中华眼镜蛇膜毒素和碘-131协同免疫导向对体外鼻咽癌细胞的抑制作用

摘    要:目的 为探索鼻咽癌新的特异性治疗方法,制备了中华眼镜蛇膜毒素(CT)和碘-131(131I)与抗鼻咽癌单克隆抗体BAC5的偶联物,观察其对体外鼻咽癌细胞CNE2的抑制作用.方法 采用氯胺T法将131I标记于BAC5,Iodogen法将125I标记于CT,用异型双功能交联剂SPDP偶联BAC5和125I-CT,并用噻唑蓝法观察抑瘤效应.结果 125I-CT-BAC5的IC<50>为9.17×10-8mol/L,131I-BAC5的IC50为5.83×108Bq/L,联合用药后抑制率明显提高.结论 125I-CT-BAC5和131I-BAC5对体外培养的CNE2细胞均有抑制作用,二者联合应用则抑制作用更为明显.

Abstract:OBJECTIVE: To explore a new specific therapy of nasopharyngeal carcinoma (NPC) using an anti-nasopharyngeal carcinoma (NPC) monoclonal antibody BAC5 conjugate with Chinese cobra (CT) and iodine-131((131)I). METHODS: BAC5 was labeled with (131)I by chloramine-T method, CT was labeled with (125)I using iodogen method, and BAC5 and (125)I-CT were conjugated by SPDP method. The inhibitory effect of the conjugate on cultured human NPC CNE2 cells was observed using MTT assay. RESULTS: The IC(50) of (125)I-CT-BAC5 conjugate was 9.17x10(-8) mol/L, and that of (131)I-BAC5 was 5.83x10(8) Bq/L, and their combined administration showed obvious inhibitory effect on the NPC cells. CONCLUSION: Both (125)I-CT-BAC5 and (131)I-BAC5 have obvious inhibition effects against NPC cells, but their combined use shows stronger effects.
