智利海洋沉积物中放线菌多样性   总被引:1,自引:0,他引:1  
【目的】认识智利海洋沉积物中放线菌的多样性。【方法】分别采用选择性分离培养和非培养的基于16SrRNA基因序列系统发育分析方法,对来自智利南部海域海洋沉积物中放线菌多样性进行研究。采用6种选择性分离培养基分离放线菌;利用放线菌特异性引物对样品总DNA进行16SrRNA基因序列扩增并构建了16SrRNA基因克隆文库。分别挑选不同培养特征的22株放线菌和59个基因克隆进行16SrRNA基因序列的系统进化分析;测定分离的放线菌对海水的依赖性及产生抗菌活性化合物的能力。【结果】共分离到328株放线菌;挑选的22株放线菌分别属于小单孢菌属、多形孢菌属、链霉菌属、迪茨氏菌属、气微菌属和短状杆菌属;挑取的59个克隆属于40个分类单元,其中60%分类单元属于放线菌门放线菌亚纲、酸微菌亚纲和红色杆菌亚纲,另外40%的分类单元在放线菌内形成几个独立的进化分支,有可能代表放线菌新类群。22株放线菌有19株具有抗菌活性,50%的生长依赖海水的放线菌也具有抗菌活性。【结论】智利海域沉积物存在丰富的放线菌系统发育多样性并能产生活性次级代谢产物,而且还蕴藏丰富的新类型的放线菌资源。  相似文献

【背景】在地衣共生系统中除了共生真菌、藻类以外,还蕴藏着丰富的放线菌资源。【目的】采集来自云南西双版纳、白茫雪山、德国波罗的海南岸3个地区的地衣,对获得的地衣纯培养放线菌进行多样性分析。【方法】采用3种放线菌选择分离培养基,通过平板稀释涂布法分离放线菌。通过比较16S rRNA基因序列相似性以及构建系统发育树,确定纯培养放线菌的分类地位。【结果】共分离菌株1 123株,鉴定417株。其中从西双版纳17份地衣样品中分离纯化到107株放线菌,分布在7个目14个科33个属,潜在新种18株,其中链霉菌为优势菌属;从白茫雪山7份地衣样品中分离纯化到103株放线菌,分布在4个目5个科9个属,潜在新种16株。其中链霉菌为优势菌属,占比39%;从波罗的海南岸5份地衣样品中分离纯化到65株放线菌,分布在4个目8个科18个属,潜在新种5株,潜在新种菌和链霉菌为优势菌属。【结论】在本研究条件下,西双版纳可培养地衣放线菌多样性较白茫雪山和波罗的海南岸丰富。白茫雪山地衣链霉菌居多,潜在新种占比15.5%。3个地区地衣放线菌的区系组成各不相同,这与3个地区地衣所处地理环境、完全不同的气候密切相关。  相似文献

印度洋红树林沉积物可培养海洋放线菌多样性及其活性   总被引:2,自引:0,他引:2  
【目的】本研究旨在了解印度洋红树林沉积物可培养海洋放线菌多样性、抗菌活性及产酶活性。【方法】选用24种碳源为唯一能源培养基,利用稀释平板涂布方法对8个印度洋红树林沉积物样品进行分离,并基于16S rRNA基因系统发育分析的方法研究样品中海洋放线菌多样性;对分离得到的菌株进行抗菌活性和产酶活性检测。【结果】24种唯一碳源分离培养基中,非糖类碳源特别是甘油、丙氨酸分离效果最好,其次是多糖物质,最后是单糖。共分离得到521株海洋放线菌,经并菌后选取其中的139株代表性菌株测序,结果发现它们主要分布在放线菌纲7个亚目10个科的16个属,其中35个为潜在新种。有43.1%、33.3%、26.9%、25.5%、15.7%的实验菌株分别对枯草芽孢杆菌、白色念珠菌、大肠埃希氏菌、金黄色葡萄球菌、黑曲霉具有抑制作用;有36.5%、26.5%、22.4%、15.9%的实验菌株分别具有蛋白酶活性、纤维素酶活、淀粉酶活性、酯酶活性。【结论】印度洋红树林沉积物蕴藏着丰富的海洋放线菌资源,并具有较高生物活性,为后续工作提供良好的实验材料。  相似文献

【目的】为了从放线菌发现新的药物先导化合物,研究了川滇4个地区的放线菌多样性及其生物活性。【方法】采集250份土样,用4种培养基分离放线菌;从中选择98株代表菌进行了初步分类鉴定;采用琼脂扩散法,检测了169株放线菌对4种细菌和7种真菌的抑菌活性;利用特异性引物扩增法,测定了它们产生的聚酮合酶(PKSI、PKSⅡ)基因、非核糖体多肽合成酶(NRPS)基因和多烯类化合物合成酶(CYP)基因。【结果】黄荆老林的放线菌有13个属,峨眉山、青城山仅5个属,九寨沟9个属,西双版纳达20个属;不同地区的放线菌具有抗菌活性的菌株平均约占10%;有27%-36%的菌株产生PKSI、II、NRPS、CPY化合物合成基因。【结论】在采集样品的地区中,人类干扰越少,放线菌的多样性越高。分离放线菌时,使用"极端"条件,虽然分离到的放线菌数量可能不多,但获得未知菌的比例较大。添加抑制剂可减少革兰氏阴性细菌和真菌,有利于分离放线菌。  相似文献

【目的】勘探干涸的九莲城淖尔土壤放线菌多样性并进行活性筛选,以期发现药用微生物资源,为新抗生素的发现奠定基础。【方法】采用15种分离培养基,以稀释涂布法分离放线菌;根据分离菌株的16S rRNA基因序列同源性分析放线菌多样性;发酵液经乙酸乙酯萃取,菌丝体经丙酮浸提,获得提取浓缩物样品;样品通过纸片扩散法进行抗菌活性初筛;抗菌阳性菌株采用PCR技术进行Ⅰ型聚酮合酶(PKS I)KS域、Ⅱ型聚酮合酶(PKS II)KS域和非核糖体多肽合成酶(NRPS)A结构域抗生素生物合成基因的检测。【结果】从11份盐湖土壤样品中分离纯化到251株放线菌,其分布于放线菌纲的10个目15个科31个属,其中优势菌属为链霉菌属和拟诺卡氏菌属;251株放线菌中包括57株耐(嗜)盐放线菌,其优势菌属为拟诺卡氏菌属(22株)和涅斯捷连科氏菌属(15株)。基于16S r RNA基因序列的系统发育分析显示,菌株J11Y309为糖霉菌科潜在新属,菌株J12GA03为分枝杆菌科潜在新种。96株放线菌活性检测结果显示,56株至少对1株检定菌具有抗菌活性,阳性率为58.3%;56株有活性的放线菌中,47株至少含有1种抗生素生物合成基因,其中17株同时具有3种抗生素生物合成基因。【结论】干涸的九莲城淖尔土壤中含有较为丰富的药用放线菌资源,具有从中发现放线菌新物种和新抗生素的潜力。  相似文献

【目的】研究大连渤海老虎滩海域可培养放线菌的多样性。【方法】利用5种不同的培养基分离、培养海洋沉积物中的放线菌,并用16S rRNA基因序列对部分放线菌株进行系统发育分析。【结果】根据菌落表型共分离到1215株放线菌。选择271株具有代表性的菌株进行16S rRNA分析,结果表明,251株(92.26%)属于放线菌门,覆盖11个科,15个属;其余20株属于厚壁门和变形菌门;有7株为潜在的新种。【结论】大连渤海老虎滩海域的沉积物中存在较为丰富的放线菌和新种资源,这些菌株为将来开发新的微生物代谢产物奠定了基础。  相似文献

【背景】红树林生境拥有丰富的微生物资源,分离获得更多的纯培养放线菌为新型抗生素的发现提供菌种资源。【目的】使用新型放线菌分离培养基,发掘广西北部湾茅尾海红树林生境放线菌资源,评估新型放线菌分离培养基分离效果。【方法】设计出新型放线菌分离培养基——椰子汁-海藻糖培养基,对广西北部湾茅尾海红树林根际土壤和红树林根皮放线菌进行分离。基于分子生物学鉴定方法,对获得的纯培养放线菌进行多样性分析。【结果】从红树林根际土壤样品中分离获得65株放线菌,分布在5个目10个科15个属,使用YIM171培养基分离到7个属,MA培养基分离到5个属,椰子汁-海藻糖培养基分离到11个属,其中包括4株潜在新种;从红树林根皮样品中分离获得19株放线菌,分布在4个目7个科10个属,使用YIM171和椰子汁-海藻糖培养基分别分离到7个属,包括4株潜在新种;使用椰子汁-海藻糖培养基在红树林生境两类样品中获得3株潜在新种。【结论】广西北部湾茅尾海红树林根际土壤和根皮中放线菌多样性丰富,使用椰子汁-海藻糖培养基分离放线菌效果较佳,可用作放线菌的分离培养基。  相似文献

【目的】比较不同红树林地区的老鼠簕内生放线菌的地理分布,了解内生放线菌与其所处环境的相关性。【方法】分别从5个不同地点的红树林采集老鼠簕全株植物,采用9种分离培养基,从植株不同部位分离内生放线菌,用16S rRNA基因序列分析鉴定到属,用添加不同NaCl浓度的ISP 2液体培养基进行耐盐度测试,用无氮基础培养基进行固氮活性测试。【结果】共分离得到内生放线菌52株,其中从叶、茎和根部分别获得5株、2株和45株,花和果中未分离到。52株内生放线菌分别属于小单孢菌属(47株),链霉菌属(3株),疣孢菌属(1株)和继生菌属(1株)。48株菌表现出耐盐或嗜盐特征,其中18株最高耐盐度20%,4株不能在无盐条件下生长,12株菌可在含有3.3%NaCl的培养基上生长良好。4株菌可在无氮培养基下生长。【结论】对47株内生小单孢菌的地理分布分析表明,老鼠簕内生小单孢菌的类群因不同地理位置有很大差异。耐盐和固氮活性测试结果表明了老鼠簕内生放线菌对环境的适应性。  相似文献

【目的】研究微波处理对于分离嗜碱和嗜盐海洋放线菌的效果。【方法】用微波处理7份海泥样品,梯度稀释后涂布于3种分离培养基,分离具有嗜碱和嗜盐特性的海洋放线菌。【结果】微波处理后的7份样品中,4份样品中嗜碱海洋稀有放线菌和3份样品的嗜盐海洋稀有放线菌数量极显著提高;7份样品中的嗜碱、嗜盐海洋小单孢菌属、游动放线菌属、诺卡氏菌属等稀有放线菌数量均有显著增加,不同样品中新分离到链孢菌属、小双孢菌属、链孢囊菌属及其他未鉴定的海洋稀有放线菌,分离到属的数量提高了1-4个。【结论】微波处理不仅显著提高嗜碱和嗜盐海洋放线菌的分离数量,而且明显增加了海洋稀有放线菌的分离种类。  相似文献

【目的】进一步了解兴义喀斯特洞穴可培养放线菌资源及产活性代谢产物的能力。【方法】选取多种分离培养基,利用稀释直接涂布平板法对贵州黔西南兴义市多个喀斯特洞穴的土壤和岩石进行可培养放线菌资源分离;利用三种发酵培养基对相关放线菌进行生物产物初筛。【结果】根据16S rRNA基因序列的比对分析,将分离得到的251株放线菌分别归类到44个属,其中链霉菌属(Streptomyces)占分离菌株的比例为24.30%,小单孢菌属(Micromonospora)占比11.95%,红球菌属(Rhodococcus)占比9.16%,微杆菌属(Microbacterium)占比7.17%,诺卡氏菌属(Nocardia)占比6.37%,该五类放线菌为该地区可培养放线菌的优势菌群。对70株细菌进行活性次级代谢产物筛选,其中35株放线菌对指示菌具有抑制活性,且主要类群为链霉菌属和小单孢菌属。【结论】贵州兴义喀斯特洞穴中存在丰富多样的放线菌类群,且蕴藏大量具有产生活性次级代谢产物能力的菌株,为医药产业提供潜力菌株资源,极具进一步发掘和研究的价值。  相似文献

Colony-forming cyanobacteria of the genus Aphanizomenon form massive blooms in the brackish water of the Baltic Sea during the warmest summer months. There have been recent suggestions claiming that the Baltic Sea Aphanizomenon species may be different from Aphanizomenon flos-aquae found in lakes. In this study, we examined variability in the morphology and 16S-23S rRNA internal transcribed spacer (ITS) sequences of A. flos-aquae populations along a salinity gradient from a string of lakes to a fjord-like extension of the Baltic Sea to the open Baltic Sea. Morphological differences among the populations were negligible. We found that the Baltic Sea was dominated (25 out of 27 sequences) by one ITS1-S (shorter band of ITS 1 [ITS1]) genotype, which also was found in the lakes. The lake populations of A. flos-aquae tended to be genetically more diverse than the Baltic Sea populations. Since the lake ITS1-S genotypes of A. flos-aquae are continuously introduced to the Baltic Sea via inflowing waters, it seems that only one ITS1 genotype is able to persist in the Baltic Sea populations. The results suggest that one of the ITS1-S genotypes found in the lakes is better adapted to the conditions of the Baltic Sea and that natural selection removes most of the lake genotypes from the Baltic Sea A. flos-aquae populations.  相似文献

Marine organisms colonizing brackish habitats such as the Baltic Sea must cope with the negative effects of low salinities on reproductive success because these may reduce gamete viability and/or increase polyspermy. Reproductive characteristics of the marine seaweed Fucus vesiculosus L. were studied in several brackish habitats, particularly in the northern Baltic Sea, to understand its ability to reproduce where few other marine species survive. Polyspermy and fertilization success were variable at the boundary of the continuous distribution of F. vesiculosus in the Baltic Sea, and polyspermy was high (10%–30%) when fertilization was successful. A strong female bias (80%–86%, ca. 5.5:1) was found at the northernmost limit of Baltic F. vesiculosus. Electrophysiological studies showed that many eggs have a high input resistance (519 ± 150 MΩ[mean ± SE, n = 14] at Drivan, 1995), which may be helpful in preventing polyspermy in this brackish habitat. The polyspermy block remains sodium-dependent in the northern Baltic. Sperm bound quickly to northern Baltic eggs in natural water, but fertilization was delayed compared to marine F. vesiculosus. A subset of northern Baltic eggs studied during an optimal reproductive period (7–11 July 1995) had a membrane potential (Em) of ca. −100 mV and an effective fertilization potential (FP) of ca. 2 min with a plateau of −25 mV, but repolarized too rapidly for the FP to be protective. Pronuclear migration and cell wall secretion occurred more slowly in Baltic than in marine zygotes. The reproductive success of theseboundary populations may be dependent upon windows of opportunity when there are favorable combinations of the levels of salinity, water motion, population density, and sex ratio. These factors and the short duration of the reproductive season in the northern Baltic Sea may result in reproductive failure in some years.  相似文献

Two freshwater populations and one marine population (Baltic Sea) of threespine stickeback (Gasterosteus aculeatus) from Northeastern Germany were studied with regard to locomotory capacity: sustained swimming performance, activities of key enzymes in axial muscle, pectoral fin muscle and heart, and morphology. We postulated that life history differences between migratory Baltic Sea and resident freshwater populations could have led to a divergence in their locomotory capacity. The activity of citrate synthase (CS) in pectoral muscle correlated with critical swimming speed. Critical swimming speed, aerobic and anaerobic capacity of the pectoral fin muscle were population-specific. The Baltic Sea sticklebacks had a higher locomotory capacity (activity of CS in pectoral muscle, critical swimming speed) than sticklebacks of one freshwater population. However, another freshwater population expressed a similar locomotory capacity as the Baltic Sea population. In addition, Baltic Sea sticklebacks had a greater mass and lower anaerobic capacity of the pectoral fin muscle than the freshwater sticklebacks. The results are interpreted as an indication of a proceeding divergence between marine and resident freshwater populations and between freshwater populations of G. aculeatus originating from marine ancestors. The migratory Baltic Sea sticklebacks had better morphological prerequisites for sustained swimming than both freshwater populations, but there was no general difference in the locomotory capacity between marine and freshwater sticklebacks. However, their morphology could favour a more effective locomotion in the Baltic Sea sticklebacks.  相似文献

Length distributions of fish were analysed based on data from the German annual (November/December) bottom trawl surveys in the Mecklenburg Bight and Arkona Basin [ICES Subdivisions (SD) 22 and 24, respectively] of the southwest Baltic Sea, covering the period from 1991 to 2000. The length spectra for all fish species, and those for demersal and pelagic species separately, all showed a consistent pattern of declining logarithmic numbers with increasing length over the fully selected size range. Such consistency allowed the shape of the spectra to be used as a community property that could be tracked in space and time. There was a rapid reduction in the number of specimens in the larger size classes, resulting in steep slopes of the spectra, which may be due to intensive fishing pressure in the Baltic. The species composition of fish communities in the southwest Baltic Sea was stable during the period considered. No consistent species replacement was found, although the clupeid group dropped sharply in the catch during the 1996 survey, which might have been due to sudden environmental changes in that year.  相似文献

Fish otoliths' chronometric properties make them useful for age and growth rate estimation in fisheries management. For the Eastern Baltic Sea cod stock (Gadus morhua), unclear seasonal growth zones in otoliths have resulted in unreliable age and growth information. Here, a new age estimation method based on seasonal patterns in trace elemental otolith incorporation was tested for the first time and compared with the traditional method of visually counting growth zones, using otoliths from the Baltic and North seas. Various trace elemental ratios, linked to fish metabolic activity (higher in summer) or external environment (migration to colder, deeper habitats with higher salinity in winter), were tested for age estimation based on assessing their seasonal variations in concentration. Mg:Ca and P:Ca, both proxies for growth and metabolic activity, showed greatest seasonality and therefore have the best potential to be used as chemical clocks. Otolith image readability was significantly lower in the Baltic than in the North Sea. The chemical (novel) method had an overall greater precision and percentage agreement among readers (11.2%, 74.0%) than the visual (traditional) method (23.1%, 51.0%). Visual readers generally selected more highly contrasting zones as annuli whereas the chemical readers identified brighter regions within the first two annuli and darker zones thereafter. Visual estimates produced significantly higher, more variable ages than did the chemical ones. Based on the analyses in our study, we suggest that otolith microchemistry is a promising alternative ageing method for fish populations difficult to age, such as the Eastern Baltic cod.  相似文献

We describe an unusually high infection rate of Gyrodactylus salaris Malmberg in juvenile Atlantic salmon Salmo salar L. of Baltic Sea origin, which are generally believed to be more resistant to G. salaris than East Atlantic salmon populations. Based on analyses of mitochondrial (complete cytochrome oxidase 1 [CO1] gene, 1548 bp) and nuclear (ADNAM1, 435 bp; internal transcribed spacer [ITS] rDNA region, 1232 bp) DNA fragments, the closest relatives of the characterized Estonian G. salaris strain were parasites found off the Swedish west coast and in Raasakka hatchery, Iijoki (Baltic Sea, Finland). Analyses of 14 microsatellite loci of the host S. salarrevealed that approximately 40% of studied fish were triploids. We subsequently identified triploid Atlantic salmon of Baltic origin as more susceptible to G. salaris infection than their diploid counterparts, possibly due to compromised complement-dependent immune pathways in triploid salmon. This is in accordance with earlier studies that have shown elevated susceptibility of triploids to various viral or bacterial pathogens, and represents one of the first reports of increased susceptibility of triploid salmonid fish to an ectoparasite. However, further experimental work is needed to determine whether triploid Atlantic salmon is generally more susceptible to G. salaris compared to their diploid counterparts, irrespective of the particular triploidization method and population of origin.  相似文献
