摘    要:利用自组装单分子层(SAMs)/普鲁士蓝(PB)/壳聚糖(Chit)界面固定葡萄糖氧化酶(GOD),构建酶生物传感器,对传感器在极性非质子有机溶剂如乙腈(AN)、二甲基亚砜(DMSO)和N,N-二甲基甲酰胺(DMF)存在条件下的电化学特性进行了研究,考察了有机溶剂体积分数对传感器响应的影响。结果表明,传感器在50%(体积分数)AN中的电流响应可达到磷酸缓冲溶液(PBS)中的73%。传感器响应速度快、重现性好、抗干扰能力强,展现了高的灵敏度(57.3mA.M.cm-2)。该传感器用于血清标本中葡萄糖的检测,测定结果与市售葡萄糖酶光度测定试剂盒测定结果相吻合。

Abstract:A biosensor was fabricated with glucose oxidase (GOD) immobilized on self-assembled monolayers (SAMs)/Prus sian blue (PB)/chitosan (Chit). The electrochemical characteristics of the biosensor were studied in the presence of dipolar aprotic organic solvents such as acetonitrile (AN), dimethyl sulfoxide (DMSO) and N, N-dimethyl formamide (DMF). The effect of volume percentage of organic solvents on biosensor response was investigated. It is observed that the response current of the biosensor in 50% AN reaches 73% of that in phosphate buffer solution (PBS). The amperometric signals are fast, re producible and exhibiting high sensitivity (57. 3 mA · M · cm-2). The biosensor was used to determine glucose in serum speci mens, and the results are consistent with those obtained with the commercially available glucose enzyme photometric kit.
