原发性高血压与血管内皮细胞损伤的研究   总被引:1,自引:0,他引:1  
目的 探讨原发性高血压 (EH )与血管内皮细胞损伤的关系。方法 对 5 0例不同临床分期EH患者 (EH组 )及2 5例体检健康的正常人 (对照组 )分别检测血浆内皮素 (ET)、血清一氧化氮 (NO)、血浆血管性血友病因子 (vWF)浓度、血清丙二醛 (MDA)含量 ,以及超氧化物歧化酶 (SOD)活性。结果 EH组ET、MDA、vWF浓度水平明显高于对照组 (P均 <0 .0 1) ;NO、SOD活性水平明显低于对照组 (P均 <0 .0 1)。结论 EH时内皮细胞损伤 ,NO和ET的平衡受到破坏 ,NO和ET及氧化应激参与EH的发生、发展。vWF可作为EH患者血管内皮细胞功能损伤程度的特异性敏感指标。  相似文献

目的 探讨肺动脉内皮细胞源性舒缩因子 (Endotheliumderivedrelaxingandconstrict ingfactor )在高原肺水肿 (HAPE )肺动脉高压发生中的作用。方法 采用右心漂浮导管检测法 ,对 8例高原肺水肿患者及 8例同海拔高度高原健康人的肺动脉压力进行了检测 ,同时也采用放免分析方法检测了HAPE患者肺动脉内皮细胞分泌到血液中的收缩因子ET - 1、TXA2 及舒张因子NO、PGI2 变化。结果 HAPE患者发病时 ,肺动脉平均压明显高于同海拔高度健康人 ,而血PaO2 、SaO2 明显低于对照组 ;HAPE患者外周血ET - 1、TXA2 含量明显高于对照组 ,而血中NO、PGI2 水平明显低于对照组 ;患者肺动脉平均压同ET - 1 /NO及TXA2 /PGI2 比值呈显著正相关。结论 肺动脉内皮细胞分泌的内皮源性舒缩因子失衡在高原肺水肿的缺氧性肺动脉高压发生中起重要作用  相似文献

目的 通过观察烧伤痂下水肿液 (STF)对人脐静脉内皮细胞合成分泌NO和ET的影响 ,以进一步阐明STF在烧伤早期内皮损伤中的作用。方法 将人脐静脉内皮细胞与分别含有 2 0 %体积的STF、烧伤患者血清 (BPS)和健康人血清 (HHS)的RPMI 16 40培养液共同培养 2 4h后 ,检测培养上清液中NO和ET含量及ET/NO。结果 STF组、BPS组、HHS血清组的NO值分别为 92 .78± 10 .49,90 .96± 19.5 8,5 0 .43± 9.13;ET值分别为 2 6 2 .2 5± 2 8.46 ,16 8.83± 11.30 ,5 8.37± 11.47;ET/NO值分别为 2 .82± 0 .42 ,1.85± 0 .31,1.15± 0 .2 6。STF组、BPS组的NO ,ET ,ET/NO值均显著高于HHS组 (P <0 .0 1) ;STF组的ET和ET/NO值显著高于BPS组 (P <0 .0 1) ;STF组与BPS组的NO值差异无显著性 (P >0 0 5 )。结论 STF可通过损伤内皮细胞而参与烧伤早期微循环障碍 ,其对内皮细胞的损伤作用强于烧伤血清。  相似文献

目的 观察不同配比的丹酚酸B(SalB)、丹参酮ⅡA(TanⅡA)对体外肿瘤坏死因子α(TNF—α)损伤大鼠心脏微血管内皮细胞(CMEC)的影响。方法 采用CMEC体外培养技术,建立TNF—α损伤模型,以细胞活力,细胞培养液中乳酸脱氢酶(LDH)释放、一氧化氮(NO)合成、内皮素(ET)分泌、超氧化物歧化酶(SOD)活性及丙二醛(MDA)含量的变化为评价指标,观察SalB和TanⅡA不同配比(10:0,8:2,5:5,2:8,0:10)对TNF-α损伤CMEC的保护作用。结果 SalB和TanⅡA(5:5)组明显提高细胞活力,SalB和TanⅡA(8:2)组显著降低LDH释放水平,TanⅡA促进NO合成、降低ET分泌以及抗氧化的作用最好。结论 SalB和TanⅡA的各配比组均能明显改善细胞的损伤,最佳配比根据检测指标的不同而不同。  相似文献

目的 :观察血管内皮生长因子 (VEGF)对兔离体主动脉内皮细胞分泌功能的影响 ,了解一氧化氮 (NO)在其中所起的作用 ,以明确VEGF的作用机制。方法 :先进行兔主动脉内皮细胞的培养和鉴定 ,然后行传代培养。在第 3~ 5代主动脉内皮细胞培养液中加入不同浓度的VEGF ,培养 6h ,取上清液测定内皮素 (ET)、血管性血友病因子 (vWF)值 ,并应用一氧化氮合成酶 (NOS)抑制剂N -硝基 -L -精氨酸甲酯 (L -NAME)后观察ET、vWF值的变化情况。结果 :VEGF可抑制血管内皮细胞 (VEC)分泌ET、vWF ,并且呈剂量依赖性 (r =- 0 .96 881P <0 .0 1,r=- 0 .9711P <0 .0 1) ,NOS抑制剂L -NAME可剂量依赖性地阻断其效应 (P <0 .0 1)。结论 :NO在VEGF抑制VEC分泌ET、vWF中起中介作用 ,NO是VEGF作用机制中的一个重要通路。  相似文献

目的:研究薯蓣皂苷对氧化低密度脂蛋白(Ox-LDL)诱导血管内皮细胞(EC)凋亡的保护作用,并探讨其作用机制.方法:实验分5组,对照组、模型组(150μg/mL的Ox-LDL)、薯蓣皂苷低中高剂量组(4.8、12、30μg/mL),用MTT法检测EC存活率;流式细胞术分析测定细胞凋亡率;硝酸还原酶法测定EC的NO浓度;双抗体夹心法检测内皮素(ET)浓度;Western Blot测定内皮细胞eNOS的蛋白表达.结果:与对照组相比,Ox-LDL使EC的凋亡率、ET的浓度显著升高(49.6±10.44vs 30.0±3.27,P<0.01;25.89±2.07vs16.93±1.64,P<0.05);使EC的存活率、NO的浓度、eNOS蛋白表达显著降低(64.89±1.02vs 100±0,P<0.01;27.03±3.12vs59.46±16.19,P<0.01;P<0.01).薯蓣皂苷低中高剂量组能明显抑制EC的凋亡率,降低ET的浓度;提高EC的存活率、NO的浓度以及eNOS蛋白表达水平,且在此药物浓度范围内成浓度依赖性.结论:著蓣皂苷可能通过减少ET的合成与分泌,提高eNOS的表达,使NO的合成与释放增加,从而对Ox-LDL诱导血管内皮细胞凋亡起到保护作用.  相似文献

目的:分析冠状动脉粥样硬化性心脏病(简称冠心病)痰热互结证患者外周血长链非编码RNA牛磺酸上调基因1(long noncoding RNA taurine up regulated gene 1,LncRNA TUG1)LncRNA TUG1与血管内皮损伤的相关性。方法:随机选取20例冠心病痰热互结证患者,另设健康对照组20例。所有入选对象取血6 mL,Real-Time PCR法检测外周血单个核细胞中LncRNA TUG1,免疫学方法测定循环内皮细胞计数,硝酸还原酶法测定NO分泌水平,放射免疫法测定内皮素分泌水平。结果:冠心病痰热互结证患者LncRNA TUG1相对表达量增加,循环内皮细胞计数增加,内皮素(endothelin,ET)分泌水平上调,NO分泌水平降低,差异有统计学意义(P0.05)。LncRNA TUG1与循环内皮细胞计数(r=0.48,P=0.00)呈显著正相关,与NO分泌(r=-0.58,P=0.00)呈显著负相关,同时与ET分泌呈显著正相关(r=0.37,P=0.02)。结论:冠心病痰热互结证患者外周血LncRNA TUG1表达具有差异性,差异表达的LncRNA TUG1与血管内皮损伤存在一定相关性。  相似文献

目的:研究刺五加叶皂苷(ASS)对糖基化终产物(AGE)作用下人脐静脉内皮细胞(HUVECs)的保护作用及其机制。方法:  体外培养HUVECs,糖孵育法制备糖基化终产物修饰牛血清白蛋白(AGE-BSA)。将细胞分为6组,即正常对照组、实验对照组(BSA组)、损伤组(AGE组)、损伤加入ASS低、中、高浓度组。将200 mg·L-1 AGE作用于不同浓度(10、50和100 mg·L-1)ASS培养4 h的HUVECs,继续培养24 h,测定各组细胞增殖活力及培养液中血管性假血友病因子(vWF)、一氧化氮(NO)及乳酸脱氢酶(LDH)的含量。结果:与BSA组比较, AGE组HUVECs增殖活力明显降低(P<0.01),AGE组HUVECs培养液vWF和LDH含量明显增加(P<0.01),NO水平明显降低(P<0.01)。与AGE组比较,10、50和100 mg·L-1 ASS组HUVEC增殖活力明显增高(P<0.05),vWF 水平显著减少(P<0.05),LDH含量明显减少(P<0.05),NO生成量明显增多(P<0.05)。结论: ASS可抑制AGE对内皮细胞的损伤作用,且可抑制AGE减少NO生成的作用,ASS保护内皮细胞免受AGE损伤的机制可能与增加内皮细胞NO生成量有关。  相似文献

目的 研究胆固醇对人血管内皮细胞的损伤作用.方法 体外培养人脐静脉内皮细胞株ECV304,分别以浓度为6.25、12.50、25.00、50.00 mg/L的胆固醇作用于ECV304细胞48 h,检测细胞培养液中乳酸脱氢酶(lactate dehydrogenase, LDH)活性,一氧化氮(nitric oxide, NO)浓度,一氧化氮合酶(nitric oxide synthase, NOS)活性及单核细胞趋化蛋白-1(MCP-1)浓度.结果 与无胆固醇组比较,胆固醇浓度为50.00 mg/L时培养液中LDH活性显著增加;25.0 mg/L,50.0 mg/L浓度组培养液中NO浓度显著降低;胆固醇浓度为6.25、12.5、25.0、50.0 mg/L组与无胆固醇组比较,随着胆固醇浓度提高,培养液中NOS活性逐渐降低,MCP-1浓度逐渐增高,有显著性差异.结论 一定浓度的胆固醇能直接损伤人血管内皮细胞结构,影响内皮细胞的部分分泌功能.  相似文献

【目的】探讨蕨麻正丁醇部位对内皮细胞缺氧损伤的保护作用。【方法】采用人脐静脉内皮细胞株(EAHY926)建立缺氧损伤实验模型,通过MTT法测定各实验组细胞代谢率;比色法测定乳酸脱氢酶(lactate dehydrogenase,LDH)、细胞内超氧化物歧化酶(superoxide dismutase,SOD)活性和NO浓度;放射免疫法测定内皮素-1(endothelin-1,ET-1)的活性。【结果】3.00 mg/ml、1.50 mg/ml和0.75 mg/ml蕨麻正丁醇部位均能显著减少缺氧损伤内皮细胞LDH的外漏量,并可显著提高细胞内SOD活性,增加NO分泌、减少ET-1的生成。【结论】蕨麻正丁醇部位对内皮细胞缺氧损伤具有显著的保护作用,其机制之一可能是清除缺氧导致的内皮细胞自由基堆积,减少ET-1的释放,增加NO分泌,从而减轻内皮细胞损伤。  相似文献

Objective: To evaluatel the value of D-dimers in patients with acute aortic dissection (AAD). Methods: This study consisted of 16 patients with AAD and 27 non-AAD patients. Serum D-dimets were measured by Sta-Liatest D-DI immunoturbidimetric assay. Results: D-dimer level was higher (P < 0.001) in patients with AAD(7.91 ± 5.52 μg/ml) than that in non- AAD group(1.57±1.24 μg/ml). D-dimer was positive (>0.4 μg/ml) in all patients with AAD and in 10 control group patients (37%). Among patients with acute AAD, D-dimers tended to be higher in Stanford A than in Stanford B (8.67 ± 4.31 μg/ml vs. 3.24±1.27 μg/ml, P <0.01). D-dimer values tended to be higher in more extended disease(3.84 ± 1.65 μg/ml, 8.57 ± 3.58 μg/ml and 11.87 ± 5.69 μg/ml in thoracic aorta, thoracic and abdominal aorta, thoracic and abdominal aorta and iliacal arteries, respectively, P < 0.05 for both 8.57 ± 3.58 and 11.87 ± 5.69 vs. 3.84 ± 1.65 ). Including the control group into the analysis, we found a sensitivity of 100%, a negative predictive value of 100%, and a specificity of 66% and a positive predictive value of 64% for D-dimer in diagnosis of AAD in our patients with suspected AAD. Conclusion: D-dimer was elevated in patients with AAD. A negative D-dimer test result could be useful in excluding AAD.  相似文献

Objective: To set up a simple and reliable rat model of combined liver-kidney transplantation. Methods: SD rats served as both donors and recipients. 4℃ sodium lactate Ringer's was infused from portal veins to donated livers,and from abdominal aorta to donated kidneys, respectively. Anastomosis of the portal vein and the inferior vena cava (IVC) inferior to the right kidney between the graft and the recipient was performed by a double cuff method, then the superior hepatic vena cava with suture. A patch of donated renal artery was anastomosed to the recipient abdominal aorta. The urethra and bile duct were reconstructed with a simple inside bracket. Results: Among 65 cases of combined liver-kidney transplantation, the success rate in the late 40 cases was 77.5%. The function of the grafted liver and kidney remained normal. Conclusion: This rat model of combined liver-kidney transplantation can be established in common laboratory conditions with high success rate and meet the needs of renal transplantation experiment.  相似文献

Objective To observe blood pressure change with age in salt-sensitive teenagers whose salt sensitivity were determined by repeated testing.Methods Salt sensitivity was determined through intravenous infusion of normal saline combined with volume-depletion by oral diuretic furosemide in 55 teenagers. After five years, salt sensitivity was re-examined and subject blood pressure was followed up. Blood pressure changes in salt-sensitive teenagers were compared to that of non-salt sensitive teenagers over five years.Results After 5 years, the repetition rate of salt sensitivity determined by intravenous saline loading is 92.7%. In teenagers with salt sensitivity on the baseline, both the systolic blood pressure increments and increment rates were much higher than non-salt sensitive teenagers (12.7±12.1 mmHg vs. 2.8±5.2 mmHg, P< 0.01; 12.2%± 12.0% vs. 2.5% ±4.4%, P< 0.001,respectively). There was a similar trend for diastolic blood pressure (8.4 ± 6.4 mmHg vs. 3.7 ± 6.4 mmHg, P = 0.052; 13.2% ±10.6 % vs. 6.8%± 10.1%, P = 0.053, respectively).Conclusions Salt sensitivity determined by intravenous saline loading showed good reproducibility. Blood pressure increments with age were much higher in salt-sensitive teenagers than non-salt sensitive teenagers, especially in terms of systolic blood pressure.  相似文献

Shock wave lithotripsy (SWL) is a treatment of choice for upper urinary stones. However, this procedure is inappropriate for obese patients because the focus is often unable to reach the target owing to the limited focal distance in shock wave source. Although treating such patients in a blast path may increase the application length of shock wave source, it's difficult to find this path on the lithotripter monitor. Excess production of reactive oxygen species(ROS)of mitochondrion mediated by hyperglycemia is the common pathogenesis of angiopathic complications of diabetes.TCM holds that the damp from the dysfunction of spleen.kidney and liver is the causative factor of complications of diabetes.This is similar to the mechanism of Ros resulting in angiopathic complications of diabetes.When the angiopathic complications of type II diabetes mellitus(T2DM)are difierentiated as caused by turbid damp in TCM can be explained as ROS.Since the obstruction of pathogenic damp in channels and collaterals is said to be the main pathogenesis,the treating principle should be dissolving the damp to remove the obstruction.  相似文献

Objective: To observe the therapeutic effects in acupunture treatment of primary dysmenorrhea combined with spinal Tui Na, and study its mechanism. Methods: Thirty cases of the treatment group were treated by acupuncture combined with spinal Tui Na, and thirty cases in the control group were treated by routine acupuncture. Results: The total effective rate was 93.3% in the treatment group, and 73.3% in the control group, with a significant difference between the two groups (P<0.05). Conclusions: Acupuncture combined with spinal Tui Na has good prospects for treatment of primary dysmenorrhea.  相似文献
