摘    要:目的:建立测定卵白蛋白(OVA)脂质体中OVA含量的方法。方法:采用高效液相色谱法。色谱柱为ZORBAX300SB-C8;流动相A为0.01%三氟乙酸水溶液,流动相B为0.01%三氟乙酸乙腈溶液,梯度洗脱,流速为0.8mL·min-1;检测波长为220nm;柱温为40℃。结果:OVA检测浓度线性范围为10~500μg.mL-(1r=0.9995);平均回收率为101.2%,RSD=0.74%(n=6)。结论:建立的测定方法灵敏度高、专属性强、简便快捷,可用于OVA脂质体中OVA的包封率以及含量的测定。

Abstract:OBJECTIVE: To establish a method for the content determination of ovalbumin (OVA) in OVA liposomes. METHODS:HPLC method was adopted. The separation was carried out on ZORBAX300SB-C8 column with mobile phase A consisted of 0.01% trifluoroacetic acid-water and mobile phase B consisted of 0.01% trifluoroacetic acid-acetonitrile (gradient elution) at the flow rate of 0.8 mL.min-a. The detection wavelength was set at 220 nm and column temperature was 40 ℃. RESULTS:The linear range of OVA was 10 to 500 μg·mL^-1 (r=0.999 5) with an average recovery of 101.2% (RSD=0.74%, n=6). CONCLUSION: The method is found to be sensitive, specific and simple. It can be used for the content and encapsulation coefficency determination of OVA in OVA liposomes.
