黄芪总苷液对瘢痕疙瘩成纤维细胞增殖凋亡的影响

摘    要:目的 探讨黄芪总苷液对人瘢痕疙瘩成纤维细胞增殖与凋亡的影响.方法 不同浓度黄芪总苷液(10、20、40ng/mL)干预瘢痕疙瘩成纤维细胞,噻唑蓝(MTT)法检测细胞增殖;实时(real-time) PCR测定成纤维细胞凋亡抑制基因survivin及细胞凋亡因子p53及Bcl-2的表达;Western blot检测成纤维细胞survivin、p53及Bcl-2蛋白水平表达.结果 MTT法检测发现,与对照组(不加黄芪总苷液)比较,各黄芪总苷液各浓度组细胞490 nm处吸光度(A)值明显降低,瘢痕疙瘩细胞增殖均明显受抑制且呈剂量依赖性(P<0.05).real-time PCR及Western blot检测结果显示,黄芪总苷液对瘢痕疙瘩成纤维细胞的凋亡相关因子survivin、Bcl-2有不同程度的抑制作用,对P53有不同程度的促进作用,且呈浓度依赖性.结论 黄芪总苷液能抑制瘢痕疙瘩成纤维细胞的增殖,调控其凋亡,从而达到有效治疗瘢痕疙瘩的效果.

Abstract:Objective To study the effect of astragaloside on proliferation and apoptosis in human keloid fibroblasts.Methods The human keloid fibroblast ceils were treated with different concentration of astragaloside(10、20、40 ng/mL).Cell proliferation was detected by MTT,the gene expreesion levels and protein levels of apoptosis-related proteins,survivin,p53 and Bcl-2.were determined by real-time PCR and Western blot,respectively.Results Comparecl with control group(treated with 0 ng/mL astragaloside),the absorbance values (A490 nm) of each concentration group were significantly reduced,which suggest that the proliferation of all keloid fibroblast were markably inhibited in a dose-dependent way (P<0.05).The gene expreesion levels and protein levels of apoptosis-related proteins,survivin、Bcl-2 were largely suppressed and P53 werelargely promoted in a dose-dependent.Conclusion The keloid fibroblasts cells proliferation and apoptosis could be regulated by astragaloside.
