卫氏并殖吸虫抗原模拟表位的筛选和免疫反应性分析

摘    要:目的 初步探讨卫氏并殖吸虫抗原模拟表位的免疫反应性。方法 应用卫氏并殖吸虫病患者血清免疫粗球蛋白(Pw-Ig)筛选噬菌体随机12肽库,对筛选到的抗原模拟表位用ELISA检测其免疫反应性,并与卫氏并殖吸虫成虫抗原(PwA)进行比较。结果 获得的6个卫氏并殖吸虫抗原模拟表位(即P5、P6、P7、P8、P13、P16)特异性和敏感性与PwA相比差异均无显著性(P>0.05),其中P5、P6和P7交叉反应性明显较PwA低(x2=4.630,P<0.05),其余则与PwA无明显差异(P>0.05)。结论 卫氏并殖吸虫抗原模拟表位对卫氏并殖吸虫病的诊断具有潜在的应用价值,有望代替天然抗原用于卫氏并殖吸虫病的诊断。

Abstract:Objective To study the immunoreactivity of mimic antigen epitope of Paragonimus westermani preliminarily. Methods Specific Ig was purified from sera of patients with P. wester-mam and used to immunoscreen the phage peptide library (ph. D. -12). The mimic antigen epitopes screened were detected by ELISA and compared with the worm antigen of P. wedtermani (PWA). Results Between each mimic antigen epitope of P. westermani (P5 ,P6,P7,P8,P13,P16) and PwA, there were no statistical significant differences on the sensitivity and specificity (P >0. 05). The cross reactivity of P5,P6 and P7 was much lower than PwA (X2 = 4. 630,P<0. 05) and the others had no statistical significant differences from PwA (P>0.05). Conclusion The mimic antigen epitopes of P. westermani have potential value in the diagnosis of paragonimiasis, and may substitute the natural antigen.
