摘    要:目的探讨硫化氢(hydrogen sulfide,H2S)对心力衰竭(heart failure,HF)大鼠心肌的保护作用及机制。方法雄性Wistar大鼠随机分为对照组(control组),心衰模型组(HF组),NaHS(外源性的H2S)干预组(NaHS组)。其中心衰模型组用异丙肾上腺素(ISO)制备,NaHS组在给予ISO的同时每天腹腔注射NaHS一次。统计各组大鼠死亡率,测定肺与体重比值;通过超声检测各组大鼠左室血流动力学的变化;HE,电镜观察心肌形态学变化,TUNEL观察心肌细胞凋亡情况。结果 H2S提高了心衰大鼠存活率,降低肺与体重比值;明显改善心衰大鼠心脏血流动力及心肌损伤情况,并且减少了心肌细胞的凋亡。结论 H2S对心衰大鼠心肌具有明显的保护作用,能阻止心衰的进一步发展。

Abstract:Objective To investigate the protective effect of hydrogen sulfide(H2S) on rat myocardium from heart failure and its mechanisms.Methods Male Wistar rats were randomly divided into control group;heart failure model group(HF group): the rats received subcutaneous injection of isoproterenol(ISO);NaHS(exogenous H2S donor) groups: the rats received intraperitoneal injection of NaHS every day after received ISO.Survival rate was to gather statistics,lung-to-body weight ratio was measured.Haemodynamic parameters were monitored by echocardiographic measurements.hematoxylin-eosin(HE) and electron microscopy(EM) were used to examine myocardial morphological changes.terminal deoxynucleotidyl transferase-mediated dUTP nick-end labelling(TUNEL) was used to examine apoptosis of cardiac tissues.Results NaHS was found to improve the survival rate and decrease the lung-to-body weight ratio.It improved significantly cardiac hemodynamics and myocardial injury in rats.Histological observation by TUNEL and EM showed that NaHS inhibited cardiac apoptosis in HF hearts.Conclusion Hydrogen sulfide could protect myocardium in rats from heart failure and cease the progression of HF.
