超声造影血流灌注定量参数与肿瘤组织氧分压相关性:实验研究

摘    要:目的 探讨超声造影(CEUS)血流灌注定量参数与肿瘤组织氧分压(PO2)的相关性。方法 以30只新西兰大白兔左后肢外侧浅肌层建立VX2皮下浅肌层肿瘤模型,于接种后第10天对荷瘤兔行常规超声及CEUS检查,于超声引导下以组织PO2传感器穿刺肿瘤,测量随机2个ROI的PO2,并对相应区域CEUS时间-强度曲线进行分析,观察血流灌注峰增强(PE)、曲线下面积(AUC)、原始曲线上升斜率(Grad)、达峰时间(TTP)与肿瘤组织PO2的相关性。结果 共对30个肿瘤获取60组数据,其平均PO2(3.98±2.62)mmHg,平均PE(15.19±6.25)dB,平均AUC(315.10±202.90)mm2,平均Grad 1.02±0.50,平均TTP(16.41±6.54)s。PE、AUC、Grad与肿瘤组织PO2均呈正相关(r=0.822、0.638、0.445,P均<0.05);TTP与PO2无明显相关性(P=...

Abstract:Objective To observe the correlations of blood perfusion quantitative parameters of contrast-enhanced ultrasound (CEUS) and oxygen partial pressure (PO2) in tumor tissue. Methods VX2 tumor pieces were implanted in the outside left thigh superficial muscle of 30 New Zealand white rabbits. On the 10th day after inoculation, ultrasound and CEUS examination were performed in the tumor-bearing rabbits. Under the guidance of ultrasound, tissue oxygen pressure sensor was set for measuring PO2 within two random ROI of tumor. Then the time intensity curve of CEUS in the corresponding areas were analyzed to investigate the relationships of the enhancement of blood perfusion peak (PE), area under the curve (AUC), gradient of the original curve (Grad), time to peak (TTP) and the tumor tissue PO2. Results A total of 60 groups data were obtained from 30 tumors. The average PO2 was (3.98±2.62)mmHg, the average PE was (15.19±6.25)dB, the average AUC was (315.10±202.90)mm2, the average Grad was 1.02±0.50 and the average TTP was (16.41±6.54)s. PE, AUC and Grad were positively correlated with PO2 of tumor tissue (r=0.822, 0.638, 0.445, all P<0.05), but no significant correlation was found between TTP and PO2 (P=0.693). Conclusion Contrast-enhanced ultrasound blood perfusion parameters PE, AUC and Grad were positively correlated with PO2 in tumor tissue. CEUS could be used to roughly assess PO2 of tumor tissue.
