Nisin expression production from Lactococcus lactis in milk whey medium

BACKGROUND: Nisin is a commercially available bacteriocin produced by Lactococcus lactis ATCC 11454 and used as a natural agent in the biopreservation of food. In the current investigation, milk whey, a byproduct from dairy industries was used as a fermentation substrate for the production of nisin. Lactococcus lactis ATCC 11454 was developed in a rotary shaker (30 °C/36 h/100 rpm) using two different media with milk whey (i) without filtration, pH 6.8, adjusted with NaOH 2 mol L^?1 and without pH adjustment, both autoclaved at 121 °C for 30 min, and (ii) filtrated (1.20 µm and 0.22 µm membrane filter). These cultures were transferred five times using 5 mL aliquots of broth culture for every new volume of the respective media. RESULTS: The results showed that culture media composed of milk whey without filtration supplied L. lactis its adaptation needs better than filtrated milk whey. Nisin titers, in milk whey without filtration (pH adjusted), was 11120.13 mg L^?1 in the second transfer, and up to 1628‐fold higher than the filtrated milk whey, 6.83 mg.L^?1 obtained in the first^t transfer. CONCLUSIONS: Biological processing of milk byproducts (milk whey) can be considered a profitable alternative, generating high‐value bioproducts and contributing to decreasing river disposals by dairy industries. An engineering process for the verification of real-time systems

The complete verification of the timing properties of a large critical system cannot be undertaken in a single step or with a single method. In this paper we present a process that links together a number of techniques and approaches that cover all stages of development from requirements analysis to code testing. The key elements of the process are: a constrained form of timed automata that uses delay and deadline to define temporal behaviour, notions of rely and guarantee to cover temporal dependencies, model checking for design verification, SPARK and Ravenscar restrictions for programming, and scheduling and response time analysis for asserting implementation compliance. Extended examples of the use of the process are given.

Biosorption of heavy metals by Saccharomyces cerevisiae: Effects of nutrient conditions

Studies took place to investigate the effects of different nutrient conditions on the biosorption ability and selectivity of heavy metals by Saccharomyces cerevisiae. After having grown in media supplemented with additional glucose, ammonium, phosphate or cysteine, the yeast was exposed to an equimolar solution of lead, cadmium, zinc and copper. Lead removal from a mixed solution was significantly higher than that of copper, followed by zinc and cadmium. Generally, yeasts from cysteine-rich media showed greatest sorption capacity whereas phosphate addition influenced zinc selectivity. In addition, glucose, fructose and sucrose as carbon sources were examined. Cultures grown in glucose had a better uptake than those cultivated with fructose at an incubation time of 30 min.     

On the algebraic formulation of certain geometry statements and mechanical geometry theorem proving

In this paper we analyze the algebraic formulations of certain geometry statements appearing in recent literature related to mechanical geometry theorem proving and give several examples to show that one of these formulations can cause serious problems. We clarify a formulation which is essentially due to W. T. Wu and, in our opinion, is the most satisfactory.This author was supported by NSF Grants DCR-8503498 and CCR-8702108.

调节对象参数和运动特性随工况变化的分析

水轮机调节对象的许多参数都是随运行工况变化的。以调节对象线性化模型为基础，从1个实例计算人手，计算了不同工况下水轮机传递系数和发电机6个参数的变化情况，进而计算这些参数变化对捌节对象特征根的影响。通过汁算表明，其水力振荡衰减速度与出力有关．而系统的其他运动特性随工况变化较小。

应用铟源的反应蒸发制备In2O3透明导电膜

本工作说明，在低压氧气氛中。应用铟源的反应蒸发很容易淀积高质量的Ｉｎ２Ｏ３透明导电膜．旦然没有使用锡杂质，但所得膜的性能可以和最好的掺锡的Ｉｎ２Ｏ３膜相比．膜电阻率达２～３×１０￣４Ω·ｃｍ，可见光透过率超过９０％，并且膜生长速率高达２１９／ｍｉｎ．文章对成膜过程作了分析，报道了膜的最佳淀积条件，对由于偏离最佳淀积参数而导致的异常膜的形成机制也进行了讨论．
