活性炭吸附蛋白质溶液中的热原物质的研究

摘    要: 目的:探索蛋白溶液中的热原物质去除问题?方法:以活性炭为吸附剂,3种常见蛋白为代表,采用定量鲎试验法,研究了活性炭吸附内毒素的有效程度和影响因素?结果:不同蛋白溶液中,活性炭吸附内毒素的有效程度相差显著,对溶菌酶溶液中的内毒素吸附最有效,去除倍数达216.5,而在牛血清白蛋白、牛血红蛋白溶液中的吸附倍数分别为33.1和51.1;酸性环境下,活性炭吸附内毒素更为有效;高离子强度促进活性炭对内毒素的疏水吸附作用;活性炭对蛋白质的吸附与蛋白质的相对疏水强度呈正相关?结论:活性炭吸附热原法适用于疏水性较低的、低分子量蛋白质或多肽,在微酸性环境中吸附去除热原物质?

Abstract:OBJECTIVE: To study that how to remove pyrogen from protein solutions. METHOD: The activated carbon was used to adsorb the pyrogen from bovine serum protein (BSA),bovine hematoglobin (BH) and lysozyme (LYZ).The quantitative limulus amebocyte lysat was used in this study.T5HZRESULTS: The adsorption effect was different apparently in these protein solutions.The clearance rate in LYZ solution was the highest,nearly 216.5,while in BSA and BH solution were 33.1 and 51.1 respectively,in acid environment,the adsorption for ET was more effective.High inonic strength promoted the interaction between the activated carbon and ET.The adsorbed protein of activated carbon increased with the relative hydrophobic strength of protein. CONCLUSION: In acid environment,activated carbon could be used to adsorb the pyrogen effectively from low hydrophobic,low molecular weight protein or polypeptide solutions.
