摘    要:该研究以蛹虫草子实体为研究对象,采用组织分离法从中分离纯化高产虫草酸的蛹虫草菌株,通过分子生物学技术对其进行鉴定,并以虫草酸含量为响应值,利用单因素试验和响应面法对其培养条件进行优化。结果表明,分离得到一株高产虫草酸的菌株YCC-1,并被鉴定为蛹草拟青霉(Paecilomyces militaris),菌株YCC-1产虫草酸的最佳培养条件为接种量7.8%、初始pH值6.0、培养温度25 ℃、转速140 r/min。在此最优条件下,虫草酸含量为132.65 mg/g,较优化前提高22.86%,为蛹虫草菌株液体发酵培养生产虫草酸提供试验依据。

Abstract:In this study, the Cordyceps militaris with high yield cordycepic acid was isolated and purified from C. militaris fruiting bodies by tissue isolation method, and identified by molecular biology technology. Using cordycepin acid content as response value, the culture conditions were optimized by single factor tests and response surface methodology. The results showed that the high-yield cordycepic acid strain YCC-1 was isolated and identified as Paecilomyces militaris. The optimum culture conditions for producing cordycepic acid by strain YCC-1 were inoculum 7.8%, initial pH 6.0, culture temperature 25 ℃, and rotational speed 140 r/min. Under the optimum conditions, the cordycepic acid content was 132.65 mg/g, which was 22.86% higher than before optimization. It provided experimental basis for the production of cordycepic acid by liquid fermentation of C. militaris strains.
