This paper describes an investigation of the uptake of Cu(II) by poly(amidoamine) (PAMAM) dendrimers with an ethylenediamine (EDA) core in aqueous solutions. We use bench scale measurements of proton and metal ion binding to assess the effects of (i) metal ion-dendrimer loading, (ii) dendrimer generation/terminal group chemistry, and (iii) solution pH on the extent of binding of Cu(II) in aqueous solutions of EDA core PAMAM dendrimers with primary amine, succinamic acid, glycidol, and acetamide terminal groups. We employ extended X-ray absorption fine structure (EXAFS) spectroscopy to probe the structures of Cu(II) complexes with Gx-NH2 EDA core PAMAM dendrimers in aqueous solutions at pH 7.0. The overall results of the proton and metal ion binding measurements suggest that the uptake of Cu(II) by EDA core PAMAM dendrimers involves both the dendrimer tertiary amine and terminal groups. However, the extents of protonation of these groups control the ability of the dendrimers to bind Cu(II). Analysis of the EXAFS spectra suggests that Cu(II) forms octahedral complexes involving the tertiary amine groups of Gx-NH2 EDA core PAMAM dendrimers at pH 7.0. The central Cu(II) metal ion of each of these complexes appears to be coordinated to 2-4 dendrimer tertiary amine groups located in the equatorial plane and 2 axial water molecules. Finally, we combine the results of our experiments with literature data to formulate and evaluate a phenomenological model of Cu(II) uptake by Gx-NH2 PAMAM dendrimers in aqueous solutions. At low metal ion-dendrimer loadings, the model provides a good fit of the measured extent of binding of Cu(II) in aqueous solutions of G4-NH2 and G5-NH2 PAMAM dendrimers at pH 7.0.  相似文献

The multiple application of reductive amination on primary amino groups of first and second generation poly(propyleneimine) dendrimers is used as a one-pot approach to introduce twice the amount of the oligosaccharide units as surface groups, compared to initially present amino groups in the first and second generation dendrimers. This was proven by (1)H NMR, MALDI-TOF-MS, and LILBID-MS analysis. The size of these dendrimers was determined by the hydrodynamic radius using pulsed field gradient NMR and dynamic light scattering. Molecular modeling confirmed the presence of dense-shell dendrimers. These dendrimers exhibit a generation dependent Cu(II)/dendrimer ratio in an aqueous environment, highlighting these materials as possible metal-carrier systems with a well-defined oligosaccharide protection shell for application in a biological environment.  相似文献

The first four generations of cobaltocenium-functionalized, diaminobutane-based poly(propylene imine) dendrimers DAB-dend-Cb,(PFb)x (x = 4, 8, 16, and 32; Cb=[Co(eta5-C5H4CONH)(eta5-C5H5)] (1-4) have been synthesized and characterized. The redox activity of the cobaltocenium centers in 1-4 has been characterized by using cyclic voltammetry and the electrochemical quartz-crystal microbalance (EQCM). All of the dendrimers exhibit reversible redox chemistry associated with the cobaltocenium/cobaltocene redox couple. Upon reduction. the dendrimers exhibit a tendency to electrodeposit onto the electrode surface, which is more pronounced for the higher generations. Pt and glassy carbon electrodes could be modified with films derived from 1-4,exhibiting a well-defined and persistent electrochemical response. EQCM measurements show that the dendrimers adsorb, at open circuit, onto platinum surfaces at monolayer or submonolayer coverage. Cathodic potential scanning past -0.75 V at which the cobaltocenium sites are reduced, gave rise to the electrodeposition of multilayer equivalents of the dendrimers. The additional material gradually desorbs upon re-oxidation so that only a monolayer equivalent remains on the electrode surface. Changes in film morphology as a function of dendrimer generation and surface coverage were studied by using admittance measurements of the quartz-crystal resonator on the basis of its electrical equivalent circuit, especially in terms of its resistance parameter. In general, we find that films of the lower dendrimer generation 1 behave rigidly, whereas those of the higher generation 4 exhibit viscoelastic behavior with an intermediate behavior being exhibited by 2 and 3. Using tapping-mode atomic force microscopy (AFM). we have been able to obtain molecularly resolved images of dendrimer 4 adsorbed on a Pt(111) electrode.  相似文献

We study the effect of the solvent (water) on the ligand exchange reaction (LER) step of Pt(II) complexation to PAMAM dendrimers. The results suggest that aquation of tetrachloroplatinate anion (PtCl(4)(2-)) inside PAMAM outer pockets occurs prior to its reaction with dendrimer atom binding sites. Thus, the active involvement of water opens up several pathways by which Pt(II) can bind to tertiary amine sites (N3). Monodentate binding pathways by which a PtCl(3)(-) moiety is obtained as a final product rather than PtCl(2)(H(2)O) are considered to be the predominant routes due to their smaller degree of complexity, including aspects such as less number of intermediates and lower energy barriers. Monodentate binding of Pt(II) to the secondary amide site (N2) is found to be feasible, in agreement with previous NMR experiments, once aquation of the tetrachloroplatinate anion has occurred. For this type of binding to occur, the dendrimer branch amide group configuration would have to switch from its equilibrium position (trans) to a cis position. It is also found that outer pockets aid Pt(II) complexation with the dendrimer mainly by making the noncovalent binding (NCB) step more favorable than that in branchless environments. Finally, our results predict that competitive monodentate binding of Pt(II) to either N3 or N2 is thermodynamically rather than kinetically driven.  相似文献

Dendrimer-Au nanocomposites are prepared in aqueous solutions using poly(amidoammine)dendrimers (PAMAM) (generation 2, 3, and 5) and poly(propyleneimine)dendrimers (PPI)(generation 2, 3, and 4) by wet chemical NaBH(4) method. The Au nanoparticles thus obtained are 2-4 nm in diameter for both dendrimers and no generation dependence on the particle size is observed, whereas the generations of the dendrimers are increased as stabilization of Au-nanoparticles is achieved with lower dendrimer concentrations. Studies of the reduction reaction of 4-nitrophenol using these nanocomposites show that the rate constants for the PAMAM dendrimers (generations 2 and 3) are higher than those for the PPI dendrimers (generations 2 and 3), while a distinct difference in the rate constants is not seen for the PAMAM dendrimer (generation 5) or the PPI dendrimer (generation 4). In addition, the rate constants for the reduction of 4-nitrophenol involving all the dendrimers decrease with increases in dendrimer concentrations.  相似文献

The poly(amidoamine) dendrimers having terminal isobutyramide (IBAM) groups were prepared by the reaction of isobutyric acid and the amine-terminated poly(amidoamine) dendrimers with generations (G) of 2 to 5 by using a condensing agent, 1,3-dicyclohexylcarbodiimide. 1H and 13C NMR revealed that an IBAM group was attached to essentially every chain end of the dendrimers. While the IBAM-terminated G2 dendrimer was soluble in water, the IBAM-terminated G3, G4, and G5 dendrimers exhibited the lower critical solution temperatures (LCSTs) at 75, 61, and 43 degrees C, respectively. Because the density of the terminal IBAM groups in the periphery of the dendrimer progressively increases with increasing dendrimer generation, the interaction of the IBAM groups might take place more efficiently, resulting in a remarkable decrease in the LCST. In addition, attachment of IBAM groups to poly(propylenimine) dendrimers could give the temperature-sensitive property, indicating that this is an efficient method to render dendrimers temperature sensitive.  相似文献

Abstract

Poly(propylene imine) dendrimers from first and third generation modified with 1,8-naphthalimide units and their Cu(II) complexes have been characterized by fluorescence and EPR spectroscopy. Cotton fabric has been modified with these dendrimers and their color characteristics were determined. The antimicrobial activity of dendrimer ligands and their Cu(II) complexes in solution and after their deposition on a cotton fabric was investigated. Good antibacterial effect of dendrimer ligands has been obtained, which is enhanced at their Cu(II) complexes. After their deposition on cotton fabric metallodendrimers exhibit good antibiofilm activity.  相似文献

Pt/gamma-Al2O3 catalysts were prepared using hydroxyl-terminated generation four (G4OH) PAMAM dendrimers as the templating agents and the various steps of the preparation process were monitored by extended X-ray absorption fine structure (EXAFS) spectroscopy. The EXAFS results indicate that, upon hydrolysis, chlorine ligands in the H(2)PtCl(6) and K(2)PtCl(4) precursors were partially replaced by aquo ligands to form [PtCl3(H2O)3]+ and [PtCl2(H2O)2] species, respectively. The results further suggest that, after interaction of such species with the dendrimer molecules, chlorine ligands from the first coordination shell of Pt were replaced by nitrogen atoms from the dendrimer interior, indicating that complexation took place. This process was accompanied by a substantial transfer of electron density from the dendrimer to platinum, indicating that the dendrimer plays the role of a ligand. Following treatment of the H(2)PtCl(6)/G4OH and K(2)PtCl(4)/G4OH complexes with NaBH4, no substantial changes were observed in the electronic or coordination environment of platinum, indicating that metal nanoparticles were not formed during this step under our experimental conditions. However, when the reduction treatment was performed with H2, the formation of extremely small platinum clusters, incorporating no more than four Pt atoms was observed. The nuclearity of these clusters depends on the length of the hydrogen treatment. These Pt species remained strongly bonded to the dendrimer. Formation of larger platinum nanoparticles, with an average diameter of approximately 10 A, was finally observed after the deposition and drying of the H(2)PtCl(6)/G4OH nanocomposites on a gamma-Al(2)O(3) surface, suggesting that the formation of such nanoparticles may be related to the collapse of the dendrimer structure. The platinum nanoparticles formed appear to have high mobility because subsequent thermal treatment in O2/H2, used to remove the dendrimer component, led to further sintering.  相似文献

Gd(III)-containing dendrimers are promising contrast agents for magnetic resonance imaging (MRI). An important issue in the effectiveness and toxicity of a Gd(III) based MRI contrast agent is knowledge of the relative locations and concentrations of Gd(III) in dendrimer drug delivery hosts. In order to provide experimental information on this issue, we have investigated the electron paramagnetic resonance (EPR) of a stable Gd(III) complex with diethylenetriaminepentaacetic acid (DTPA) in various polyammidoamine (PAMAM) dendrimers as a function of dendrimer generation (G2, G4, and G6), dendrimer core (ethylenediamine = EDA, and cystamine = cys), and dendrimer surface functionality (NH(2), 5-oxo-3-pyrrolidinecarboxylic acid methyl ester = pyr, and tris(hydroxymethyl) methylamine = tris). The dendrimer systems were investigated in the presence and absence of paramagnetic probes, that is, Cu(II) and nitroxide radicals (4-(trimethylammonium and dodecyl-dimethylammonium) 2,2,6,6-tetramethylpiperidine 1-oxyl bromide = CAT1 and CAT12, respectively). The analysis of the EPR spectra revealed anisotropic locations of Gd-DTPA inside the dendrimer. Computer analysis of the EPR spectra of the probes identified the interactions of the Gd-dendrimers with ions and organic molecules. The interaction between the probes and the dendrimer internal and external surface depends on the type of core, the composition of the external surface and the generation of the dendrimer. The negatively charged Gd-DTPA complex attracts the positively charged species and this provokes spin-spin interactions between Gd and the probes, which increases with a decrease in generation, mainly from G6 to G4, and with an increase in both the Gd-dendrimer concentration and the probe concentration. The cys core increases the internal volume and decreases the packing of the branches.  相似文献

A new class of pi-conjugated dendrimers G0, G1, and G2 was developed through a double-stage divergent/convergent growth approach, in which 5,5,10,10,15,15-hexahexyltruxene was employed as the node and oligo(thienylethynylene)s (OTEs) with different lengths as the branching moieties. The dendrimers were fully characterized by (1)H and (13)C NMR, elemental analysis, gel permeation chromatography, and MALDI-TOF MS. Also, by using atomic force microscopy, it was observed that dendrimer G2 laid nearly flat on the mica surface as a single molecule. Dynamic light scattering results showed that the molecule retained its relatively flat shape in solution. To our best knowledge, dendrimer G2, with a radius approaching 10 nm and a molecular weight of 27 072 Da, was the largest among reported second generation dendrimers. The energy gradient in G2 was constructed by linking OTEs of increasing effective conjugation lengths from the dendritic rim to the core. The intramolecular energy transfer process was studied using steady-state UV-vis absorption and photoluminescent spectroscopies, as well as time-resolved fluorescence spectroscopy. Our structurally extended dendrimers showed an excellent energy funneling ability (their energy transfer efficiencies were all over 95%). All results demonstrate that these dendrimers are promising candidates as light-harvesting materials for optoelectronic devices.  相似文献

6FDA-polyimide films modified by polyamidoamine (PAMAM) dendrimers with generations of 0, 1, and 2 are reported in this article. The actual molecular conformation and bulk size of these three generation dendrimers immobilized on polyimide surface were characterized by atomic force microscopy. After comparing with the results of dynamic simulation, we believe that the disk-shape cluster structure of dendrimers has been developed on the polymer surfaces. The amidation and cross-linking reaction between dendrimers and polyimide were examined and quantified by X-ray photoelectron spectroscopy, attenuated total reflection Fourier transform infrared spectroscopy, and gel content measurements. Modification time and the generations of PAMAM dendrimer have been verified as two important factors in determining the properties of modified polyimide films. These modified polyimide films exhibit excellent gas separation performance. The ideal selectivity of He/N(2) increases tremendously to about 200% as compared to that of the original polyimide film. Particularly, the separation performance of CO(2)/CH(4) gas pair can be improved beyond the upper bond limit possibly due to the strong interactions of dendrimer molecules with CO(2), which was verified by sorption tests.  相似文献

Terminal amine groups of poly(amidoamine) (PAMAM) dendrimers can be substituted with different functional groups for various applications. In this study, PAMAM derivatives with acetamide, hydroxyl, and carboxyl termini were synthesized from ethylenediamine (EDA) core generation 4 and 5 primary amine-terminated PAMAM dendrimers. The reaction products were purified with dialysis and subsequently characterized by polyacrylamide gel electrophoresis (PAGE), capillary electrophoresis (CE), size exclusion chromatography (SEC), matrix-assisted laser desorption ionization time-of-flight (MALDI-TOF) mass spectrometry, potentiometric titration, 1H NMR, and 13C NMR. PAGE and CE electropherograms provide data regarding the purity, charge distribution, and electrophoretic mobility of the dendrimers and their derivatives. SEC and MALDI-TOF mass spectrometry detect the average absolute molar mass and the individual mass fractions, respectively. The combination of SEC with potentiometric titration provides quantitative evidence of the degree of the functional group substitution, while NMR techniques (both 1H NMR and 13C NMR) confirmed the changes in dendrimer surface functionalization. This study provides a general example for the comprehensive characterization of surface-functionalized PAMAM dendrimer nanoparticles. The synthesized dendrimer derivatives hold promise for environmental and medical applications.  相似文献

Amphiphilic polyester-co-polyether (PEPE) dendrimers synthesized from poly(ethylene glycol) (PEG) were examined to understand the influence of alterations in the architecture of dendrimers on their conformation at interfaces and distribution of various groups on their surface. Effect of changes in the number of branching points, type of terminal functional groups and generation of dendrimer was primarily evaluated. Dendrimers were deposited on mica by spin coating at 0.1 mg/mL. Tapping mode atomic force microscopy (AFM) was employed for the visualization of dendrimer topographies while, X-ray photoelectron spectroscopy (XPS), AFM phase and force imaging were used as the tools for characterization of their surfaces. Individual dendrimer molecules could be imaged by AFM, which showed that they are round or oval in topography. Dendrimers were also flattened on mica but the extent of flattening differed with the chemical structure; for instance, third generation dendrimers were more flattened than second generation dendrimers whereas, dendrimers with higher number of branches had greater height above the mica surface. Hydrophilic and hydrophobic groups present towards the aerial interface existed in distinct zones rather than being distributed randomly, except in dendrimer with higher number of branches. The percentage of various hydrophobic groups on the surface of dendrimer was enhanced by increase in the number of branches but, was lowered by the presence of hydroxyl groups as the pendant terminal groups. Furthermore, the core of dendrimers was not always located towards the centre, its position was found to be altered by the number of branching points, type of terminal functional groups and the generation of dendrimer.  相似文献

A series of chiral phosphine-functionalized poly(propyleneimine) (PPI) dendrimers was synthesized by the reaction of carboxyl-linked C(2)-chiral pyrphos ligand (pyrphos=3,4-bis(diphenylphosphino)pyrrolidine) with zeroth-fourth generation PPI using ethyl-N,N-dimethylaminopropylcarbodiimide (EDC)/1-hydroxybenzotriazol as a coupling reagent. The dendrimers obtained were characterized by NMR spectroscopy and elemental analysis as well as FAB and MALDI-TOF mass spectrometry, which established their molecular masses of up to 20 700 amu. Metalation of the multi-site phosphines with [Rh(COD)(2)]BF(4) cleanly yielded the cationic rhododendrimers containing up to 32 metal centers (for the fourth generation species), representing the largest chiral phosphine dendrimer catalyst studied to date. The complete metalation of the chiral phosphine sites was demonstrated by (31)P NMR spectroscopy and the observation of the coordination-shifted AB part of the ABX spin system (delta(A)=33.9, delta(B)=32.9; (1)J(Rh,P)=150, 153 Hz; (2)J(P,P)=28 Hz). The relationship between the size/generation of the dendrimer and its catalytic properties was established in the asymmetric hydrogenation of Z-methyl-alpha-acetamidocinammate and dimethyl itaconate. A decrease in both activity and selectivity of the dendrimer catalysts was observed on going to the higher generations.  相似文献

Generation 5 ethylenediamine (EDA)-cored poly(amidoamine) (PAMAM) dendrimers (E5, E denotes the EDA core and 5 the generation number) with different degrees of acetylation and carboxylation were synthesized and used as a model system to investigate the effect of charge and the influence of dendrimer surface modifications on electrophoretic mobility (EM) and molecular distribution. The surface-modified dendrimers were characterized by size-exclusion chromatography, 1H NMR, MALDI-TOF-MS, PAGE, and CE. The focus of our study was to determine how EM changes as a function of particle charge and molecular mass, and how the molecular distribution changes due to surface modifications. We demonstrate that partially modified dendrimers have much broader migration peaks than those of fully surface functionalized or unmodified E5 dendrimers due to variations in the substitution of individual dendrimer surfaces. EM decreased nonlinearly with increases in surface acetylation for both PAMAM acetamides and PAMAM succinamic acids, indicating a complex migration activity in CE separations that is not solely due to charge/mass ratio changes. These studies provide new insights into dendrimer properties under an electric field, as well as into the characterization of dendrimer-based materials being developed for medical applications.  相似文献

[structure: see text]. A novel, rapid, inexpensive, and highly efficient divergent approach for the synthesis of a 32-amine-terminated G4 polyamide dendrimer has been developed. Each generation dendrimer was successfully obtained by the condensation of the preceding generation dendrimer with the building block and the deprotection with hydrazine in one pot. All the dendrimers were easily purified by precipitation in alkaline water, and the purity was confirmed by NMR, MALDI-TOF mass spectra, and elemental analysis.  相似文献

The physicochemical properties of quaternized poly(amidoamine) dendrimers (generation 4) with methyl or octyl groups and of their mixtures with sodium dodecyl sulfate (SDS) in aqueous solutions have been investigated using several techniques including surface tension, fluorescence of pyrene, and dynamic light scattering. In the single systems of the dendrimers, the dendrimer with octyl groups shows lower surface tension and lower micropolarity than the dendrimer with methyl groups. The hydrodynamic radii of two quaternized poly(amidoamine) dendrimers are considerably large, indicating the formation of aggregates. In the mixed systems of quaternized poly(amidoamine) dendrimers and SDS, the dendrimer with octyl groups-SDS mixed system shows very low surface tension and low micropolarity even in the presence of extremely low SDS concentration compared to those of the dendrimer with methyl groups-SDS mixed system. Maximum turbidity for both systems is observed at around the mixed molar ratio of dendrimer:SDS=1:1.5 where distinct changes have also been confirmed by surface tension, fluorescence of pyrene, and electrical conductivity measurements.  相似文献

A series of pyrazine (pz) complexes containing cis-(NH(3))(2)Pt(II), (tmeda)Pt(II) (tmeda = N,N,N',N'-tetramethylethylenediamine), and trans-(NH(3))(2)Pt(II) entities have been prepared and characterized by X-ray crystallography and/or 1H NMR spectroscopy. In these compounds, the pz ligands act as monodentate (1-3) or bidentate bridging ligands (4-7). Three variants of the latter case are described: a dinuclear complex [Pt(II)]2 (4b), a cyclic tetranuclear [Pt(II)](4) complex (5), and a trinuclear mixed-metal complex [Pt2Ag] (7). Mono- and bidentate binding modes are readily differentiated by 1H NMR spectroscopy, and the assignment of pz protons in the case of monodentate coordination is aided by the observation of (195)Pt satellites. Formation of the open molecular box cis-[{(NH3)2Pt(pz)}4](NO3)8.3.67H2O (5) from cis-(NH3)2Pt(II) and pz follows expectations of the "molecular library approach" for the generation of a cyclic tetramer.  相似文献
