乌司他丁对脂多糖诱导的巨噬细胞NO释放和iNOSmRNA表达的影响

摘    要:目的 探讨乌司他丁对脂多糖(LPS)诱导的RAW 264.7细胞一氧化氮(NO)释放和诱生型一氧化氮合酶(iNOS mRNA)表达的影响。方法 应用LPS激活RAW264.7细胞株,与不同浓度组乌司他丁(100~10000U/ml)共同孵育,Griess试剂法测定NO释放量;实时荧光定量反转录聚合酶链反应(RT-PCR)技术分析iNOS mRNA的表达。结果 高浓度的乌司他丁(1000~10000U/ml)能抑制LPS诱导的RAW264.7细胞NO的表达(P<0.05),下调iNOS mRNA含量(P<0.05);低浓度乌司他丁(100U/ml)对LPS诱导的RAW 264.7细胞NO、iNOS mRNA表达与LPS单独处理组比较,差异无统计学意义(P>0.05)。结论 高浓度乌司他丁抑制LPS诱导的巨噬细胞NO释放和iNOS mRNA表达,这种抑制与浓度相关。

Abstract:Objective To investigate the effect of ulinastatin on the lipopolysaccharide(LPS)-induced production of nitric oxide(NO) and the mRNA expression of inducible nitric oxide synthase(iNOS) in macrophages.Methods The RAW264.7 macrophage cells activated with LPS were pretreated with ulinastatin of different concentrations(100-10 000 U/ml).The production of NO was determined by the Griess test.The mRNA expression of iNOS was determined by Real-time RT-PCR.Results Ulinastatin,especially in high concentration ranges(1 000-10 000 U/ml),inhibited LPS-induced production of NO(P<0.05),decreased the mRNAs expression of iNOS(P<0.05);There was no significant difference in NO and the mRNA expression of iNOS in LPS-induced RAW 264.7 cells between ulinastatin of low concentration(100 U/ml) and LPS alone(P>0.05).Conclusion High concentration ulinastatin can inhibit the production of NO and mRNA expression of iNOS in LPS-induced macrophages,and the effect is correlated with the concentration of ulinastatin.
