Urinary 1-hydroxypyrene (1-OHP), a biomarker of polycyclic aromatic hydrocarbons (PAHs) exposure, may be influenced by metabolic gene polymorphisms. Such knowledge could benefit us in understanding the inter-individual difference in the mechanism of PAHs-induced carcinogenesis. We investigated the influence of gene polymorphisms on urinary 1-OHP concentrations in 447 coke oven workers from two coking plants in south China. After adjustment for age, plant, level of occupational exposure, body mass index, level of education, alcohol consumption, cigarette smoking and respirator usage, AhR R554K (rs2066853), UGT1A1 -3263T>G (rs4124874) and GSTP1 I105V (rs1695) were associated with urinary 1-OHP excretion with the p-value of 0.053, 0.006 and 0.021, respectively. The concentrations of urinary 1-OHP (Geometric mean, micromol/mol creatinine) in the homozygous major variant carriers and homozygous minor variant carriers for AhR R554K, UGT1A1 -3263T>G and GSTP1 I105V were listed as follows: 4.20 and 5.12, 5.11 and 3.92, 4.93 and 2.91, respectively. GSTT1 present carriers had a significantly higher urinary 1-OHP level than that in null carriers in the case with AhR R554K GA/AA carriers (5.17 vs. 3.64 micromol/mol creatinine, p=0.038), as well as in the case with UGT1A1 -3263T>G TG/GG carriers (5.67 vs. 3.38 micromol/mol creatinine, p=0.001). These results showed that AhR, UGT1A1, GSTP1 and GSTT1 polymorphisms were associated with urinary 1-OHP concentrations in Chinese coke oven workers. No influence was found in the association between urinary 1-OHP and other genetic polymorphisms such as CYP1A1, CYP1A2, CYP1B1, CYP2E1, EPHX1, EPHX2 in this population.  相似文献

Particulate air pollution is significantly elevated during the winter in Christchurch, New Zealand, largely attributable to use of wood burners for domestic home heating, topography, and meteorological conditions. Polycyclic aromatic hydrocarbons (PAHs) are a key component of airborne particulate matter (PM) and urinary 1-hydroxypyrene (1-OHP) has previously been used to assess exposure of people to PAHs. We examined urinary 1-OHP in Christchurch male non-smoking schoolchildren (12-18 yr) on two occasions after high pollution events (48 and 72 microg PM(10)/m(3) 24-h average) and two occasions during periods of low pollution (19 and 12 microg PM(10)/m(3)). Concentrations of urinary 1-OHP were significantly elevated in the students during high pollution events (median (mean+/-SD) 0.043 (0.051+/-0.032) and 0.042 (0.060+/-0.092) micromol OHP/mol creatinine respectively) compared with low pollution periods (median (mean+/-SD) 0.019 (0.026+/-0.032) and 0.025 (0.028+/-0.018) micromol/mol creatinine respectively). The observed 1-OHP concentrations are at the lower end of those determined in children and non-occupationally exposed adults in international studies and suggest a generally low exposure to PAHs. The increased urinary 1-OHP concentrations following nights of elevated particulate concentrations in ambient air suggest increased exposure to ambient air pollution during winter time, and could potentially be used as a biomarker of exposure in this population.  相似文献

It was postulated that a population in sub-Saharan Africa, known to be at high risk for aflatoxicosis due to frequent ingestion of aflatoxin (AF)-contaminated foods could also be exposed to polycyclic aromatic hydrocarbons (PAHs) from a variety of environmental sources. Previously, participants in this population were shown to be highly exposed to AFs, and this exposure was significantly reduced by intervention with NovaSil clay (NS). Objectives of this study were 1) to assess PAH exposure in participants from the AF study using urinary biomarker 1-hydroxypyrene (1-OHP); 2) examine the effect of NS clay and placebo (cellulose) treatment on 1-OHP levels; and 3) determine potential association(s) between AF and PAH exposures. A clinical trial was conducted in 177 Ghanaians who received either NS capsules as high dose or low dose, or placebo (cellulose) for a period of 3 months. At the start and end of the study, urine samples were analyzed for 1-OHP. Of the 279 total samples, 98.9% had detectable levels of 1-OHP. Median 1-OHP excretion in nonsmokers was 0.64 µmol/mol creatinine at baseline and 0.69 µmol/mol creatinine after 3 months. Samples collected at both time points did not show significant differences between placebo and NS-treated groups. There was no linear correlation between 1-OHP and AF-albumin adduct levels. Results show that this population is highly exposed to PAHs (and AFs), that NS and cellulose treatment had no statistically significant effect on 1-OHP levels, and that this urinary biomarker was not linearly related with AF exposure.  相似文献

1-hydroxypyrene (1-OHP) is a biomarker of exposure to polycyclic aromatic hydrocarbons (PAHs). PAHs are pollutants produced by the combustion of organic material, and could be found both indoors and outdoors. Prenatal exposure to these compounds has been associated with retarded intrauterine growth and respiratory problems in children. The objective of this study is to describe the 1-OHP levels in urine in pregnant women in the INMA cohort in Valencia, and to study the association with socio-demographic and lifestyle variables, as well as with indicators of air pollution. The study population is a sub-sample (n: 204) of a cohort of pregnant women in the multicentre INMA project in Valencia (Spain). Urine samples were taken in week 12 of gestation, and 1-OHP was analyzed, and adjusted for creatinine. Socio-demographic and lifestyle information was collected using a questionnaire. Data from the monitoring stations in Valencia were used to assign levels of exposure to nitrogen dioxide (NO2), sulphur dioxide (SO2), total suspended particulates (TSP) and carbon monoxide (CO). The median 1-OHP level was 0.061 muicrool/mol of creatinine. Women who smoked had higher 1-OHP levels (0.095 micromol/mol of creatinine) than non-smokers (0.050 micromol/mol of creatinine). The variables predicting 1-OHP levels on linear multivariate analysis were: tobacco consumption, consumption of fried and smoked foods. There was also a significant association with NO2 levels 4 and 5 days before the samples were taken. The 1-OHP levels found in pregnant women in Valencia were higher than those found among women in reports in USA. 1-hydroxypyrene could be a good marker of exposure to PAHs from tobacco and diet, and also a useful marker of short term exposure to air pollution.  相似文献

Indoor air pollution is considered to be a serious public health issue in Mexico; therefore, more studies regarding this topic are necessary. In this context, we assessed exposure to polycyclic aromatic hydrocarbons (PAHs) and volatile organic compounds in: (i) women who use firewood combustion (indoor) for cooking and heating using traditional open fire; (ii) women who use firewood combustion (outdoor) for cooking and heating using traditional open fire; and (iii) women who use LP gas as the principal energy source. We studied 96 healthy women in San Luis Potosi, México. Urine samples were collected, and analyses of the following urinary exposure biomarkers were performed by high-performance liquid chromatography: 1-hydroxypyrene (1-OHP), trans, trans-muconic acid, and hippuric acid (HA). The highest levels of 1-OHP, trans, trans-muconic acid, and HA were found in communities where women were exposed to indoor biomass combustion smoke (or products; geometric mean ± s.d., 3.98 ± 5.10 μmol/mol creatinine; 4.81 ± 9.60 μg/l 1-OHP; 0.87 ± 1.78 mg/g creatinine for trans, trans-muconic acid; and 1.14 ± 0.91 g/g creatinine for HA). Our findings indicate higher exposure levels to all urinary exposure biomarkers studied in women who use indoor firewood combustion for cooking and heating (using traditional open fire). PRACTICAL IMPLICATIONS: High mean levels of 1-hydroxypyrene, t,t-muconic acid, and hippuric acid were found in women who use firewood combustion (indoor) for cooking and heating using traditional open fire and taking into account that millions of women and children in Mexico are living in scenarios similar to those studied in this report, the assessment of health effects in women and children exposed to polycyclic aromatic hydrocarbons and volatile organic compounds is urgently needed. Moreover, it is immediately necessary an intervention program to reduce exposure.  相似文献

The exposure of temple workers to polycyclic aromatic hydrocarbons   总被引:1,自引:0,他引:1  
Five temples, each a different size, were selected for this study. Two of the temples were located in Taichung City (in central Taiwan), and three were in Tainan City (in Southern Taiwan). Aerosols smaller than 10 microm aerodynamic diameter (PM(10)) were collected by using personal collection samplers during pilgrim days (the first and fifteenth day of each lunar month) and normal days (all other days). Regression analysis showed that about 1.6 microg/m(3) of PM(10) contributed to the workers' exposure in the temples for each joss stick increase in the censer. The concentrations of total polycyclic aromatic hydrocarbons (t-PAHs) and carcinogenic PAHs (car-PAHs) on pilgrim days were higher than those on normal days. Mean concentrations of urinary 1-hydroxypyrene (1-OHP) in the pre-shift and post-shift workers of the five temples on normal days were 1.20 and 1.61 microg/g creatinine, respectively. Furthermore, the post-shift concentrations of 1-OHP in the workers of temples on pilgrim days were all higher than those of the workers of corresponding temples on normal days. Significant relationships between the urinary concentrations of 1-OHP and the exposure concentrations of pyrene, total PAHs and BaP(eq) were found in the workers of the temples. Results of linear regression showed that the increase of unit concentration (1 ng/m(3)) of pyrene led to a 0.05 microg/g creatinine increase of urinary 1-OHP, while the increase of unit concentration (1 ng/m(3)) of BaP(eq) resulted in an increase of 0.03 microg/g creatinine of urinary 1-OHP.  相似文献

Aims:

Exposure to polycyclic aromatic hydrocarbons (PAH), among the main compounds present in polluted urban air, is of concern for children's health. Childhood exposure to PAH was assessed by urinary monitoring of 1-hydroxypyrene (1-OHP), a pyrene metabolite, investigating its association with exposure to air pollution and other factors related to PAH in air.

Methods:

A group of 174 4-year-old children were recruited and a questionnaire on their indoor and outdoor residential environment was completed by parents. At the same time, environmental measurements of traffic-related air pollution (NO2) were carried out. A urine sample was collected from each child in order to analyze 1-OHP using HPLC with fluorescence detection, correcting for creatinine concentrations. Non-parametric tests and regression analyses were used to identify environmental factors that influence 1-OHP excretion.

Results:

Mean urinary 1-OHP concentration was 0.061 μmol/mol creatinine, ranging from 0.004 to 0.314 μmol/mol. Non-parametric tests and regression analysis showed positive and significant associations (P ≤ 0.05) between 1-OHP and predicted residential exposure to NO2 (which was based on outdoor environmental measurements and geo-statistical analysis), self-reported residential vehicle traffic, passive smoking and cooking appliance. 1-OHP levels tended to be higher among children living in urban areas (0.062 μmol/mol vs. 0.058 μmol/mol for children living in rural areas) but differences were not significant (P = 0.20).

Conclusion:

In Southern Spain, concentrations of urinary 1-OHP were in the lower range of those generally reported for children living in non-polluted areas in Western Europe and the USA. Traffic-related air pollution, passive smoking and cooking appliance influenced urinary 1-OHP level in the children, which should be prevented due to the health consequences of the inadvertent exposure to PAH during development.  相似文献

Cytochrome P450 1B1 (CYP1B1) is induced through the Ah receptor and is involved in the activation of polycyclic aromatic hydrocarbons (PAHs). To determine the validity of a quantitative analysis of CYP1B1 mRNA in peripheral human blood cells for the estimation of PAH exposure, a real-time quantitative polymerase chain reaction method was used to measure the relative levels of CYP1B1 mRNA in 37 Chinese coke oven workers and 13 control workers. A large inter-individual difference in the levels was observed. The average level of the CYP1B1 mRNA in workers at the top work site, where the PAH exposure level from the coke ovens was highest, was significantly higher than in workers at the middle site (P<0.01) or the controls (P=0.02). A non-significant positive correlation was found between the CYP1B1 mRNA levels and urinary 1-hydroxypyrene (R=0.22, P=0.13), and a significant correlation between these mRNA levels and urinary cotinine (R=0.33, P=0.02). It was interesting that a significant positive correlation between CYP1B1 mRNA and 1-hydroxypyrene was observed in subjects with the Leu/Leu type of CYP1B1 Leu432Val polymorphism (R=0.33, P=0.02, n=38) and a non-significant correlation in subjects with the Leu/Val and Val/Val types (R=-0.36, P=0.25, n=12), although the number of subjects in this strata analysis was small. Our preliminary study suggests that PAH exposure in coke ovens and smoking maybe associated with CYP1B1 mRNA levels in peripheral blood cells although mRNA is generally unstable and could be expressed following exposure to other agents.  相似文献

Indoor air pollution can be an important risk factor for human health, considering that people spend more than 60% of their time indoors. Fifty percent of the world population and approximately 90% of the rural population in developing countries are using biomass as energy source. Latin America represents 12% of the global consumption of biomass; in Mexico, 27 million people use wood as an energy source. Therefore, in this study we evaluated a 3-stage risk reduction program. The stages were: 1) removal of indoor soot adhered to roofs and internal walls; 2) paving the dirt floors; and 3) introduction of a new wood stove with a metal chimney that expels smoke outdoors. The complete intervention program was applied. In 20 healthy subject residents from an indigenous community in San Luis Potosí, Mexico, we measured blood carboxyhemoglobin (% COHb), DNA damage (comet assay) in nucleated blood cells, and urinary 1-OHP levels before and after the program. Before intervention individuals had a geometric mean COHb level of 4.93% and 53% of the population presented levels above 2.5% considered a safe level. However, in all the studied individuals the levels of COHb were reduced to below 2.5% (mean level 1.0%) one month after the intervention. Moreover, when compared, DNA damage in people exposed before the intervention was higher (5.8+/-1.3 of Tail Moment) than when the program was introduced (2.8+/-0.9 of Tail Moment) (P>0.05) and a same trend was observed with urinary 1-OHP levels; 6.71+/-3.58 micromol/mol creatinine was the concentration before intervention; whereas, 4.80+/-3.29 micromol/mol creatinine was the one after the program. The results suggest that the intervention program offers an acceptable risk reduction to those families that use biomass for food cooking.  相似文献

Levels of selenium and mercury in blood and urine were analysed in 37 male workers exposed to elemental mercury vapour in a chloralkali plant and in 39 unexposed controls of the same age. Mean urinary Hg was 223 nmol l-1 (15 nmol/mmol creatinine) in the exposed group and 26 nmol l-1 (2.0 nmol/mmol creatinine) in the controls. Mean blood and plasma Hg levels were 46 and 36 nmol l-1, respectively, in the exposed group, as compared with 17 and 7 nmol l-1 in the controls. The concentrations of Se in plasma and erythrocytes did not differ between the two groups. Urinary Se levels were, however, slightly but significantly lower in the exposed group (median values 23 vs 29 nmol/mmol creatinine), and there was a negative correlation between urinary Se and plasma Hg in the exposed group. This may be due to a retention of Se in the kidneys. In a subgroup of exposed workers and controls, glutathione peroxidase, superoxide dismutase and catalase were also analysed. No differences were found between the groups with respect to these antioxidative enzymes. The effect on Se status of moderate Hg exposure seems to be of minor clinical importance.  相似文献

A study was carried out on cobalt (Co) excretion in the urine of 12 workers exposed to known cobalt concentrations in the stone cutting diamond wheel production and in six volunteers: four of these were exposed in the same work environment for a whole workshift and the other two were exposed to cobalt in a cabin under experimental conditions. The kinetics of the urinary excretion was multiphase: (i) a first stage of rapid elimination ( ); (ii) a second phase of slower elimination ( ); (iii) a longer period of retention, of the order of years, in subjects with higher exposure. In the control group (4 subjects), the excretion proved to be much faster in the first stage ( h). The different behaviour of the two groups could be related to the different body burden, of cobalt and/or to the possibility of different kinetics induced by continuous exposure to the metal. Moreover, 3 weeks after the removal of the workers from exposure the urinary cobalt concentrations were not within the normal limits of CoU for the general population, (even for workers exposed to cobalt levels of the same order as the TLV). The increase of CoU concentrations in the first 3 h after the end of exposure, stresses the problem of when urine samples for biological monitoring of the workers should be collected. The present study confirms the utility of CoU in discriminating between exposed and non-exposed subjects as well as in assessing high and low level exposure.  相似文献

Using a sensitive ETA ASS method, urinary thallium (Tl-U) has been measured in a group of non-exposed individuals and in two groups of workers with suspected industrial exposure. In non-exposed subjects (72 healthy males aged 41 +/- 11 years) the Tl-U mean value was 0.22 mu/l (range 0.05-0.61). Significantly higher values were found in two groups of workers (30 and 21 subjects) employed in two cement factories and two cast iron foundries (mean Tl-U value: 0.38 and 0.33 mu/l, respectively; range 0.06-1.2). Thallium has been determined in coal, raw materials and, in one factory, also in environmental samples, but no detectable levels were found. The analytical problems connected to urinary thallium determination and the preliminary results of biological monitoring in non-exposed individuals and in workers are discussed.  相似文献

Exposure to genotoxic compounds in ambient air has been studied in Bangkok, Thailand, by analysis of polycyclic aromatic hydrocarbons (PAHs) associated with particles and using different biomarkers of exposure. Eighty-nine male, non-smoking Royal Thai police officers were investigated. The police officers were divided into a high exposure group (traffic police) and low exposure (office duty). Particulate matter was collected using personal pumps (2 l/min) and the eight carcinogenic PAHs were analysed by standard procedures. The traffic police was exposed to a 20-fold higher level of total PAHs than office police (74.25 ng/m3 vs. 3.11; P= 0.001). A two-fold variation was observed between the different police stations. The major PAHs in all groups was benzo[g,h,l]pyrelene. Large inter-individual differences in biomarker levels were observed, but the level of all markers was statistically significantly higher in the traffic police group than in the office group. The level of 1-hydroxypyrene (1-HOP) was 0.181+/-0.078 (range 0.071-0.393) micromol/mol creatinine in the traffic group and 0.173+/-0.151 (P = 0.044) in the office group. The bulky carcinogen DNA-adduct level, determined by P32-post-labelling, was 1.6+/-0.9 (range 0.4-4.3) adducts/10(8) nucleotides in the traffic group and 1.2+/-1.0 (0.2-4.9) in the office group (P = 0.029; Mann-Whitney U-test). The serum PAH-albumin adduct level was 1.76 (0.51-3.07) fmol adducts/microg albumin in the traffic group and 1.35+/-0.77 (0.11-3.45; P = 0.001) in the office group. Lower biomarker levels were observed during the period when the traffic police officers were wearing a simple facemask, indicating that these masks protect against particle-associated PAHs. No statistically significant correlations were observed between biomarker levels and the level of individual PAHs or total PAH. Our data show, that people in Bangkok, who spend most of the day outside air-conditioned offices, are exposed to high levels of genotoxic PAHs. However, for people who spend their working day in offices, the exposure is similar to people living in other metropolitan areas.  相似文献

Personal exposure to total volatile organic compounds (TVOCs), benzene and toluene of 100 Milan office workers was assessed through personal air monitoring at home, in the office, and during commuting. Biological monitoring was performed by measuring blood benzene and toluene concentrations together with urinary trans-trans-muconic acid (t,t-MA) and cotinine at the end of the monitoring period. The geometric means of the total 24-h personal exposure were 514 micrograms/m3 for TVOCs, 21.2 micrograms/m3 for benzene and 35.2 micrograms/m3 for toluene. Daily exposure to the volatile organic compounds was almost totally determined by indoor exposure at home and in the office, with a minor contribution in the transport means. An important factor determining exposure to benzene was found to be tobacco smoke, both for active smokers and for non-smokers exposed to environmental tobacco smoke (ETS). All the mean levels of the biological indicators were significantly higher in active smokers than in non-smoking subjects non-exposed to ETS; urine cotinine and t,t-MA levels were also significantly higher in non-smokers exposed to ETS than in non-smokers non-exposed to ETS.  相似文献

The contents of the elements antimony, arsenic, cadmium, copper, lead, mercury, selenium, silver, tin and zinc in bone tissue from autopsy specimens of the femur of workers who had been exposed to a large number of metals in a smeltery and refinery in Northern Sweden as well as of a control group have been quantitatively assayed.The analytical techniques used were atomic absorption spectrophotometry, neutron activation analysis and particle induced X-ray emission analysis (PIXE) in a proton microprobe. Increased levels of lead in the bone tissue of exposed workers compared to a non-exposed control group were observed. The median level of lead in the group of exposed workers exceeded the corresponding value of the control group about 5 times.Using the proton microprobe in the PIXE-mode, concentration profiles of copper, lead and zinc were examined within the Haversian system of the bone samples.  相似文献

To evaluate lead exposure among secondary lead-smelting workers with a focus on erythrocyte pyrimidine 5'-nucleotidase (P5N) activity, blood lead concentration (PbB), activity of P5N and other biological variables were examined in 88 exposed workers in five secondary lead smelters and in 24 non-exposed workers in Korea. All of the mean values of air lead concentration (PbA) in the three processes, scrap pretreatment, blast furnace smelting, and refining and casting of the secondary lead smelters, markedly exceeded 0.05 mg/m3. In this survey, 29 (97%) of 30 air samples for lead exceeded 0.05 mg/m3. The highest mean PbA and PbB values were found in the section of blast furnace smelting. All of the mean PbB values in all the sections were higher than 30 microg/dl. PbB of 71 (81%) of the 88 exposed workers exceeded 30 microg/dl. In 31 (35%) of the exposed workers, PbB was above 60 microg/dl. Compared with the non-exposed group, zinc protoporphyrin (ZPP) in the exposed group was significantly increased, whereas erythrocyte P5N activity and activity of erythrocyte delta-aminolevulinic acid dehydratase (ALAD) were significantly inhibited. Erythrocyte P5N activity had valid biological correlation with PbB and with other biological variables, such as ALAD activity or ZPP. Lead exposure affected hemoglobin levels via inhibition of P5N activity, as well as the heme biosynthetic pathway, in the high-exposure state.  相似文献

A method for the rapid and sensitive analysis of 1-hydroxypyrene in human urine was improved by high performance liquid chromatography (HPLC) with fluorescence detection. The limit of detection decreased approximately threefold when triethylamine was added to the extracts prior to the chromatographic determination. The sensitivity was increased mainly due to triethylamine blockage of the secondary-retention effects. The analysis time was approximately 20 min per run. This method was applied to the analysis of 1-hydroxypyrene in 125 urine specimens collected from cigarette smokers who had non-occupational exposure to polycyclic aromatic hydrocarbons. The average concentrations of urinary 1-hydroxypyrene were 0.04 micromol/mol creatinine in non-smokers, 0.20 micromol/mol creatinine in light smokers, 0.46 micromol/mol creatinine in medium smokers, and 1.16 micromol/mol creatinine in heavy cigarette smokers. The levels of urinary 1-hydroxypyrene correlated strongly with cigarette intake (r2 = 0.93, P < 0.001).  相似文献

The aim of the study was to determine if occupational exposure to hydrofluorocarbons (HFC) and hydrochlorofluorocarbons (HCFC) generates autoimmune responses against CYP2E1. HFCs and HCFCs have replaced the chlorofluorocarbons (CFC) in e.g. refrigeration installations and air-conditioning systems. During the substitution period, refrigeration mechanics reported symptoms like asthma, influenza-like reactions, and joint troubles. These symptoms resemble those of chronic inflammatory diseases with an autoimmune component. Since exposure to structurally similar chemicals, e.g. halothane, has previously been associated with autoimmune responses and diseases, autoimmunity among the refrigeration mechanics might hypothetically explain the reported inflammatory symptoms. Serum from 44 Swedish men, occupationally exposed to halogenated hydrocarbons, was screened for antibodies against CYP2E1 with enzyme-linked immunosorbent assay. Thirty of the workers had asthma, joint problems or influenza-like symptoms whereas 14 of them had no such symptoms. They were all selected from a cohort of 280 refrigeration mechanics. Unexposed, healthy, Swedish men (n=35) constituted control group. The study was approved by the Ethics Committee at Karolinska Institutet. No increase in autoantibodies against CYP2E1 was detected among the occupationally exposed workers as compared to the unexposed controls. Further, there was no difference in antibody titer between the exposed workers with symptoms and the exposed, asymtomatic workers or the unexposed controls. The present study does not completely exclude a connection between exposure and effect but makes the relation less likely at these exposure levels.  相似文献

Extremely Low Frequency-Magnetic Fields (ELF-MF) are possible carcinogens to humans and some data suggest that they can act as promoters or progressors. Since NK cells play a major role in the control of cancer development, an adverse effect on ELF-MF on NK function has been hypothesized. We examined NK activity in 52 workers exposed to different levels of ELF-MF in various activities. Individual exposure was monitored during 3 complete work-shifts using personal dosimeters. Environmental exposure was also monitored. ELF-MF levels in the workers were expressed as Time-Weighted Average (TWA) values. NK activity was measured in peripheral blood lymphocytes (PBL). In the whole group the median occupational TWA was 0.21 microT. According to the TWA levels, workers were classified as low exposed (26 subjects, TWA < or =0.2 microT) and higher exposed workers (26 subjects; TWA >0.2 microT). In higher exposed workers, we observed a trend to reduce NK activity compared to low exposed, but the difference was not significant. Then we selected a subgroup of highest exposed workers (12 subjects; TWA >1 microT); no difference was observed between low and highest exposed subjects in the main personal variables. Considering both E:T ratios from 12:1 to 50:1 and Lytic Units, a significant reduction in NK activity was observed in the highest exposed workers compared to the low exposed. Multivariate analysis showed a significant negative correlation between exposure and LU, while no correlation was evidenced with other personal characteristics. ELF-MF are considered possible carcinogens, and existing data suggest that they can act as promoters. Due to the role of NK activity in host defence against cancer, the results obtained in this study in workers exposed to ELF-MF levels exceeding 1 microT are in agreement with this hypothesis, and support the need for further investigation in this field.  相似文献

Mercury [Hg] emissions from old plant collections treated with mercuric chloride (HgCl(2)) may present a high health risk for staff working in certain herbaria. The present study evaluated Hg concentrations in ambient air, plant specimens and biological samples from staff working in the Pretoria National Herbarium (PRE) and the H.G.W.J. Schweickerdt Herbarium (PRU), University of Pretoria. Biological samples from a group of 15 people exposed to HgCl(2) in herbaria and a non-exposed control group of five people were studied. Additionally, plant samples from herbarium specimens treated and non-treated with HgCl(2) were analysed. Plant materials treated with HgCl(2) had persistent high concentrations of Hg in the range of 114-432 microg g(-1), whereas untreated materials were in the range of 0.20-0.45 microg g(-1). The HgCl(2)-treated plant specimens induced elevated concentrations of Hg into the herbarium rooms near storage cabinets, where up to 1 microg m(-3) of Hg was measured in the air of both herbaria. However, no significant difference in mean Hg concentrations in hair was found between herbarium workers and members of the control group, 0.46 and 0.64 microg g(-1) respectively (p0.05, Student's t-test). For both groups, Hg concentrations were lower than that indicated by the World Health Organization [WHO] for non-exposed adults, namely 2 microg g(-1). The mean concentration of total Hg in urine from the mercury-exposed herbarium group, 2.28 microg g(-1) creatinine, was significantly higher than in the control group, 1.05 microg g(-1) of creatinine. For both populations, the concentrations of Hg in their urine were below the threshold Hg values set by the WHO, i.e., 5 microg g(-1) creatinine. We concluded that there was no strong response by individual herbarium staff from long-term exposure to Hg concentrations in the range of 0.28-1.1 microg m(-3).  相似文献
