Simultaneous nitrification and denitrification using a mixed methanotrophic culture was investigated. When both NO3 -N (108 mg l–1) and NH3-N (59 mg l–1) were added into batch reactors, nitrate removal was complete within 10 h at the rate of 47 mg NO3 -N g VSS–1 day–1 when dissolved oxygen (DO) concentration was maintained at 2 mg DO l–1. Ammonia removal started simultaneously with nitrate removal at a slower rate of 14 NH3-N g VSS–1 day–1. No significant accumulation of nitrite or nitrate during ammonia utilization suggested the occurrence of simultaneous nitrification and denitrification.  相似文献

Hydrological and chemical structures off the Rhône River estuary resulting from the introduction of the river flow into the Mediterranean Sea are described. The effect of the fresh-water/sea-water interface on the distribution of inorganic and organic matter off the Rhône river is investigated. Strong vertical gradients of inorganic and dissolved organic matter such as lipids characterized the first few meters in this area (from 83.7 to 0.6 N-NO3 µgat l–1, from 6.39 to 0.92 N-NH4 µgat l–1 and from 299 to 73 µg l–1 of total dissolved lipids). At the interface, substantial increases of particulate organic (PON: from 45 µg l–1 at surface to 118 µg l–1 at the interface, POC: from 462 to 876 µg l–1, total particulate lipids: from 33 to 648 µg l–1) and suspended matter in general (from 18 to 22.2 mg l–1) were observed. High phytoplanktonic production may account for some of this enrichment, although passive accumulation might also be involved.  相似文献

Summary The growth parameters ofPenicillium cyclopium have been evaluated in a continuous culture system for the production of fungal protein from whey. Dilution rates varied from 0.05 to 0.20 h–1 under constant conditions of temperature (28°C) and pH (3.5). The saturation coefficients in the Monod equation were 0.74 g l–1 for lactose and 0.14 mg l–1 for oxygen, respectively. For a wide range of dilution rates, the yield was 0.68 g g–1 biomass per lactose and the maintenance coefficient 0.005 g g–1 h–1 lactose per biomass, respectively. The maximum biomass productivity achieved was 2 g l–1 h–1 biomass at dilution rates of 0.16–0.17 h–1 with a lactose concentration of 20 g l–1 in the feed. Summary Submerged batch cultivation under controlled environmental conditions of pH 3.8, temperature 30°C, and KLa200 h–1 (above 180 mMO2 l –1 h–1 oxygen supply rate) produced a maximum (12.0 g·l –1) SCP (Candida utilis) yield on the deseeded nopal fruit juice medium containing C/N ratio of 7.0 (initial sugar concentration 25 g·l –1) with a yield coefficient of 0.52 g cells/g sugar. In continuous cultivation, 19.9 g·l –1 cell mass could be obtained at a dilution rate (D) of 0.36 h–1 under identical environmental conditions, showing a productivity of 7.2 g·l –1·h–1. This corresponded to a gain of 9.0 in productivity in continuous culture over batch culture. Starting with steady state values of state variables, cell mass (CX–19.9 g·l –1), limiting nutrient concentration (Cln–2.5 g·l –1) and sugar concentration (CS–1.5 g·l –1) at control variable conditions of pH 3.8, 30°C, and KLa 200 h–1 keeping D=0.36 h–1 as reference, transient response studies by step changes of these control variables also showed that this pH, temperature and KLa conditions are most suitable for SCP cultivation on nopal fruit juice. Kinetic equations obtained from experimental data were analysed and kinetic parameters determined graphically. Mathematical model parameters for the methanogenic degradation of propylene glycol were estimated in a sequential manner by means of an optimization technique. Model parameters determined from an initial experimental data set using one bioreactor were then verified with the results from a second bioreactor. The proposed methodology is a useful tool to obtain model parameters for continuous flow reactors with completely mixed regime. A modified Rotating Biological Contactor (RBC) was used for the treatability studies of synthetic tapioca wastewaters. The RBC used was a four stage laboratory model and the discs were modified by attaching porous nechlon sheets to enhance biofilm area. Synthetic tapioca wastewaters were prepared with influent concentrations from 927 to 3600 mg/l of COD. Three hydraulic loads were used in the range of 0.03 to 0.09 m3·m–2·d–1 and the organic loads used were in the range of 28 to 306 g COD· m–2·d–1. The percentage COD removal were in the range from 97.4 to 68. RBC was operated at a rotating speed of 18 rpm which was found to be the optimal rotating speed. Biokinetic coefficients based on Kornegay and Hudson models were obtained using linear analysis. Summary Chaetomium cellulolyticum (ATCC 32319) was cultivated on glucose, Avicel and/or Sigmacell in a 20-1 stirred tank batch reactor. The substrate (cellulose) concentration, the cell mass concentration (through protein and/or nitrogen content), reducing sugar concentration, the enzyme activity, the alkali consumption rate, the dissolved O2 and CO2 concentrations in the outlet gas were measured. The specific growth rate, the substrate yield coefficient, cell productivity, the oxygen consumption rate, the CO2 production rate and the volumetric mass transfer coefficient were determined. At the beginning of the growth phase the oxygen utilization rate exhibits a sharp maximum. This maximum could be used to start process control. Xylose production by Candida guilliermondii FTI 20037 was carried out in a synthetic medium in the presence of 0–100 g methanol l–1, 0–0.7 g furfural l–1 or 0–1.3 g acetic acid l–1. Kinetic results show a mixed inhibition mechanism in all three cases. Maximum specific productivity and saturation constant for product formation were, in the absence of inhibition, 3.6 gP gX –1 h–1 and 232 gS l–1, respectively, while the inhibition constants, K i and K i, were 17 and 50 g methanol l–1, 0.62 and 7.0 g furfural l–1, 0.69 and 3.5 g acetic acid l–1, which suggests the following order of inhibition: furfural > acetic acid > methanol.  相似文献

Summary Optimal growth conditions for Zymomonas mobilis have been established using continuous cultivation methods. Optimal substrate utilization efficiency occurs with 2.5 g l–1 yeast extract, 2.0 g l–1 ammonium sulfate and 6.0 g l–1 magnesium sulfate in the media. Catabolic activity is at its maximum with glucose uptake rates of 16–18 g l–1 h–1 and ethanol production rates of 8–9 g l–1 h–1, Qg values of 22–26 and Qp values between 11 and 13, which results in 40 g l–1 h–1 ethanol yields using a 100 g l–1 substrate feed. Any increase in these parameters goes on cost of substrate utilization efficiency. Summary The influence of temperature on the growth of the theromophilic Bacillus caldotenax was investigated using chemostat techniques and a chemically defined minimal medium. All determined growth constants, that is maximal specific growth rate, yield and maintenance, were temperature dependent. It was striking that the very large maintenance requirement was about 10 times higher than for mesophilic cells under equivalent conditions. A death rate, which was very substantial at optimal and supraoptimal growth temperatures, was estimated by comparing the maintenance for substrate and oxygen. Impacts of solar radiation, humic substances and nutrients on phytoplankton abundance at different depths were investigated in a temperate dimictic lake, Lake Solumsjö. Penetration of solar radiation profiles at different depths, represented as light attenuation coefficient (K d) were examined. Water sampling and downward irradiance of photosynthetically active radiation (PAR), ultraviolet-A (UV-A, 320–400 nm) and ultraviolet-B (UV-B, 280–320 nm) radiation were performed once a week and at three different times of the day (08.00, 12.00 and 16.00 hrs, local time) between September 13 and November 1, 1999. During the period of investigation, solar radiation above the water surface declined from 474 to 94 mol m–2 s–1 for PAR, from 1380 to 3.57 W m–2 for UV-A and from 13.1 to 0.026 W m–2 for UV-B, respectively. The attenuation coefficient (K d) for UV-B radiation ranged from 3.7 to 31 m–1 and UV-B radiation could not be detected at depths greater than 0.25 m. Humic substances measured at 440 nm ranged from 35.5 to 57.7 Pt mg l–1. Mean values of biomass, estimated from chlorophyll a, in the whole water column (0–10 m) varied between 2.3 and 5.6 g l–1 and a diel fluctuation was observed. During stratified conditions, high levels of iron (1.36 mg l–1) and manganese (4.32 mg l–1) were recorded in the hypolimnion, suggesting that the thermocline played a major role in the vertical distribution of phytoplankton communities in Lake Solumsjö. The high levels of iron and manganese stimulated the growth of Trachelomonas volvocinopsis in the hypolimnion at a depth of 10 m. Negative impacts of UV-B radiation on phytoplankton in lake Solumsjö are reduced due to the high levels of humic substances and the high degree of solar zenith angle at the latitude studied.  相似文献

Summary Recently we proposed that cytoplasmic acidification of low K+ (LK) sheep erythrocytes may stimulate ouabain-resistant Cl-dependent K+ flux (K+Cl cotransport), also known to be activated by cell swelling, treatment with N-ethylmaleimide (NEM), or removal of cellular bivalent cations. Here we studied the dependence of K+ transport on intracellular and extracellular pH (pH i , pH o ) varied either simultaneously or independently using the Cl/HCO 3 exchange inhibitor 4,4, diisothiocyanatostilbene-3,2-disulfonic acid (DIDS). In both control and NEM-treated LK cells volumes were kept near normal by varying extracellular sucrose. Using DIDS as an effective pH clamp, both K+ efflux and influx of Rb+ used as K+ congener were strongly activated at acid pH i and alkaline pH o . A small stimulation of K+ (Rb+) flux was also seen at acid pH i in the absence of DIDS, i.e., when pH i pH o . Anti-L l serum, known to inhibit K+Cl cotransport, prevented the pH i -stimulated K+ (Rb+) fluxes. Subsequent to NEM treatment at pH 6, K+ (Rb+) fluxes were activated only by raising pH, and thus were similar to the pH activation profile of K+ (Rb+) fluxes in DIDS-treated cells with pH o varied at constant physiologic pH i . Anti-L l , which inhibited NEM-stimulated K+ (Rb+) fluxes, failed to do so in NEM-plus DIDS-treated cells. Thus, NEM treatment interferes with the internal but not with the external pH-sensitive site.  相似文献

The kinetic properties of a microsomal gill (Na+,K+)-ATPase from the blue crab Callinectes danae were analyzed using the substrate p-nitrophenylphosphate. The (Na+,K+)-ATPase hydrolyzed PNPP obeying cooperative kinetics (n=1.5) at a rate of V=125.4±7.5 U mg−1 with K0.5=1.2±0.1 mmol l−1; stimulation by potassium (V=121.0±6.1 U mg−1; K0.5=2.1±0.1 mmol l−1) and magnesium ions (V=125.3±6.3 U mg−1; K0.5=1.0±0.1 mmol l−1) was cooperative. Ammonium ions also stimulated the enzyme through site–site interactions (nH=2.7) to a rate of V=126.1±4.8 U mg−1 with K0.5=13.7±0.5 mmol l−1. However, K+-phosphatase activity was not stimulated further by K+ plus NH4+ ions. Sodium ions (KI=36.7±1.7 mmol l−1), ouabain (KI=830.3±42.5 μmol l−1) and orthovanadate (KI=34.0±1.4 nmol l−1) completely inhibited K+-phosphatase activity. The competitive inhibition by ATP (KI=57.2±2.6 μmol l−1) of PNPPase activity suggests that both substrates are hydrolyzed at the same site on the enzyme. These data reveal that the K+-phosphatase activity corresponds strictly to a (Na+,K+)-ATPase in C. danae gill tissue. This is the first known kinetic characterization of K+-phosphatase activity in the portunid crab C. danae and should provide a useful tool for comparative studies.  相似文献

A bacterial strain PNS-1, isolated from activated sludge derived from a domestic wastewater treatment unit, could utilize 4-aminobenzenesulphonate (4-ABS) as a sole organic carbon and energy source under aerobic conditions. Degradation rate varied with the initial concentration of 4-ABS and maximum specific substrate removal rate was observed at 400mg 4-ABS l–1 (2.3mM). Average biomass yield was 0.31mg/mg 4-ABS degraded. Biokinetic parameters for the degradation, determined using the Haldane relationship, were 0.26h–1 (max), 6mg\,l–1 (KS) and 4020mg\,l–1 (Ki). Strain PNS-1 could not utilize other isomers of benzenesulphonate and 5-sulphosalicylate as growth substrates whereas protocatechuate, pyrocatechuate and p-hydroxybenzoate could be degraded. In mixed substrate batch cultivations, where 4-ABS was one of the component, protocatechuate and 4-ABS were simultaneously utilized. Presence of 2- or 3-ABS decreased the growth and substrate degradation rates of 4-ABS. With 4-ABS and pyrocatechuate, although a lag phase was observed prior to pyrocatechuate degradation, a diauxic growth pattern was not seen.  相似文献

Denitrification was studied in anoxic batch cultures of a simulated fish processing wastewater at 37 r C and pH 7.5, using a denitrifying enrichment culture from fishery wastewater. Different initial nitrate to biomass ratios (So/Xo) were used: nitrate and biomass varied from 7.5 to 94.7 mg NO3-N l–1, and from 20 to 4300 mg volatile suspended solids l–1, respectively. The specific maximum denitrification rate (r m) and the cell yield (Y X / S) depended on the So/Xo ratio under anoxic conditions: r m increased from 1.2 to 1584 mg NO3-N g–1 VSS h–1 and Y X / S decreased from 42 to 0.03 mg VSS mg–1 NO3-N when So/Xo varied from 5.5 10– 3 to 9.3 mg NO3-N/mg VSS. The potential of a cyanobacteriumPhormidium sp., for the tertiary treatment of piggery waste-water chemical oxygen demand , 3000 mg·1–1, using an aerobically stabilized secondary effluent, was studied. Batch cultures were carried out in 2-1 glass reactors and in a 30-1 glass-fibre carousel reactor. The nutrient removal efficiency as well as biomass production were compared in synthetic mineral medium and in different concentrations of aerobically stabilized piggery effluent. The best performance ofPhormidium sp. ocurred in diluted stabilized secondary effluent (1:1). Removal efficiencies were 100% for P-PO4 –3, 50% for N-NH4 –1 and 35% for N-NO3 in small-volume cultures; and 31% for P-PO4 –3, 100% for N-NH4 and 70% for N-NO3 in an open carousel reactor. Biomass production on the aerobically stabilized effluent was six times higher in the 2-1 reactors and 1.7 times higher in the carousel reactor when compared to the synthetic medium.  相似文献

Summary We have measured the intracellular potassium activity, [K+]i and the mechanisms of transcellular K+ transport in reabsorptive sweat duct (RSD) using intracellular ion-sensitive microelectrodes (ISMEs). The mean value of [K+]i in RSD is 79.8±4.1mm (n=39). Under conditions of microperfusion, the [K+]i is above equilibrium across both the basolateral membrane, BLM (5.5 times) and the apical membrane, APM (7.8 times). The Na+/K+ pump inhibitor ouabain reduced [K+]i towards passive distribution across the BLM. However, the [K+]i is insensitive to the Na+/K+/2 Cl cotransport inhibitor bumetanide in the bath. Cl substitution in the lumen had no effect on [K+]i. In contrast, Cl substitution in the bath (basolateral side) depolarized BLM from –26.0±2.6 mV to –4.7*±2.4 mV (n=3;* indicates significant difference) and decreased [K+]i from 76.0±15.2mm to 57.7* ±12.7mm (n=3). Removal of K+ in the bath decreased [K+]i from 76.3±15.0mm to 32.3*±7.6mm (n=4) while depolarizing the BLM from –32.5±4.1 mV to –28.3*±3.0 mV (n=4). Raising the [K+] in the bath by 10-fold increased [K+]i from 81.7±9.0mm to 95.0*±13.5mm and depolarized the BLM from –25.7±2.4 mV to –21.3*±2.9 mV (n=4). The K+ conductance inhibitor, Ba2+, in the bath also increased [K+]i from 85.8±6.7mm to 107.0*±11.5mm (n=4) and depolarized BLM from –25.8±2.2 mV to –17.0*±3.1 mV (n=4). Amiloride at 10–6 m increased [K+]i from 77.5±18.8mm to 98.8*±21.6mm (n=4) and hyperpolarized both the BLM (from –35.5±2.6 mV to –47.8*±4.3 mV) and the APM (from –27.5±1.4 mV to –46.0* ±3.5 mV,n=4). However, amiloride at 10–4 m decreased [K+]i from 64.5±0.9mm to 36.0*±9.9mm and hyperpolarized both the BLM (from –24.7±1.4 mV to –43.5*±4.2 mV) and APM (from –18.3±0.9 mV to –43.5*±4.2 mV,n=6). In contrast to the observations at the BLM, substitution of K+ or application of Ba2+ in the lumen had no effect on the [K+]i or the electrical properties of RSD, indicating the absence of a K+ conductance in the APM. Our results indicate that (i) [K+]i is above equilibrium due to the Na+/K+ pump; (ii) only the BLM has a K+ conductance; (iii) [K+]i is subject to modulation by transport status; (iv) K+ is probably not involved in carrier-mediated ion transport across the cell membranes; and (v) the RSD does not secrete K+ into the lumen.  相似文献

Summary Transbasal electrical potential (V b) and intraepithelial potassium chemical activity ((K+) i ) were measured in isolated midgut epithelium of tobacco hornworm (Manduca sexta) using double-barrelled glass microelectrodes. Values ofV b ranging from +8 to –48 mV (relative to blood side) were recorded. For all sites, (K+) i is within a few millivolts of electrochemical equilibrium with the blood side bathing solution. Sites more negative than –20 mV show relatively high sensitivity ofV b to changes in blood side K+ concentration: 43% of these sites can be marked successfully with iontophoresed Lucifer yellow CH dye and shown to represent epithelial cells of all three types present in the midgut. In about half of successful marks, dye-coupling of several adjacent cells is seen. Low potential sites — those withV b less negative than –20 mV —typically do not show high sensitivity ofVb to changes of external K+, but rather (K+) i rapidly approaches the K+ activity of blood side bathing solution. These sites can seldom be marked with Lucifer yellow (4% success). The mean (K+) i of the high potential sites is 95±29 (sd)mm under standard conditions, a value which is in accord with published values for the whole tissue.  相似文献

A method for isolation of d-amino acid oxidase (DAAO) from disrupted Trigonopsis variabilis cells has been developed. In an aqueous two-phase system consisting of PEG6000 (220 g l–1), potassium phosphate (110 g l–1, K2HPO4 + KH2PO4 = 10.1:1, mol mol–1) and dl-methionine (11 g l–1), the major portion of cellular proteins (87%) was partitioned into the salt phase. By sequential extraction, 48% of DAAO was recovered in PEG phase, giving a yield of 211 U mg protein–1.  相似文献
