核心蛋白聚糖Decorin对骨肉瘤细胞生长的研究

目的:探讨核心蛋白聚糖Decorin对骨肉瘤细胞生长的影响及其作用机制。方法:用不同浓度的Decorin、TGF-β1作用于人成骨肉瘤细胞Saos-2,通过RT-PCR、Western Blot、MTT等方法检测其细胞增殖的情况及其凋亡的改变,并用流式细胞仪分析细胞的凋亡情况。结果:Decorin在体外能抑制骨肉瘤细胞的增殖,诱导骨肉瘤细胞凋亡,并且凋亡的出现是通过促进Fas/FasL来实现的。结论:Decorin可以通过促进Fas/FasL的表达促进细胞凋亡,进一步明确了Decorin作为抗肿瘤药物的可能。

Study on the effect of Decorin on the growth of osteosarcoma cells

Objective:To explore the effect of Decorin on the growth of osteosarcoma cells and the mechanism.Methods:Human osteosarcoma cells Saos-2 were treated with Decorin and TGF-β1 of different concentrations.RT-PCR,Western blot and MTT method were used to detect the proliferation and apoptosis of osteosarcoma cells,flow cytometry was used to analyze the apoptosis of osteosarcoma cells.Results:Decorin inhibited the proliferation of osteosarcoma cells in vitro,induced the apoptosis of osteosarcoma cells,which was achieved by promoting Fas/FasL.Conclusion:Decorin can promote the apoptosis of osteosarcoma cells by promoting the expression of Fas/FasL,which confirms the probability of Decorin as a antineoplastic agent.
