed in the colon. In contrast to serotonin-positive cells, most somatostatin-positive cells were also positive for proguanylin. Thus, proguanylin (1-15) or its related protein appears to coexist with somatostatin in intestinal endocrine D cells which may be a source of circulating proguanylin. Proguanylin, like somatostatin, may also regulate intestinal function as a local regulator.  相似文献

A thrombus in a coronary artery causes ischemia, which eventually leads to myocardial infarction (MI) if not removed. However, removal generates reactive oxygen species (ROS), which causes ischemia–reperfusion (I/R) injury that damages the tissue and exacerbates the resulting MI. The mechanism of I/R injury is currently extensively understood. However, supplementation of exogenous antioxidants is ineffective against oxidative stress (OS). Enhancing the ability of endogenous antioxidants may be a more effective way to treat OS, and exosomes may play a role as targeted carriers. Exosomes are nanosized vesicles wrapped in biofilms which contain various complex RNAs and proteins. They are important intermediate carriers of intercellular communication and material exchange. In recent years, diagnosis and treatment with exosomes in cardiovascular diseases have gained considerable attention. Herein, we review the new findings of exosomes in the regulation of OS in coronary heart disease, discuss the possibility of exosomes as carriers for the targeted regulation of endogenous ROS generation, and compare the advantages of exosome therapy with those of stem-cell therapy. Finally, we explore several miRNAs found in exosomes against OS.  相似文献

Inhomogeneities in semiconductor solids can be imaged by two-dimensional mapping of the amplitude of periodically modulated tip current in scanning tunneling microscopy that is induced by illumination of semiconductor samples with a chopped light. It has been shown that it is possible to distinguish between plural origins of the photo-modulated current by analyzing the response properties of the current signal. A judicial choice of the modulation frequency is important for the required contrasts to be obtained.  相似文献

We theoretically analyze the relationship between the electric field envelope shape of an optical pulse launched into an optical fiber and the power spectrum of the spontaneous Brillouin backscattered light it produces. The electric field envelope is characterized by the pulse width, leading-trailing time, and steepness. The peak power of the launched, pulsed-light power spectrum is proportional to the square of the pulse width regardless of the pulse leading-trailing time and steepness, and the power spectrum broadens in inverse proportion to the pulse width. The peak power of the spontaneous Brillouin backscattered light produced by the launched, pulsed light is proportional to the pulse width when it is above approximately 100 ns and is proportional to the square of the pulse width when it is below approximately 1 ns. The power spectrum of the spontaneous Brillouin backscattered light also broadens rapidly corresponding to the pulse width, when the pulse width falls below approximately 30 ns. As the pulse leading-trailing time is shortened or the pulse leading-trailing part becomes steep, the Brillouin backscattered-light power spectrum broadens greatly, even if the launched pulse width remains constant. Our analysis showed that an optical pulse with a triangular-shaped electric field envelope forms the Brillouin backscattered-light power spectrum with the narrowest profile and consequently gives the minimum error in measuring the peak-power frequency, when the pulse width is below approximately 50 ns. The measurement error with the triangular-shaped pulsed light is 1/ radical2 times smaller than that for a rectangular-shaped pulsed light, when the pulse width falls below several nanoseconds. By contrast, the rectangular-shaped envelope gives the minimum error when the pulse width exceeds ~50 ns.  相似文献

A spherical aberration (Cs)-corrected 200 kV TEM was newly developed. The column of the microscope was extended by 25 cm and the inner yoke of the objective lens was modified to insert some parts of the corrector elements. The corrector has two hexapole elements that play a main role in Cs correction and they are placed at a position equivalent to the coma-free point of the objective lens by using two transfer doublet lenses. The Cs correction was successfully carried out by means of the third-order aberration that was generated in the two extended hexapoles. The Cs can be corrected to the desired value and also can be overcompensated in order to produce a negative Cs, as with the corrected Cs of -23 microm shown in this work. The optical system of the corrector does not produce second- and fourth-order aberrations, and can correct residual aberrations up to the third order. All of the corrector elements are computer-controlled and the third-order aberrations are quite stable after they are properly corrected. The resolution of 0.135 nm was experimentally confirmed by the Young's fringe method. Image simulations of a silicon [110] single crystal were made with various Cs and defocus values to demonstrate the effectiveness of arbitral control of Cs.  相似文献

固体燃料和固体废物在燃烧和气化过程中的汞排放问题在全球已经受到广泛关注。该文首先选取3个煤种和3种干城市污泥作为样本,通过热重(TG)实验了解汞在燃烧和气化过程中的基本析出行为。然后选取一个污泥样在滴管炉里燃烧,研究汞在接近实际燃烧状况下的析出行为。通过热平衡计算预测了在和实验相同条件下汞析出的变化趋势。结果表明,污泥燃烧过程中汞的析出行为受到污泥中硫和固定碳含量的抑制,煤燃烧过程中汞的析出行为受到煤中固定碳含量和Cl含量的重要影响。另外,因为单质汞在还原性气氛下极易形成,在气化过程中,即使是低温汞也很快析出。  相似文献

Schwann cells play an important role in peripheral nerve function, and their dysfunction has been implicated in the pathogenesis of diabetic neuropathy and other demyelinating diseases. The physiological functions of insulin in Schwann cells remain unclear and therefore define the aim of this study. By using immortalized adult Fischer rat Schwann cells (IFRS1), we investigated the mechanism of the stimulating effects of insulin on the cell proliferation and expression of myelin proteins (myelin protein zero (MPZ) and myelin basic protein (MBP). The application of insulin to IFRS1 cells increased the proliferative activity and induced phosphorylation of Akt and ERK, but not P38-MAPK. The proliferative potential of insulin-stimulated IFRS1 was significantly suppressed by the addition of LY294002, a PI3 kinase inhibitor. The insulin-stimulated increase in MPZ expression was significantly suppressed by the addition of PD98059, a MEK inhibitor. Furthermore, insulin-increased MBP expression was significantly suppressed by the addition of LY294002. These findings suggest that both PI3-K/Akt and ERK/MEK pathways are involved in insulin-induced cell growth and upregulation of MPZ and MBP in IFRS1 Schwann cells.  相似文献

Optical near-field interactions allow energy localization at scales smaller than the diffraction limit of light. They also show intrinsic hierarchical responses, meaning that optical near-fields exhibit different physical behavior at different scales. In this paper, by combining the localized energy dissipation and hierarchy properties, we present an architecture for a novel traceable optical memory that records memory access events to each bit, which is useful in applications such as high-security information transfer. The basic principles are demonstrated by numerical simulations using a metal nanostructure.  相似文献

The influence of miscibility of an acrylic PSA and several tackifier resin systems upon PSA performance was investigated. When the acrylic copolymer and the resins were blended in various proportions, three types of mixing state were found: miscible system, partially miscible system and immiscible system. In the case of miscible systems, PSA performance (tack, peel strength and shear resistance) depended upon the viscoelastic properties of the PSA. In the case of completely immiscible systems, the above PSA performance depended primarily upon the viscoelastic properties of a continuous matrix phase, and the separated resin phase acted as a kind of filler. In the case of partially miscible systems, the PSA performance changed discontinuously at the resin concentration where phase separation occurred. It suggests that the phase structure of a PSA greatly influences the PSA's performance.  相似文献
