Kinetic,thermodynamic parameters and in vitro digestion of tannase from Aspergillus tamarii URM 7115

Tannase is an enzyme used in various industries and produced by a large number of microorganisms. The aim of this study was to evaluate tannase production to determine the biochemical, kinetic, and thermodynamic properties and to simulate tannase in vitro digestion. The tannase-producing fungal strain was isolated from “jamun” leaves and identified as Aspergillus tamarii. Temperature at 26°C for 67?h was the best combination for maximum tannase activity (6.35-fold; initial activity in Plackett–Burman design—15.53?U/mL and average final activity in Doehlert design—98.68?U/mL). The crude extract of tannase was optimally active at 40°C, pH 5.5 and 6.5. Moreover, tannase was stimulated by Na+, Ca2+, Mg2+, and Mn2+. The half-life at 40°C lasted 247.55?min. The free energy of Gibbs, enthalpy, and entropy, at 40°C, was 81.47, 16.85, and ?0.21?kJ/mol?·?K, respectively. After total digestion, 123.95% of the original activity was retained. Results suggested that tannase from A. tamarii URM 7115 is an enzyme of interest for industrial applications, such as gallic acid production, additive for feed industry, and for beverage manufacturing, due to its catalytic and thermodynamic properties. 相似文献

Conservation of food products depends on product quality and packaging suitability. The objective of this work was to develop and evaluate the antimicrobial efficiency of natamycin‐incorporated film in the production process of Gorgonzola cheese. It aims to optimize the production process and increase shelf‐life and food safety for the consumer. Films with different concentrations of natamycin were produced and tested in Gorgonzola cheeses to evaluate its efficiency against Penicillium roqueforti on the cheese surface. Films with 2 and 4% natamycin presented satisfactory results for fungus inhibition and the amount of natamycin released to the cheese was below that allowed by the legislation. A business process is a set of activities performed in a coordinated manner within an organizational and technical environment that is aimed toward a business goal. The flexibility of a process is related to an understanding of the unexpected events that occur when people, systems and resources interact and require adjustments. Thus, business processes must be designed to respond to information about different events and their specificity. This information defines what the literature calls “context”. To broaden the perception of context in the case of a business process, this work proposes an approach to characterize the context of a business process activity in a given domain through conceptual models structured in layers. The aim of this research was to study spray drying as potential action to protect chlorophyllide from environmental conditions for shelf‐life extension and characterisation of the powders. Six formulations were prepared with 7.5 and 10 g of carrier agents [gum Arabic (GA), maltodextrin (MA) and soybean protein isolate (SPI)]/100 mL of chlorophyllide solutions. The powders were evaluated for morphological characteristics (SEM), particle size, water activity, moisture, density, hygroscopicity, cold water solubility, sorption isotherms, colour and stability, during 90 days. All the powders were highly soluble, with solubility values around 97%. A significant lower hygroscopicity was observed for GA powders, whilst the lower Xm values obtained by GAB equation fitting of the sorption isotherms was observed for the 7.5 g MA/100 mL samples. All formulations, but the 1 (7.5 g SPI/100 mL of chlorophyllide), provided excellent stability to the chlorophyllide during 90 days of storage even at room temperature. 相似文献

Compatibility of the new environmentally-friendly alternative of diesel engine fuels, biodiesel, with storage and engine part materials, is still an open issue. In this work, the interaction between three fuels (petroleum diesel and two types of biodiesel — soybean and sunflower) and two materials (carbon steel and high density polyethylene) used in storage and automotive tanks, is analyzed in detail. A wide set of characterization techniques was used to evaluate the changes in both solid and fluid materials, as weight change measurement, optical, scanning electron and atomic force (AFM) microscopies, Raman and FTIR spectroscopies, and differential scanning calorimetry. The AFM technique allowed detecting surface roughness and morphology changes in the metallic material following the trends in the weight losses. In the case of polymeric material, weight gain by fluid absorption occurred, being detected by the spectroscopic techniques. The biodiesel fuels underwent some ageing however this phenomenon did not affect the interaction between the biodiesel fuels and the substrates. The petrodiesel, which did not age, caused more significant degradation of the substrates. 相似文献

Fructooligosaccharides have been isolated from roots and leaves of Stevia rebaudiana, by hot aqueous extraction, followed by precipitation with ethanol. Their structure has been determined using methylation and NMR analysis, MALDI-TOF, and ESI-MS. Fructooligosaccharides contained almost exclusively (2→1)-linked β-fructofuranosyl, with terminal α-glucopyranosyl and β-fructofuranosyl units. MALDI-TOF and ESI-MS analyses showed the wide range of degree of polymerisation (DP) present in various extracts. From roots and leaves, three different fractions gave profiles of homologous series, with DPs ranging up to 17 with MALDI-TOF and 19 using ESI-MS. These inulin-type fructooligosaccharides were the major component of extracts from S. rebaudiana roots and significant components from the leaves. 相似文献

In this work, we demonstrated a novel and effective approach on the use of low-cost electrodes, an eco-friendly substrate and zinc oxide (ZnO) micro or nanorods (MRs or NRs, respectively) for building triboelectric devices (TENGs). The reported strategy focuses on using low-cost materials and fabrication processes. For the first time and without any pre-treatment, an aluminum recyclable paper from the milk carton (named ARP) was used as a substrate and TENG bottom electrode. A systematic study on the growing of ZnO structures on ARP by chemical bath deposition has been carried out. We found that the ZnO rods size, and resistivity of the TENG upper electrode considerably influence the power density of the device. Such sustainable, low-priced ZnO-based TENGs can produce up to 1.6μW/cm2 output power density when operated at 50?Hz. The fabrication of an eco-friendly nanogenerator demonstrates the possibility of manufacturing low-cost, flexible, and large-area energy harvesting devices for future applications. 相似文献
