肾性高血压大鼠主动脉对血管活性物质反应的性别差异

摘    要:目的:探讨肾性高血压大鼠主动脉对血管活性物质反应的性别差异.方法:雄性与雌性SpragueDawley大鼠,用二肾一夹(2K1C)方法复制肾性高血压模型.术后随机分4组:雄性假手术、雄性2K1C、雌性假手术、雌性2K1C组,4 w后测定血压和左心室重/体重,并取胸主动脉进行离体血管环实验.结果:术后4w,与假手术组比较,2K1C组平均动脉压显著升高,左心室重/体重比显著增大,主动脉对乙酰胆碱(Ach)的舒张反应明显减弱,对NE和高钾溶液的收缩反应明显增强.在假手术组,雄性与雌性大鼠术后4 w上述各项指标改变差异无显著性.在2K1C组,与雌性大鼠相比较,雄性大鼠的改变更为明显.结论:2K1C肾性高血压的发展过程中,主动脉对血管活性物质的反应存在性别差异.

Abstract:Objective:To explore the gender differences of the response of aorta to vaso-active substance in rats with renal hypertension.Methods:Animals were divided into 4 groups: the male-sham, male-2K1C (two-kidney and one-clip),female-sham, and male-2K1C group. Four weeks after operation,arterial pressure and the left ventricular weight/body weight were measured and and experiment of the isolated aortic ring cut from thoracic aorta were performed.Results:Four weeks after operation,the mean arterial pressure compared with the sham groups,was significantly elevated and in ratio of the left ventricular weight/body weight was remarkably icreased,the diastolic response of aorta to Ach was signifecantly decreased and the systolic response of aorta to non-epinephrine and high-KCl solution remarkably elevated.In the sham group,there was no significant difference on the indices mentioned-above in the male and female rats after four weeks of operation.In 2K1C group,changes of the male rats,compared with the female one,were more significant.Conclusion:In the developing process of renal hypertension with 2K1C method,there are gender differences of the aorta vaso-active substances in Sprague-Dawley rats.
