大黄对奥沙利铂加5-氟尿嘧啶化疗大鼠肠黏膜屏障的保护作用

目的观察大黄对奥沙利铂加5-氟尿嘧啶(FOLFOX方案)化疗大鼠肠黏膜屏障的保护作用。方法 40只Wistar大鼠随机分为实验组和对照组,每组20只。对照组予奥沙利铂、5-氟尿嘧啶分别按生药8、30mg/kg,容积10mL腹腔注射,每日1次。实验组在对照组用药基础上加用大黄按生药0.5g/kg、容积10mL灌胃,每日3次。2组均连续用药6d。6d后检测2组大鼠血清内毒素含量、肠系膜淋巴结及肝脏细菌移位阳性率;测量回、结肠黏膜厚度,并光镜下观察回、结肠组织结构病理变化。结果实验组血清内毒素含量、肠系膜淋巴结及肝脏细菌移位阳性率与对照组比较差异有统计学意义(P0.05),实验组低于对照组;实验组回、结肠黏膜厚度与对照组比较差异有统计学意义(P0.05),实验组高于对照组。光镜下实验组病理图片明显改善。结论大黄对奥沙利铂加5-氟尿嘧啶化疗大鼠肠黏膜屏障具有保护作用。

Protection of Rhubarb on barrier of intestinal mucosa of rat suffered from Oxaliplatin and 5-Fluorouracil

Objective To investigate the protection of Rhubarb on barrier of intestinal mucosa of rat suffered from Oxaliplatin and 5 - Fluorouracil. Methods 40 Wistar rats were randomly divided into two groups. Rats in control group received intraperitoneal injection of Oxaliplatin and 5 - Fluorouracil 8 mg/kg, 30 mg/kg, 10 mL, once a day. Rats in treatment group received intragastic Rhubarb, 0.5 g/kg, 10 ml on the basis of control group medication. The course was six days in two groups. The contents of endotoxin, lymphonodi mesenterici and bacterial translocation rate were measured after a course. The mucosa thickness of ileum and colon were measured and the pathological changes in ileum and colon tissue were observed with optical microscope. Results There was significant difference between two groups on the contents of endotoxin, lymphonodi mesenterici and bacterial translocation rate （ P 〈0.05）. These in treatment group were less than those in control group. The mucosa thickness of ileum and colon in treatment group was more than those in control group （ P 〈 0.05 ）. The pathological changes in ileum and colon tissue were improved. Conclusion Rhubarb has protection on barrier of intestinal mucosa of rat suffered from Oxaliplatin and 5 - Fluorouracil.
