G-CSF-pJGW1/pGP1-2/DH5α工程菌的系统稳定性研究

摘    要:将构建好的rhG-CSF工程菌(G-CSF-PJGWI/pCP1-2/DH5a)由-70℃冰箱取出,连续培养140代,每20代观察工程菌质粒的遗传稳定性及rhG-CSF工程菌蛋白表达稳定性。结果表明:rhG-CSF工程菌在连续培养140代后,其抗生素抗性无改变,质粒DNA酶切鉴定与原始菌相同,其蛋白表达率与原始菌基本一致。

Abstract:The constructed engineering bacteria E.coli DH5a that contain plasmid pJGWl-hGCSF and pGPl-2 were cultured up to 140 generations. The antibiotics resistance and expressed protein level of rhG-CSF of the engineering bacteria is very stable. The recomhinant plasmids of pJGWl-hGCSF and pGPl-2 of different generations also showed the same electrophoretic patterns after restriction enzyme cleavage as those of the primary recombinant bacteria.
