高效液相法测定香菇菇柄麦角甾醇的含量

摘    要:采用高效液相色谱法(High performance liquid chromatography,HPLC)建立了测定菇柄麦角甾醇的含量测定方法。确定提取过程中皂化剂的种类和醇碱比后,将样品皂化,萃取后蒸干溶剂,乙醇定容测定。采用Phe-nomenex-C18色谱柱,V(流动相甲醇)∶V(水)=98∶2,流速1.0 mL/min,检测波长282 nm。结果表明:麦角甾醇线性回归方程为Y=9E+9×106X-8919.9(X:质量浓度,mg/mL),R2=0.998 9,0.01~0.30 mg/mL范围内线性关系良好,回收率为97.31%~101.95%。与紫外分光光度法所测结果比较,HPLC法测定菇柄中麦角甾醇含量灵敏、快速、准确,适用菇柄中麦角甾醇的含量测定。

Abstract:A determination method of ergosterol in Lentinus edodes stem by high performance liquid chro- matography (HPLC) was established. After type of saponifier and alcohol-base ratio were determined in the extraction process, the sample was analyzed by saponifiing, extracting to dissolve in ethanol, then de- termined by HPLC. The conditions were Phenomenex-Cls, mobile phase methanol: water = 98:2, 1.0 mL/min velocity, detection wavelength of 282 nm. The calibration curve was Y = 9E + 9 x 106X- 8 919.9 (X:concentration, mg/mL), R2 = 0.998 9. The standard curve was linear over the range of 0.01-0.3 mg/mL, and the average recovery was 97.31%-101.95%. The results showed that the method is simple and accurate, so it is suitabie for content determination of ergosterol in Lentinus edodes stem.
