摘    要:我院从猪肺灌洗液中提取肺表面活性物质(PS),取得成功。先将猪肺灌洗液离心取得PS粗制品,然后用乙醚乙醇提炼得到精制品。每1000ml猪肺灌洗液大约可提取0.247gPS,其中主要成分磷脂占78.5%,蛋白质含量仅0.33%,磷脂和蛋白质比值高达224.8,说明磷脂纯度极高,其表面活性能将表面张力降至10mN/m以下,与人羊水和牛肺PS基本相似。

Abstract:We have succeeded in isolating pulmonary surfactant (PS) from the porcine lung, which is readily available in this country. Crude PS was obtained from high speed centrifugation of the washings of porcine lungs. Refinement by extraction with ether and alcohol produced 0.247 grams of PS per 1 000 ml of the washing fluid. Phospholipid was the main component, composing 78.5%, while protein was only 0.33%. The phospholipid to protein ratio was as high as 224.8, indicating a high purity of the former. Surface activity of the PS reduced minimal surface tension to less than 10 mN/m, showing that the chemical composition and surface activity of porcine PS are essentially similar to those of PS from human amniotic fluid and bovine lung. Further assessment and refinement are required before the product can be put to clinical use.
